4.DETERMINATIONS OF cAMP, cGMP AND CORTISOL LEVELS IN BLOOD AND cAMP AND cGMP LEVELS IN THE TISSUES OF THE INTERNAL ORGANS IN EXPERIMENTAL SEPTIC SHOCK
Journal of Chongqing Medical University 1986;0(04):-
Forty-four mice were used in this experiment. The animal model of septic shock was produced by intraabdomcnal injection of the viable B. coli. The animals were divided into 5 groups. During the course of the development of the septic shock.some of the animals were killed and blood from inferior vena cava was withdrawn for the determination of cAMP, cGMP and cortisol. Tissues from both sides of the adrenal glands, lower pole of the left kidney, left ventricle and right lobe of liver were removed for determination of cAMP and cGMP. The results showed that, during the early stagenof septic shock, the levels of cAMP, cGMP and cortisol were raised, followed by steady lowering of these levels as the shock became more profound.Base on the results of this experiment the author pointed out that the large dose glucocor-ticosteroids may be useful during the early stage of septic shock
5.Research progress of DNA methylation in lens development and cataract
Chinese Journal of Experimental Ophthalmology 2017;35(8):747-751
Epigenetics is the study of genomic structural modifications that affects gene expression without DNA sequence change.Epigenetic mechanisms for the regulation of gene expression include DNA methylation, histone modifications, and microRNAs.DNA methylation may contribute to silencing gene expression which is a major mechanism of epigenetic gene regulation.DNA methylation regulatory mechanisms in lens development and pathogenesis of cataract represent exciting areas of research that have opened new avenues for association with aging and environment. This review concludes current understanding of the major mechanisms and function of DNA methylation in lens development, pathogenesis of age-related cataract, secondary cataract and complicated cataract.By understanding the role of DNA methylation in the lens disease and development,it may help to develop a new therapeutic approach to clinical treatment of cataract.

Chinese Journal of Biologicals 2024;37(11):1327-1333
Objective To investigate the regulatory effects of resveratrol on proliferation, adhesion, migration, invasion,epithelial-mesenchymal transition(EMT), and Janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3) signaling pathway of human glioma cells T98G, so as to provide theoretical basis for the related research of resveratrol in the treatment of glioma. Methods Human glioma T98G cells were cultured in vitro and divided into control group(without intervention), experimental groups(12.5, 25, and 50 μmol/L resveratrol groups), positive drug group(50 mg/L 5-fluorouracil), inhibitor group(50 μmol/L resveratrol + 50 μmol/L JAK2/STAT3 signaling inhibitor AG490) and activator group(50 μmol/L resveratrol + 0. 5 μmol/L JAK2/STAT3 signaling activator colivelin), which were intervened for 24 h. The cell viability, proliferation rate, adhesion ability, migration ability, invasion ability, EMT and JAK2/STAT3 signaling pathway related protein expression levels were analyzed by cell count(CCK-8), 5-ethynyl-2′deoxyuridine(EdU), adhesion test, scratch test, Transwell and Western blot. Results Compared with the control group, the cell viability of 12. 5 and 25 μmol/L resveratrol groups had no significant change(t = 1. 501 and 2. 167, respectively, P > 0. 05), while that of 50 μmol/L resveratrol group and the positive drug group decreased significantly(t = 7. 918 and 8. 020, respectively, P < 0. 05). The cell viability of 12. 5 and 25 μmol/L resveratrol groups increased significantly compared with the positive drug group(t = 6. 519and 5. 853, respectively, P < 0. 05), and that of 50 μmol/L resveratrol group was close to 50% and had no significant difference from that of the positive drug group(t = 0. 102, P = 0. 921, P > 0. 05). Compared with the control group, the proliferation rate, adhesion, migration, invasion ability and the protein expression of N-cadherin, vimentin, fibronectin(FN), p-JAK2,and p-STAT3 in 50 μmol/L resveratrol group and positive drug group significantly decreased(t = 4. 090-28. 510, respectively, P < 0. 05), while the expression of E-cadherin significantly increased(t = 16. 782 and 17. 291, respectively, P <0. 05). Compared with 50 μmol/L resveratrol group, the proliferation rate, adhesion, migration, and invasion ability, and the protein expression levels of N-cadherin, vimentin, FN, p-JAK2 and p-STAT3 of T98G cells in the inhibitor group significantly decreased(t = 3. 801-17. 980, respectively, P < 0. 05), while the expression level of E-cadherin protein significantly increased(t = 15. 256, P < 0. 05); the proliferation rate, adhesion, migration, and invasion ability, and the protein expression of N-cadherin, vimentin, FN, p-JAK2 and p-STAT3 of T98G cells in the activator group significantly increased(t =4. 235-27. 951, respectively, P < 0. 05), while the expression of E-cadherin significantly decreased(t = 14. 748, P < 0. 05).Conclusion Resveratrol can significantly inhibit the proliferation, adhesion, migration and invasion of human glioma T98G cells, which may be related to the inhibition of EMT process by inhibiting JAK2/STAT3 pathway signal transduction.

10.Early efficacy comparison between independent exercises under the doctors guidance and passive exercises by using CPM in rehabilitation after TKA
The Journal of Practical Medicine 2015;(7):1120-1122
Objective To compare the early efficacy of two different ways of rehabilitation exercises in total knee arthroplasty (TKA) patients. Method From January 2011 to January 2014,136 patients having undergone TKA were randomly divided into two groups. In group A , patients exercised under the guidance with doctor. In group B, patients exercised by using continuous passive movement (CPM) machine. Compared the data from the two groups including range of motion (ROM) of the knee joint, knee function score (KFS) and circumference of the knee joint, t test of independent sample was utilized in the research. Results All patientswere followed up in 4 weeks. The ROM and KFS of Group A was significantly better than Group B (P < 0.05). The knee circumference had no statistically significant difference between two groups (P> 0.05).Conclusion Group A and B are effective methods in TKA patients in early rehabilitation training. But training under the guidance of doctor has greater advantages than CPM.
