1.Chloroquine increased radiosensitivity of radioresistant MDA-MB 231 cells and its molecular mechanism

Objective: To study the genes associated with CQ(Chloroquine) increased radiosensitivity in radioresistant MDA-MB 231 cells.Methods: The survival curve of MDA-MB231 breast cancer cells was establish in the presence or absence of CQ after irradiation by colongenic assay.The relationship between certain protein expressions(MLH1,ERCC1,PARP,RAD51,XRCC4) and CQ increased radiosensitivity in radioresistant MDA-MB 231 cells was examined by Western blot method.The genes which are associated with CQ increased radiosensitivity were studied by microarray analysis.Results: CQ increased deaths of parental MDA-MB231 cells,radiosensitive MDA-MB231 cells and redioresistant MDA-MB231 cells were compared with those of the CQ-untreated control cells after irradiation,and the Radiation Potentiation Factor were 1.21-1.23.The expression of ERCC1 in the presence and absence of CQ in the radioresistant cells was significantly higher than that of sham-irradiated control cells by Western blot method.After irradiation,RAD51 level was increased up to 3-hour radiation and then gradually reduced.The expression of MLH1 was lower in 24 hours than that of other time points after irradiation.There were no differences on the expressions of MLH1,ERCC1,RAD51 in cells with and without CQ,which suggested that these genes weren't correlated with CQ increased radiosensistivity.XRCC4 and PARP levels were not changed by irradiation.Microarray analysis showed that TNP3,FRIH,CA21,ADTA genes might be related to CQ increased radiosensistivity in radioresistant cells.Conclusion: CQ can increase radiosensistivity in radioresistant MDA-MB231 breast cancer cells.It is not related to MLH1,ERCC1,PARP,RAD51,XRCC4 genes.(TNP3) may play an important role in CQ increase radiosensistivity in radioresistant MDA-MB231 breast cancer cells.
2.Prevalence and clinical value of the thymus superiorly extending to the neck in children

Objective To determine the proportion and the clinical value of the thymus extending superiorly into the lower neck in children.Methods The MR images of 400 normal children were retrospectively reviewed.According to the childhood staging criteria.all children were divided into four groups:the infancy and toddlerhood group(0-3 yo,42),the pre-school age group(>3-6 yo,87),the school age group(>6-12 yo,168),and the adolescence group(>12-18 yo,103).The presence or absence of the superior extension of the thymus above the manubrium Was determined on sagittal T2-weighted images.If so,the distance between manubrium and thymic apex was measured.The associations between frequency of ccrvicaI extension and age and sex were evaluated using correlation analysis.Results Superior extension of the thymus was detected in 288(288/400,72.0%)children,with 39(39/42,92.9%)in the infancy and toddlerhood group,72(72/87,82.8%)in the pre-school age group,118(118/168,70.0%)in the school age group,and 59(59/103)in the adolescence group(57.3%).There Was a statistically significant relationship between age group and frequency of cervical extension of the thymus (X2=4.9684,P<0.05).There was no significant decreasing trend in the mean distance of cervical extension between age increasing groups(F=2.28,P>0.05).Conclusions Cervical extension of the thymus occurs in the majority of children,and should be considered as a normal finding.Through recognizing this finding,doctors can avoid misdiagnosing it as a pathologic mass.
3.Follow-up study of peer education program on HIV/AIDS among senior high school students in Shanghai

Objective To evaluate the effects of peer education program on HIV/AIDS prevention among senior high school students. Methods Three senior high schools were randomly selected from Hongkou District of Shanghai as sample schools. One hundred and forty-three students from one class of each school were randomly selected as intervention group,and 152 students from another class of each school were served as control group. Six class hours of peer education program on HIV/AIDS prevention were conducted among intervention group. The same questionnaires which contained knowledge of reproductive health,HIV/AIDS and sexually transmitted diseases were administered before the program,one week and one year after the program. Results The knowledge scores before the program,one week and one year after the program in intervention group were 21.66?8.71,31.72?7.78 and 30.97?8.46,respectively,and those in control group were 22.32?9.24,22.61?8.66 and 22.51?9.16,respectively. There was no significant difference in the knowledge scores before the program between intervention group and control group,while there were significant differences in the knowledge scores one week and one year after the program between intervention group and control group (P

5.Effects of exercise training on myocardial mitochondrial miR-499-CaN-Drp-1 apoptotic pathway in mice.

OBJECTIVETo detect the levels of miR-499 and relative proteins in hearts of mice after exercise training, and investigate the mechanism of exercise-regulative apoptosis.
METHODSMale C57BL/6 mice were randomly divided into 3 groups( n = 14): sedentary (SE), exercise training 1 (ET1) and exercise training 2 (ET2) group. SE did not do any exercise. ET1 performed swimming training for 8 weeks. ET2 performed the same work as ET1 until the 5th week. Then, mice trained twice a day until the end of training. TUNEL assay was applied to test myocardial apoptosis, RT-PCR and Western blot were used to detect miR-499 and proteins levels respectively.
RESULTSCompared with SE, stress in ET1 failed to affect apoptotic index (AI) and miR-499-CaN-Drp-1 pathway (P > 0.05). In contrast, exercise load in ET2 increased miR-499 level, decreased Drp-1 level and AI with statistical significance respectively (P < 0.05), but neither CaN expression nor CaN activity was changed significantly (P > 0.05).
CONCLUSIONSwimming training can inhibit myocardial apoptosis, and the decrease in Drp-l may be responsible for the reduced myocardial apoptosis. CaN, the upstream protein, does not participate in exercise-regulative apoptosis.

6.Two different kinds of fixation materials in repair of distal clavicle fracture:stability of reconstruction

BACKGROUND:For distal clavicle fracture, clavicular hook plate and Kirschner wire tension band were used in the clinic, but they cannot keep the dual function of stability and functional protection. However, the application of microporous locking plate of distal clavicle makes it possible to double efficacy. OBJECTIVE:To compare and analyze the clinical outcomes of distal clavicle microporous locking plate and clavicular hook plate. METHODS:From March 2011 to March 2013, 40 patients with distal clavicle fractures were involved in this study. They were divided into two groups with the voluntary principles:distal clavicle microporous locking plate group and clavicular hook plate group. We compared and observed perioperative indicators (operation time, blood loss, and hospitalization days), clinical curative effects (University of California at Los Angeles scoring system and American Shoulder and Elbow Surgeons Scale), the score of shoulder joint function (Karlsson standard) and the overal treatment (removed the plate and functional training costs). RESULTS AND CONCLUSION:Al patients successful y underwent surgery. At 1 year after surgery, internal fixators were taken out. Necessary relevant rehabilitation training was performed for more than 3 months. There was no significant difference in perioperative indicators (P>0.05). However, clinical curative effects and the score of Karlsson shoulder joint function showed significant advantages in the microporous locking plate group (P<0.05). Although the distal clavicle microporous locking plate material cost was high, but there was no significant statistic difference in the overal treatment for better protection shoulder function (P>0.05). We draw the conclusion that distal clavicle microporous locking plate can effectively reconstruct the stability of the fracture site, and better retain the function of the joints at the same time.
7.The diagnostic value of ABCD2 for stroke in hospitalized patients with dizziness

Objective To evaluate whether ABCD2 score can discriminate dizziness patients with and without stroke. Methods This retrospective case-control observational study was conducted in 403 hospitalized patients. According to the final discharge diagnosis, the patients were divided into two groups:the stroke group and non-stroke group. The areas un-der the receiver-operator curves and 95%confidence intervals were then generated to estimate the diagnostic value. Re-sults ABCD2 score was higher in stroke group than in non-stroke group. Conclusions ABCD2 score can be used to quickly identify dizziness patients with cerebrovascular diseases.

10.Expression of MEK-1 in regulation of ?V integrin and E-cadherin of MCF-7 breast cancer cells

Purpose:Expression of MEK-1 was studied by regulating ?V integrin and E-cadherin on MCF-7 breast cancer cells.Methods:Using cell transfected techniques site-mutated genes were inserted into MCF-7 cells and made MCF-7 cells containing MEK-1-GFP,Ala 222 MEK-1-GFP,Asp 222 MEK-1-GFP ,respectively. These cells were tested with immunoblot analysis and immunofluoresence technique to determined the change of MEK-1 expression and location of MEK-1 in cells.Results:Deactivated Ala 222 MEK-1-GFP inhibited the activity of MAPK,active Asp 222 MEK-1-GFP increased the activity of MAPK. Ala 222 MEK-1-GFP increased the expression of E-cadherin,did not increase the expression of ?V integrin and inhibited FAK phosphorylation. MEK-1-GFP and Asp 222 MEK-1-GFP did not affect the expression of E-cadherin ,but upregulated the expression of ?V integrin,increased FAK phosphorylation.Conclusions:MEK-1 regulated the expression of ?V integrin and E-cadherin,FAK is the key factor that induced signal pathway of ?V integrin. It is possible that MEK-1 regulated FAK phosphorglation by regulating expression of ?V integrin and E-cadherin.
