1.Impulsion therapy of children's nephrotic syndrome

Objctive To search for the therapeutic method of children's nephrotic syndrome.Methods Sixty-six cases oe children's nephrotic syndrome were randomly divided into 2 groups,impulsion group (34 cases) and control group (32 cases). Dexamethasone (1.5~3 )/mg (kg?d) added into (100~150)ml 10%GS solution, intravenous drip in impulsion group, one time a day, totat 3 days, the fourth day stoped. The fifth day started again and used one time evcry two days, total 6 times. Prednisone(1.5~3)mg/(kg?d) were taken next day and total 4 weeks, then grandually decreased the dose. Only prodnisone was used in control group, the method and dose were the same as impulsion group.Results Complete remission. partial remission inefficacy ere 23, 7 and 4 cases respectiye1y in impulsion group and 22, 5 and 5 cases respectively in control group, the effective rates of the 2 group are 88.23%and 84.38% (P>0.05). The times of state of illness stabilization are respectively 11.3?7.2 and 10.48?6.34 months in the 2 groups. The side effect of impulsion group is bigger than that of control group.Conclusion Children's primary nephrotic syndrome should be treated for 8 weeks by routine hormone induction therapy, if no remission, impulsion therapy could be used.
2.One Hundred and Seventy-Two Cases with Ulcerative Colitis in Children

Objective To investigate the diagnostic and therapeutic experience in children with ulcerative colitis(UC) from clinical data.Met-hods The literature on UC in children publicated from 1995 to 2004 were retrieved by CBM,CNKI and VIP database.The clinical manifestations,X-ray,endoscopic and histologic features,management and prognosis of 172 patients in this hospital correspond with the inclusion criteria were retrospectively investigated and a definite diagnosis with UC was made.Results Of 172 patients,the ratio of maleold female was 1.07:1.0,and the age of patients ranged from 2 months to 15 years old.Thirty-six patients(36.7%)were less than 3 years-old;the course ranged from 3 days to 4 years.One patient had family history.The main manifestations were chronic diarrhea,mucus bloody stool,bloody purulent stool and abdominal pain.The systemic symptoms were fever,weight loss,malnutrition and anemia,no enteral symptoms and complications.The lesions were mainly seen in the whole colon with moderate to severe degree.The therapies based on salicylaxosulfapyridine(SASP) or 5-acetylsalicylic acid(5-ASA) together with corticosteroid in the short period have clinical remission in different levels.From long follow-up,the complete remittence was less than one third.Conclusions It is not rare to see UC in infantile.The clinical characteristics of UC in children are different from those in adults,the same is true before and after 3 year-old.The UC in children has high misdiagnosis and is difficult to manage.It is very necessary to establish a diagnostic and therapeutic guideline for children with UC in China.

4.Isolation and Identification of Endophytic Bacteria H-6 with High Antifungal Activity from Huperzia serrata

In this study,nearly 200 endophytic bacteria were isolated from different part of Huperzia serrata, over 60 bacterium with clear antifungal activity were selected from those cultures.Among them,strain H-6 exhibited the highest antifungal activity which was strongly inhibits the growth of many plant pathogenic fungi such as Sclerotinia scleroliorum,Fusarium graminearumt,Sclerotinia libertiana,Phytophthora capsici Leonia and Sesame fusarium wilt.According to the characteristics of morphology,physiology and biochem- istry tests and the comparison of 16S rDNA sequence,the strain H-6 was similar to the Burkholderia.So strain H-6 was identified as Burkholderia sp.H-6.The results also showed that Burkholderia sp.H-6 was markedly different from Burkholderia cepacia that was applied widely in agriculture as antagonistic bacteria. The medium and culture conditions of the strain all were optimized by single factor and orthogonal experiments.In the investigation of the culture condition,growth was carried out in a basal medium(potato juice)and gradually supplemented with the various ingredients to be investigated.The major ingredients be- ing investigated included carbon sources and nitrogen sources.The optimal antifungal activity production condition is growth in a medium(potato juice with 2.5%mannitol and 0.1%NaNO3),initial pH 4.0 at 28℃.
5.Bioavailability and bioequivalence of erythromycin ethylsuccinate granules in healthy volunteers

Objective To study the bioavailability and bioequivalence of erythromycin ethylsuccinate granules in healthy male volunteers.Methods In a randomized two-period crossover study,20 healthy male volunteers received single 500 mg test and reference formulations of erythromyein ethylsuccinate granules.The plasma concentrations of erythromycin were assayed by microbiological method.Results The parameters of test and reference preparations were as follows:T_(max)(0.86?0.22)and (0.80?0.13)h,C_(max)(2.13?0.64)and(2.16?0.61)mg/L,t_(1/2)(2.04?0.2)and(1.97?0.4)h,AUC_(0-t)(4.96?1.73)and(4.63?1.52)mg?h/L,respectively.There was no significant difference between the two preparations.The rela- tive bioavailahility of the test granules was(109.1?22.8)%.Conclusions The two preparations of erythromycin ethylsucci- nate granules are bioequivalent.
6.Low frequency ultrasound combined with microbubbles and brain-derived neurotrophic factor drugs in prevention of ischemic stroke in rats

Objective To explore the preventive therapeutic effects of brain-derived neurotrophic factor (BDNF) on acute ischemic stroke in rats under ultrasound combined with microbubbles.Methods Twenty-seven healthy SD rats were randomly divided into sham-operation group (n=9),BDNF group (n=9) and BDNF plus ultrasound group (n=9),then middle cerebral artery occlusion (MCAO) models were established.The concentration of BDNF in targeted brain tissue was measured at 24 h after operation.MRI examination and behavioral assessment were performed at 1,3 and 7 days after operation.Results The concentration of BDNF in BDNF plus ultrasound group was significantly higher than that in BDNF group and sham operation group (all P<0.05).The infarct size in BDNF group gradually reduced with time increase,and the infarct size was significantly smaller than that of BDNF group at the 7th day.The behavioral assessment had a significant statistical difference between BDNF plus ultrasound group and BDNF group at the 7th day after the operation (P<0.05).Conclusion The preventive therapeutic effect of BDNF could be enhanced by ultrasound in the rat model with acute ischemic stroke,which can significantly reduce the size of the infarct and improve the behavior of the rats.
7.Research progress on molecular genetics of forest musk deer.

Forest musk deer is one of the large-scale farming musk deer animals with the largest population at the same time. The male musk deer can secrete valuable medicines, which has high medicinal and economic value. Due to the loss of habitat and indiscriminate hunting, the numbers of wild population specie and the distribution have been drastically reduced. Therefore, in-depth understanding of the molecular genetics progress of forest musk deer will pave a way for musk deer protection and breeding. In this review, the progress associated with the molecular marker, genetic classification, artificial breeding, musk secretion and disease in past decades were reviewed, in order to provide a theoretical basis for subsequent molecular genetic researches in forest musk deer.

P300/CBP-associated factor(PCAF),an important member of histone acetyltransferase family(HATs) within eukaryotic cells,is capable of inducing the acetylation of histone,promoting the transcription of specific genes and involving in many biological effects.In the present study,full-length cDNA of PCAF was inserted into plasmid pGEX-5x-1,then the soluble protein GST-PCAF was expressed in E.coli BL21(DE3) after the optimization of inducing conditions.The recombinant protein was further purified with affinity chromatography and tested the activity by in vitro acetylation assays.High efficient PCAF protein produced by this method could serve for the study on the role of PCAF in gene regulation and the interaction between PCAF and other proteins.

Objective To explore the investigate of X-chromosome-linked inhibitor of apoptosis pro- tein(XIAP)and its effect on chemotherapeutic sensitivity in bladder carcinoma.Methods Using immu- nohistochemistry methods,the expression of XIAP was evaluated in 47 bladder carcinomas and 6 normal bladder tissues.The XIAP gene was transfected into bladder cancer cell line T24 by liposome and the positive clone was screened by G418.Cellular XIAP mRNA level was detected by RT-PCR.The apoptosis of T24 cells was induced by low-dose of mitocycin C(0.005 mg/ml and 0.05 mg/ml,respectively).The in vitro cellular growth activities were assayed by MTT color imetry;and the apoptosis rate was assayed by TUNEL methods. Results The expression rate of XIAP was 78.7%(37/47)in bladder carcinoma samples,with no corre- lation with carcinoma stages and grades(P>0.05).XIAP mRNA level in transfected T24 ceils was signifi- cantly increased by 3.8 times.Treated with 0.005 mg/ml and 0.05 mg/ml of mitomycin C,the growth rates of XIAP transfected T24 cells were increased [(11.60?0.25)% and(16.51?0.87)% ,respectively,P<0.05];and the apoptosis rates were decreased [(10.1?0.2)% and( 11.9?0.2)% ,respectively,P<0.05]compared with those in control cells.Conclusions XIAP is highly expressed in humun bladder car- cinoma samples.Overexpression of XIAP in T24 cells results in decrease in bladder carcinoma cell apoptosis induced by MMC,which may decrease the chemotherapeutic sensitivity of T24 cells.
10.Mechanism and clinical significance of anti-apoptotic gene bcl-2 expression in diffuse large B-cell lymphoma.

OBJECTIVETo evaluate the molecular mechanism and prognostication of bcl-2 protein expression in different subgroups of diffuse large B-cell lymphoma(DLBCL) in Guangxi Zhuang Autonomous Region, China.
METHODSImmunohistochemical stains for CD10, bcl-6, MUM-1, bcl-2 and NF-κB were performed in 214 cases of DLBCL. The Hans immunologic classification was applied to classify DLBCL into GCB and non-GCB subgroups. Using a dual-probe fluorescence in-situ hybridization (FISH) assay, IgH/bcl-2 gene translocation and bcl-2 amplification were analyzed.
RESULTSIn 214 cases of DLBCL, 30.8% (66/214) of cases were GCB and 69.2% (148/214) were non-GCB. Twenty-seven point three percent (18/66) of GCB subgroups and 59.5% (88/148) of non-GCB subgroups had bcl-2 protein expression, with a significant difference (P < 0.01). IgH/bcl-2 translocation was positive in 3.7% (8/214) of cases, even majority of them (6/8) was found in GCB subgroup, while represented only 9.1% of GCB case. There was a significant difference (P = 0.02) in bcl-2 gene amplification between GCB (27/66, 40.9%) and non-GCB subgroup (86/148, 58.1%). Among non-GCB cases, the expression of bcl-2 was correlated with that of NF-κB expression and bcl-2 gene amplification (r = 0.216 and 0.219, respectively, P < 0.05). No similar correlation was observed in GCB cases. The overall survival time of bcl-2-positive patients (31.4 ± 3.8) months was shorter than that of bcl-2-negative patients (40.2 ± 4.2) months. In conjunction with immunophenotypes and clinical stages, the bcl-2 positive patients had a 1.89 times higher risk than that of the bcl-2 negative patients.
CONCLUSIONSMajority of the cases were prognostically unfavorable non-GCB subgroups among DLBCL, which were characterized by high frequency of bcl-2 gene amplification and low frequency of IgH/bcl-2 translocation. The anti-apoptotic gene bcl-2 was frequently expressed in non-GCB subgroups and closely related to the gene amplification and NF-κB activation. bcl-2 positive patients had more short overall survival times, would face significant higher risk of death, these results suggested that bcl-2 could be a prognostic marker independent to clinical staging and immunophenotyping.
