1.Analysis of the Utilization of Antidepressant Drugs in 31 Hospitals of Nanjing During 2011 to 2013

OBJECTIVE:To provide reference for rational use of antidepressant drugs in the clinic. METHODS:The utilization of antidepressant drugs in Nanjing during the period of 2011-2013 were analyzed statistically in respect of DDD,DDDs,DDC,av-erage annual growth rate and ratio of ranking method . RESULTS:From 2011 to 2013,consumption sum and DDDs of antidepres-sant drugs increased year by year in 31 hospitals of Nanjing;paroxetine occupied the first place in the list of consumption sum. Se-lective serotonin reuptake inhibitors (SSRI) got the top place ranked by DDDs,followed by serotonin/norepinephrine reuptake in-hibitors and special serotonin antidressant. CONCLUSIONS:The application of antidepressant drugs in Nanjing area is basically rea-sonable. SSRIs,such as fluvoxamine,fluoxetine,citalopram,sertraline and paroxetine,are still the first-line antidepressant drugs recent years in Nanjing.
2.In situ hybridization and analysis of core sequence of follicle-stimulating hormone and its receptor in rats submaxillary

Objective To explore if the expression of follicle-stimulating hormone ( FSH ) and its receptor ( FSHR) exist in the rat submandibular gland tissue , which may provide a theoretic evidence for further studying on adjusting function of the FSH in submaxillary gland .Methods Twenty normal SD rats , with a mean body weight of (200 ±20)g, were adopted to collect out the submandibular gland after abdominal cavity anesthesia .The method of in situ hybridization was employed on cellular localization of FSH and FSHR .RT-PCR was used to detect if FSH and its receptor mRNA exist in the rat submandibular gland .RNA was extracted from the submandibular gland , then FSH and the core cDNA of its receptor gene were obtained with reverse transcription polymerase chain reaction and analyzed on its sequence . Results The epithelial cells of serous acinus and granular convoluted tubule in submaxillary gland of rats had FSH and FSHR mRNA hybridized signals , which were detected in cytoplasm with negative nuclei .Specific bands of FSH cDNA and FSHR cDNA were identical to be 193bp and 413bp respectively.Conclusion FSH and FSHR can be synthesized by the epithelial cells of serous acinus and granular convoluted tubule in submaxillary gland of rats , thus this study further proves that the submandibular gland is the target organ of FSH .
3.Abnormal expression of BRMS1 and Cx43 protein in thyroid cancer occurrence and progression

Objective:To investigate the protein expression levels of breast cancer metastasis suppressor 1 (BRMS1) and Con-nexin 43 (Cx43) in thyroid cancer and their correlation with clinicopathologic parameters. Methods:Immunohistochemistry Streptavi-din-Peroxidase method was used to detect the BRMS1 and Cx43 protein expression in 195 tissue samples, including 90 cases with thy-roid carcinoma, 45 cases with thyroid adenoma, 30 cases with nodular goiter, and 30 cases with normal thyroid glands. Results:The positive rates of BRMS1 and Cx43 expression was 56.7%(51/90) and 41.1%(37/90) in thyroid carcinoma, respectively, which are sig-nificantly lower than the rates in thyroid adenoma, nodular goiter, and normal thyroid gland tissues (P<0.001). Of the three pathological types of thyroid cancer, the positive expression rate of Cx43 was 61.9%in papillary carcinoma, 27.8%in medullary carcinoma, 27.3%in follicular carcinoma, and 12.5%in undifferentiated carcinoma. Statistical differences in the BRMS1 expression among papillary car-cinoma and the other pathological types were also noted. Unlike the patients without lymph node metastasis, the positive expression of BRMS1 and Cx43 proteins were both significantly low among patients suffering from nodal metastasis. Subgroup analysis shows that the positive expression of BRMS1 and Cx43 protein gradually decreased with TNM staging. In addition, a positive correlation was ob-served between the BRMS1 and Cx43 protein expression (r=0.494, P=0.032). Conclusion:The decreased expression of BRMS1 and Cx43 proteins is significantly correlated with the metastasis of thyroid cancer and malignant grade. The combined detection of the two proteins can be ideal biomarkers for judging the prognosis of thyroid carcinoma.
4.A new type of fusion apparatus of seaphoid-trapezium-trapezoid for limited interearpal arthrodesis

Objective To develop a new type fusion apparatus for scaphoid-trapezium-trapezoid (STT) arthrodesis.Methods The apparatus was designed on the basis of the anatomic data of STT measured from Chinese cadaveric wrists.Simulated arthrodeses of STT joints were conducted to test the fixation performance of the apparatus made of titanium alloy on the STT joint specimens.Several modifications were made before the apparatus was made into a finished product.Results The fusion apparatus consists of a plate and screws.The fusion plate for STT arthrodesis is made into a taper one connected to a cylinder.On the plate are three screw holes through which the three carpal bones (STT) are fixed by three screws respectively.The hollow cylinder is designed for bone grafting,and its outer threads ensure a very tight contact between the fusion plate and the carpal bones.Auxiliary tools are also made.Conclusions As the novel design of the fusion apparatus improves the mechanical strength of fixation of STT and prevents the possible rotation and collapse of fused STT,the apparatus is a reliable internal fixation device for the limited intercarpal arthrodesis.
5. Effect of Buyanghuanwu decoction on preventing atherosclerosis induced by Hey via inhibiting NF-κB-dependent

OBJECTIVE: To investigate the effects of Buyanghuanwu decoction on preventing atherosclerosis induced by Hey in rats in vivo. METHODS: The model of atherosclerosis in rats was induced by intragastric gavage of Hey. The intervention group was administered Buyanghuanwu decoction (5, 10, 20 g · kg-1 · d-1). After 8 weeks treatment, the area of atherosclerotic plaques, blood vessel walls and their ratio, and ROS contention in the aorta were analyzed. The positive cell ratio of NF-κB P65 in vascular was detected by immunohistochemistry, and the expression of I-κBα was determined by Western-blotting. RESULTS: The rats in the model group showed evident AS lesions which had significant difference with others groups. Compared with control group, the ROS content of model group was elevated (P < 0.01), the I-κBα protein in the vascular tissue was significantly reduced, and NF-κB was significantly activated. Different doses of Buyanghuanwutang decoction could decrease ROS content (P < 0.01), improve I-κBα protein and inhibit the activation of NF-κB. CONCLUSION: Buyanghuanwutang decoction can prevent atherosclerosis induced by Hcy via inhibiting NF-κB-dependent pathway. Copyright 2012 by the Chinese Pharmaceutical Association.
6.Influence of dexmedetomidine on SAS score and Ramsay score in patients with hypertensive cerebral hemorrhage after borehole drainage

Objective:To explore influence of dexmedetomidine on Riker sedation -agitation scale (SAS) score and Ramsay sedation score in patients with hypertensive cerebral hemorrhage after borehole drainage . Methods :A total of 96 patients with hypertensive cerebral hemorrhage after borehole drainage treated in our hospital were selected . According to analgesic drugs taken after operation ,they were divided into dexmedetomidine group (n=48) and mid‐azolam group (n= 48) .Riker SAS score ,Ramsay sedation score and adverse reactions after operation were com‐pared between two groups . Results:Compared with midazolam group ,there were significant rise in Ramsay seda‐tion score [6h :(1.59 ± 0.52) scores vs .(2.13 ± 0.15) scores] and significant reduction in Riker SAS score [6h :(4.09 ± 0.82) scores vs .(3.73 ± 0.76) scores] on 6h ,12h and 24h after operation ,P<0.05 or <0.01 ;and signifi‐cant reduction in incidence rate of adverse reactions (37.5% vs .10.4% , P< 0.05 ) in dexmedetomidine group . Conclusion:Dexmedetomidine possesses definite analgesic and sedative effects on patients with hypertensive cerebral hemorrhage after borehole drainage ,it's safe andreliable ,which is worth extending .
7.Study on immunotherapy by house dust mite allergen recombinants

Objective:To build murine asthma model with Dermatophagoides pteronyssinus. To compare the curative effects of the immunotherapy regimens with either Dermatophagoides pteronyssinus allergen extracts or a recombinant Derp2(rDerp2). To offer a novel thread for immunotherapy of allergic diseases.Methods:The BALB/c mice were divided into four groups randomly. They were: negative control, positive control, allergen extracts and rDerp2. Mice were sensitized by i.p. injection of mixture of allergen extracts and Al(OH)3 except the negative control,and were i.p. given a treatment with either allergen extracts or rDerp2 respectively, and then were i.n. given allergen extracts as a challenge. Negative control was treated by saline always. Assays for IgE,IgG1,IgG2a in serum,IL-4,IFN-? in BALF and SCCS were performed by ELISA. Total cell number and assorted cell number in BALF samples were determined. The lungs kept in 10% formalin were made into slices stained by HE in order to observe the pathologic changes in lungs.Results:Compared to the negative control, in the positive control the total cell number and the number of inflammatory cell in BALF were significantly increased, BALF eosinophil counts were significantly increased, neutrophils and eosinophils were the predominant BALF inflammatory; and the Th2 cytokine, e.g. IL-4, was the main cytokine in BALF and SCCS. Compared with the positive control, the allergic responses in the mice given relevant immunotherapy were obviously alleviated, and the predominant cytokine in BALF and SCCS was Th1 pattern, e.g. IFN-?.Conclusion:We established successfully the murine asthma model. Immunotherapy with Dermatophagoides pteronyssinus allergen extracts had better curative effects than immunotherapy with recombinant Derp2.
8.Effect of lipopolysaccharide on stimulating high mobility group box-1 protein expression in peritoneal macrophages in rats and its potential signaling mechanism

Objective To investigate the effect of lipopolysaccharide (LPS) on stimulating high mobility group box-1 protein (HMGB1) mRNA expression in peritoneal macrophages in rats and its potential signaling mechanism. Methods Abdominal macrophages obtained from male Wistar rats were seeded on 24-well (1?10 6 cells/well) tissue culture plates. The cells were incubated for 3 days before they were stimulated with LPS, fludarabine (specific inhibitor of signal transducer and activator of transcription 1, STAT1), or rapamycin (specific inhibitor of signal transducer and activator of transcription 3, STAT3). After being stimulated, macrophages were denatured directly in tissue culture plates to determine HMGB1 mRNA expression. The time-dependent and dose-dependent responses between LPS stimulation and HMGB1 mRNA expression were analyzed, and the effect of treatment with fludarabine and rapamycin on HMGB1 mRNA expression was also observed. Results After being stimulated with 75-100ng/ml LPS, the HMGB1 mRNA expressions in macrophages were up-regulated markedly, peaking at 24-36 hours, and remained elevated up to 48 hours. It was found that the HMGB1 mRNA expression was significantly inhibited by treatment with either fludarabine (100?mol/L) or rapamycin (25ng/ml). Conclusions These data suggest that LPS stimulation can result in up-regulation of HMGB1 mRNA expression in peritoneal macrophages in rats. Janus kinase/STAT pathway may be involved in modulating HMGB1 mRNA expression in LPS-activated macrophages.
9.Advances and problems in allograft tendon transplantation

Given the limited sources of autogenic tendon and the difficulties of the tissue engineering tendon to clinical application, the allograft tendon transplantation was a good way for tendon repairing defects. The domestic and foreign scholars have done a lot of studies on the acquisition, preservation, immunological characteristics, clinical application and prognosis of allograft tendon transplantation technology. The allograft tendon transplantation has been more and more used to repair the tendon tissue defects, but there still were some problems to be solved, such as the preservation, immunological characteristics, mechanical strength and postoperative adhesions.
10.The hearing protection of NGF to mimetic aging guinea pigs

OBJECTIVE To explore the function and mechanism of the apoptosis in cochlear of mimetic aging guinea pigs treated with NGF. METHODSThe twenty-four four-month-old guinea pigs which auricle reflex were sensitive were divided randomly into three groups: A group were injected to abdominal cavity with physiological saline (2 ml/kg) ; B group were injected to abdominal cavity with 5 % D-galactose (300 mg/kg) ; C group were given 5 % D-galactose (300 mg/kg-) and NGF (1000 U/kg) respectively to abdominal cavity. Three groups had been treated two times a day for four consecutive weeks. All the guinea pigs were tested with auditory brainstem response (ABR) before the experiment and twenty-four hours after the last injection. All the guinea pigs were killed by decapitation. The cochlears were removed and fixed in different fixatives. The specimens were treated by a series of special process and the Corti organs were tested with terminal deoxynucleotidy1 transmission-mediated dUTP nick-end-labelling (TUNEL) and transmission electron microscopy respectively. RESULTS The ABR threshold of C group was apparently lower than that of B group (P
