Objective To survey the significance of specific immune response in the relationship between Sagnation the Liver-qi syndrome and the spleen Deficiency syndrome.Methods The rats with the Liver-qi Stagnation syndrome and the Spleen Deficiency syndrome were respectively corresponding and crossely treated with Radix Bupleuric dispersing the stagnated Liver-qi powder Decolion of Four Noble Drugs. Their T, B lymphocyte activity and specific immune response indexes changes (such as IgG, IgM in the serum) were respectively observed by lymphocyte transformation test and immunoturbidimetry. Results There were no notable changes about the IgG level among all kinds of groups. The IgM levels in the Liver-qi Stagnation syndrome and the Spleen Deficiency syndrome decreased, especially in the Liver-qi Stagnation syndrome (P0 05), Decolion of Four Noble Drugs could increase the IgM level of the patients with the Liver-qi Stagnation syndrome and the Spleen Deficiency syndrome (P0 05); The T lymphocyte transformation rate of the Spleen Deficiency syndrome obviously decreased, and Decolion of Four Noble Drugs could recover it to the normal level (P0 05). The rate of B lymphocyte transformation the Liver-qi Stagnation syndrome's also showed the increasing tenderly (P0 05). Conclusions The relationship between the Liver-qi Stagnation syndrome and the Spleen Deficiency syndrome could occur in the phase of antigen presentation, especially in the phase of the T H cell activation stimulated by antigen. Radix Bupleuric dispersing the stagnated Liver-qi powder could enhance T, B lymphocyte activities by reverse the Liver-qi Stagnation syndrome,and Decolion of Four Noble Drugs could enhance T lymphocyte by improving the Spleen Deficiency syndrome.
6.The Effect of Thalidomide and Triamcinolone Acetonide on Human Umbilical Vein Endothelial Cells

Objective: To investigate the effect of thalidomide and triamcinolone acetonide (TA) on human umbilical vein endothelial cells (HUVEC). Methods: The primary culture of HUVEC with collagenase Ⅰ was identified with FⅧAg. There were 3 groups in the study including thalidomide group(10, 25, 50 and 75 mg/L), TA and thalidomide group and control group. The absorbance ratio was measured through MTT-test. Results: Both thalidomide group and TA and thalidomide group showed inhibitor effect on the growth of HUVEC significantly(P < 0.01). The inhibitory effect was dose dependent. Compared to thalidomide group, the inhibitory effect was more significant in TA and thalidomide group(P < 0.05). Conclusion: Thalidomide can inhibit the growth of HUVEC; however, the effect was more significant in TA and thalidomide group.
7.The modulatory effect of transcutaneous electrical nerve stimulation (TENS) on tonic heat pain in the human brain: topographic mapping of absolute EEG power spectra

Objective To investigate the effect of transcutaneous electrical nerve stimulation (TENS) on baseline eyes-closed brain activation and on pain-related EEG activity when TENS is applied contralatarally to the hand of pain. Method 128-channel electroencephalography (EEG) was recorded in four conditions including baseline,heat pain test,TENS stimulation and contralateral TENS modulation in 15 right-handed healthy young males. The Short Form McGill Pain Questionnaire was employed to assess pain sensation. Fast Fourier Transformation analysis (FFT) was performed to calculate absolute EEG power spectra based on 7 bands spectrum. Results ① The fronto-central beta-1 activity was significantly enhanced by TENS compared to eyes-closed [eyes-closed:(290±201)μV2,TENS:(385±224)μV2,t=3.323,P<0.01].②TENS markedly relived pain sensation assessed by SF-MPQ and significantly increased the posterior-anterior alpha-1 power [pain:(332±221)μV2,TENS modulation:(378±243)μV2,t=3.683,P<0.01] and fronto-central and posterior beta-1 activities [pain:(348±213)μV2,TENS modulation:(397±240)μV2,t=2.362,P<0.05]. Conclusion TENS,used as a peripheral nerve stimulation,mainly activates sensorimotor cortex.The increased Alpha-1 activity implies that TENS is likely able to relieve pain affective.
8.Expression of VEGF gene mediated by adenovirus in human bone marrow stromal cell

[Objective]To investigate the possibility of stable expression of VEGF~65 in human bone marrow stromal cell.[Method]hBMSc were divided into transfection group, empty vector group and control group, hBMSc were infected with Ad-VEGF165(MOI=20 pfu). The hBMSc proliferation and transfection efficiency were confirmed by fluorescence microscope.The expression of VEGF165 gene of cultured hBMSc transfected with VEGF165 gene was assayed by RT-PCR, ELISA and Western blot.[Result]VEGF165 gene was amplified with RT-PCR.GAPDH gene(internal control) was located at 549 bp in the three groups, VEGF165 gene was located at 500 bp in transfection group but not in the other groups. 9.Study on the GC-MS Fingerprint Spectrum of Volatile Components in Danqi Piantan Capsule

OBJECTIVE:To establish the GC-MS fingerprint spectrum of volatile components in Danqi piantan capsule. METH-ODS:GC-MS was performed. HPLC conditions:The column was BPX 5,temperature of injection was 250 ℃,respectively;carri-er gas was He,flow rate was 1 ml/min,split injection,split ratio was 50:1 and volume injection was 0.02 μl. MS conditions:ion source was EI,bombarding energy was 70 eV,temperature of ion source and transmission line was 200 ℃ and 250 ℃,respective-ly;scan range was 40-500 amu,solvent delay of 2 min. The peak with largest chromatographic peaks areas was reference peak,com-mon peaks of 10 batches of Danqi piantan capsule were analyzed. Similarity evaluation system for chromatographic figerprint of TCM was adapted for the similarity analysis and NIST standard MS database was used to retrieve and identify the common peak. RESULTS:There were totally 13 common peaks in the 10 batches of Danqi piantan capsule with similarity degree>0.92;accord-ing to the verification,the fingerprint spectrum was a dapted for the similarity analysis and reference fingerprint of 10 batches of Danqi piantan capsule showed good consistency. The No.3 peak was Angelica sinensis unique,the No.2,4,5,7,8,12 peak was Acorus tatarinowii unique. CONCLUSIONS:The established method is specific and stable,and can provide basis for the quality as-sessment for Danqi piantan capsule.

The effect of chitin on human umbilical vein endothelial cells (HUVECs) injured by oxygen free radicals was investigated in vitro. The degree of injury was determined by the amount of malondialdehyde (MDA) and lactic dehydrogenase (LDH) in the culture medium. The results indicated that chitin could reduce the amount of MDA and LDH significantly, showing that chitin could protect HUVECs from oxygen free radicals injury.
