1.Expression of gC1qR in cervical cancer and its effects on the biological behaviors of human cervical cancer cells

[Abstract ] Objective Globular C1q receptor (gC1qR), a highly acidic receptor protein, is expressed in almost all mamma-lian cells in addition to exoerythrocytic, which can mediate a variety of biological responses.The study aimed to explore gC1qR expres-sion in cervical cancer and itd effects on the biological behaviors of human cervical cancer cells. Methods Retrospective analysis was made on 100 cervical tissue samples of patients in Nanjing Maternal and Child Health Hospital from August 2014 to April 2015, in-cluding 50 cervicitis tissues and 50 cervical cancer tissues.Immunohistochemical SP method was applied in the research of cervical cancer cell line C33a to detect the expression and the location of gC1qR in cervical tissues.Real-time PCR and Western blot analysis were respectively applied to detect the levels of gC1qR mRNA and gC1qR protein expression.Besides, the abilities of C33a cells mi-gration, invasion and apoptosis were respectively assessed by in vitro cell wound healing experiment, transwell assay and flow cytome-try. Results The expression of gC1qR gene was dramatically decreased in the group of cervical cancer tissues when compared with chronic cervictis group (2.18 ±0.37 vs 7.23 ±0.69, 0.27 ±0.09 vs 0.74 ±0.02, P<0.001).gC1qR overexpression could result in sig-nificant up-regulation of cervical celluer apoptosis([22.89 ±1.46]%vs [12.98 ±0.57]%) and down-regulation in migration ([42.60 ± 3.29]%vs [141.83 ±4.71]%) and invasion([26.20 ±2.89]%vs [67.13 ±0.95]%).Typical apoptosis was also observed in cervi-
cal cells by transmission electron microscope. Conclusion gC1qR expression might play an important role in inhibiting the invasion and migration of cervical cancer cells and inducing the apoptosis of cervical carcinoma cells, which provides new clues and potential targets for the treatment of cervical cancer in further research.
2.Effect and mechanism of IKK-2 receptor blocker on corneal stromal fibroblasts

BACKGROUND:Corneal stromal fibroblasts have been shown to express interleukin-8 in the stimulation of lipopolysaccharide. Different reactions of fibroblasts to lipoprotein or other inflammatory mediators may constitute different characteristics of different tissues in the inflammatory response. OBJECTIVE:To observe the inhibitory effect of IKK-2 receptor blocker TPCA-1 on human corneal stromal fibroblasts to secrete inflammatory cytokines, chemokines and cel adhesion molecules under the stimulation of lipopolysaccharide and its signal transduction pathway, to compare with dexamethasone, and to explore alternative or synergistic effects after their combination. METHODS:This study measured the secretion of interleukin-1, tumor necrosis factor alpha, interleukin-6, and interleukin-8 from cultured human corneal stromal fibroblasts under the action of basic state and lipopolysaccharide, and their changes after the intervention with IKK-2 receptor blocker TPCA-1 and dexamethasone. We also detected expression level of intercelular adhesion molecule-1 and interleukin-6 in cel surface, and verified the changes in expression levels of intercelular adhesion molecule-1, interleukin-6, and interleukin-8 from mRNA level, as wel as examined the expression of nuclear factor kappa B under above conditions.
3.De-noising of Impedance Cardiography Differential Signal and Detection of the Feature Points Based on Wavelet Transformation.

Calculation of cardiac hemodynamic parameters is based on accurate detection of feature points in impedance cardiogram. According to these parameters, doctors can determine heart conditions, so it is very important to accurately detect the feature point of impedance differential signals. This article presents a process in which we used wavelet threshold method to de-noise signals, and then detected the feature points after six layers wavelet decomposition by using bior3. 7. The experimental data were collected from healthy persons in our laboratory and twenty two clinical patients in Chongqing Daping Hospital by using KF_ICG instrument. The results indicated that this method could precisely detect feature points whether it was from healthy people or clinical patients. This helps to achieve the application of noninvasive detection cardiac hemodynamic parameters in clinical treatments by using impedance method.

4.Repairing sacrococcygeal decubitus ulcers grade Ⅲ with direct polymerizing suture after using rhEGF during wound bed preparation

Objective To study the clinical efficacy of recombinant human epidermal growth factor (rhEGF) in the treatment of sacrococcygeal decubitus ulcers grade Ⅲ with direct polymerizing suture after appling rhEGF to reinforce wound bed preparation (WBP). Methods From January 2007 to October 2009, 60 patients with sacrococcygeal decubitus ulcers Grade Ⅲ, were divided into control group and treatment group. The ulcer size was 3 cm × 4 cm to 10 cm × 12 cm and all ulcers were infected for 15-70 d. Treatment group received traditional dressing change and appling rhEGF to reinforce wound bed preparation. Control group received traditional dressing change only. The operative technique that we used in two groups was direct polymerizing suture. Cure rate of stage Ⅰ and complication morbidities were analyzed. Results Cure rate of stage Ⅰ was 87% in treatment group and 70% in control group. Complication morbidities were 13% in treatment group and 30% in control group. The difference between these two groups was statistically significant (P < 0.05). Conclusion Appling rhEGF to reinforce wound bed preparation before operation could make subsequent treatment more effective and improve the cure ratio of operation with decreasing complications and morbidities. And more, dissecting under fascia possesses the advantages of easiness to perform and rich blood supply.The method of appling rhEGF with direct polymerizing suture is a simple, high efficient approach for the first repairment of sacrococcygeal decubitus ulcers grade Ⅲ, especially desirable for the elderly.

6.Expression of calcium handling protein in diastolic heart failure

AIM: To elucidate molecular mechanisms underlying calcium handling protein in diastolic heart failure (DHF) from mRNA level and protein expression, including calcium adenosine triphosphatase (Ca 2+ -ATPase), phospholamban, ryanodine receptor, calsequestrin and L-type calcium channel. METHODS: The mRNA of these calcium handling genes were detected by RT-PCR, and the protein levels were analyzed by Western blot analysis. RESULTS: Compared with sham-operated rabbits, the steady-state levels of mRNA encoding the SR Ca 2+ -ATPase and cardiac L-type calcium channel were decreased significantly in rabbits with DHF, and protein level of SR Ca 2+ - ATPase was greatly reduced, whereas the mRNA and protein levels of other calcium handling protein were unchanged. CONCLUSION: L-type calcium channel and the sarcoplasmic reticular Ca 2+ -ATPase were down regulated in DHF. These changes may be a contributory factor for DHF.
8.Study on biological activity of thrombopoietinⅡ in vivo

AIM: To investigate the biological activity of thrombopoietin Ⅱ(TPOⅡ) in vivo , which consists of two new kinds of ligand binding with thrombopoietin receptor. METHODS: Purified ligandⅠof TPOⅡ, artificial compound ligandⅡ of TPOⅡand rhTPO were injected into purebred Babl/c mice respectively in 7 days by intraperitoneal injection once for a day. Then the biological activity of TPOⅡ was analyzed by measuring peripheral platelet counts by the end of the seventh day. RESULTS: On the seventh day, the platelet counts of mice treated by ligandⅠof TPOⅡ were higher than that in the negative control group( P 0.05). On the fourteenth day, the platelet counts increased in two all experimental groups of TPOⅡcompared with negative control group( P 0.05). Moreover the platelet counts of mice in two experimental groups of TPOⅡ and the positive group showed increase with experimental days. CONCLUSION: The purified ligandⅠof TPOⅡ had obvious activity in increasing platelet production, which is not different from the effect of rhTPO.

10.Intranasal administration of low dosage recombinant human erythropoietin inhibits seizure in rats.

OBJECTIVETo investigate the effect of intranasal administration of low dosage recombinant human erythropoietin (r-HuEPO) on seizure in rats.
METHODSAfter intranasal or intraperitoneal administration of r-HuEPO, the behavioral and electroencephalographic changes were observed in pentylenetetrazol (PTZ) and maximal electroshock (MES) induced seizure or electrical amygdaloid-kindled seizure of rats.
RESULTIntranasal administration of low dosage r-HuEPO increased the seizure latency, and decreased the seizure grade and duration, and the number of convulsive episodes in PTZ induced seizure, with the most potential dosage of 2.4 IU. Intraperitoneal administration of r-HuEPO (3 000, 4 000 IU/kg) only decreased the seizure duration and number of convulsive episodes. The seizure grade, forelimb or hindlimb extension duration were decreased in MES-induced seizure by intranasal administration of 2.4 IU r-HuEPO. In addition, intranasal administration of 2.4 IU r-HuEPO decreased the seizure grade, generalized seizure duration and afterdischarge in electrical amygdaloid-kindled rats stimulated with generalized seizure threshold.
CONCLUSIONIntranasal administration of low dosage r-HuEPO can inhibit the seizure in rats.
