Journal of Shanghai Jiaotong University(Medical Science) 2006;0(04):-
Objective To clarify the combined ability of viral macrophage inflammatory protein (vMIP) to receptors through comparison research of vMIP binding to chemokine receptors of peripheral blood macrophages(PBMCs). Methods Combined ability of vMIP to receptors was measured with radioligand assay and identified with saturation, kinetic and specificity analyses. Results Kd is 11.1 nmol/L to human receptors of PBMC and 14.3 nmol/L to monkey receptors. The IC50 is 3.4 nmol/L to CCR5 and 4.5 nmol/L to CXCR4. Conclusion vMIP has high affinity with receptors and high specificity to CCR5 and CXCR4.This may be of great significance in the prevention and treatment of inflammation and HIV infection.

4.Inflammatory response and immune regulation of high mobility group box-1 protein in treatment of sepsis
World Journal of Emergency Medicine 2010;1(2):93-98
Sepsis is an infection induced systemic inflammatory response syndrome and is a major cause of morbidity as well as mortality in intensive care units. A growing body of evidence suggests that the activation of a proinflammatory cascade is responsible for the development of immune dysfunction, susceptibility to severe sepsis and septic shock. The present theories of sepsis as a dysregulated inflammatory response and immune function, as manifested by excessive release of inflammatory mediators such as high mobility group box 1 protein (HMGB1), are supported by increasing studies employing animal models and clinical observations of sepsis. HMGB1, originally described as a DNA-binding protein and released passively by necrotic cells and actively by macrophages/monocytes, has been discovered to be one of essential cytokines that mediates the response to infection, injury and inflammation. A growing number of studies still focus on the inflammation-regulatory function and its contribution to infectious and inflammatory disorders, recent data suggest that HMGB1 formation can also markedly influence the host cell-mediated immunity, including T lymphocytes and macrophages. Here we review emerging evidence that support extracellular HMGB1 as a late mediator of septic complications, and discuss the therapeutic potential of several HMGB1-targeting agents in experimental sepsis. In addition, with the development of traditional Chinese medicine in recent years, it has been proven that traditional Chinese herbal materials and their extracts have remarkable effective in treating severe sepsis. In this review, we therefore provide some new concepts of HMGB1-targeted Chinese herbal therapies in sepsis.

Objective To examine the relationship between the A118G polymorphism of mu-opioid receptor(OPRM1) gene and analgesic effects of oxycodone hydrochloride in patients with severe cancer pain. Methods Fifty-nine patients with severe cancer-induced pain were divided into 3 groups by genotype(AA group,AG group and GG group,23,28,8 patients, respectively). They were orally treated with oxycodone hydrochloride sustained-release tablets, and the treatment dosage and adverse reactions(including nausea, vomiting, dizziness, constipation, etc.)were compared between groups. Results The variation of allele frequencies(118G)was 37.3%.The dosage of oxycodone hydrochloride sustained-release tablets used in AA, AG and GG groups was(27.0±14.3),(36.4±22.5)and(55.0±35.1)mg,respectively,and the differences were statistically significant between groups( P = 0. 01). On the part of adverse reactions, the incidence of nausea and vomiting, dizziness, constipation was 28.8%,22.0%,and 52.5%,in AA,AG and GG groups, respectively, and there was no significant difference between groups(P>0.05). Conclusion The analgesic effect of oxycodone hydrochloride is affected by opioid receptor gene polymorphism.Patients with G allelic variation(AG or GG genotype)require larger doses of oxycodone hydrochloride than those with AA genotype. However, adverse reactions are not associated with polymorphism.

Objective To provide a clinical anatomic basis for the bi-pedicled gastrocnemius myocuta- neous flap which is to be used to repair defects of the Achilles tendon and posterior skin of the ankle. Methods In anatomic study, out of 30 cadaver specimens,bi-pedicled V-Y gastrocnemius myoeutaneous flaps were harvested to measure the downward sliding lengths of the flap when the knee was at flexion of different degrees. In clinic,12 patients with the Achilles tendon and posterior ankle skin defects were repaired with the bi-pedicled gastrocnemius myocutaneous flaps. The areas of composite defects ranged from 10 cm?6 cm to 6 cm?4 cm.They were followed up for four months to 12 years. Results The maximal sliding length of the flap reached (9.2?0.9)cm when the knee was at flexion of 90?.All the clinical flaps survived and ambulation of the patients recovered. The dorsiflexion and plantarflexion of the ankles reached 11.0??1.4?and 35.0??4.6?respectively.Conclusion This flap is suitable for one-stage repair of composite Achilles tendon defects.

ObjectiveTo analyze the relationship between result of hemorheology and non-pathological factors such as sex, age and life habit.Methods3483 healthy adults who had health examination were divided into different groups according to sex and age, and results of hemorheological test of them were analyzed and compared with reference values.ResultsAll hemorheological indexes of men were higher than that of women. The whole blood viscosity of female had an increasing trend along with the age increasing. However, the result of hemorheology of male showed that the index of the age of 30~49 was higher than the age of more than 50, and had a decreasing trend along with the age increasing after the age of 50. The index of high shear viscosity, low shear viscosity and hematocrit of both male and female were all higher than the reference values offered by apparatus.ConclusionEffect of non-pathological factors such as age, sex and life habit on index of hemorheology should be considered.

Objective To investigate the effects of Pulmicort Respules on the airway resistance and airway remodeling in rats with allergic asthma. Methods Thirty-two SD rats were randomly divided into blank control group, asthma model group,Pulmicort Respules group and normal saline control group, with 8 rats in each group. On the 14th day of allergization, rats were challenged by ovalbumin inhalation and were treated correspondingly. On the 28th day, airway resistance was determined,HE staining was conducted in lung tissues of each group, and airway remodeling related parameters were also measured. Results The airway resistance of Pulmicort Respules group was significant lower than that of blank control group, asthma model group and normal saline control group(P
