1.Application of fetal DNA in maternal plasma in noninvasive prenatal diagnosis.

To explore the application of fetal DNA in maternal plasma for noninvasive prenatal diagnosis, the DNA template was extracted by hydroxybenzene-chloroform from 44 maternal (7-41 weeks) plasma. The Fetus-derived Y sequence DYZ-1 gene (149bp) was chosen to be amplified by PCR. The fragment was identified in all the plasma of male bearing pregnant women with the diagnostic accordance rate being 100.00%. Two of the 22 female bearing pregnant women had false positive results. Among the 44 pregnant women, the diagnostic accordance rate was 88.89% at early pregnant stage, 100.00% at medium pregnant stage, and 96.55% at late stage respectively. The final accuracy of 95.45% was obtained in all cases. It was concluded that by means of hydroxybenzene-chloroform extraction the authors of this article promoted the concentration and purity of the DNA template, and diagnosed more accurately. The results showed that free fetal DNA in the maternal plasma could be regarded as the gene resource for noninvasive prenatal diagnosis.

2.Neoadjuvant chemotherapy in patients with breast cancer

Objective To analyze the effect of neoadjuvant chemotherapy in patients with breast cancer , and further to explore its mechanism in order to provide a reliable basis for its clinical application .Methods 425 cases of breast cancer patients admitted from Jan .2010 to Jun.2014 were the research object , and they were divided into the control group(n=322)and the observation group(n=103).The control group received operation treatment and the observation group were added with neoadjuvant chemotherapy besides routine operation treat -ment.Incidence of clinical efficacy and adverse reactions of the 2 groups were statistically analyzed .Changes of vascular endothelial growth factor ( s-VEGF) and microvessel density ( MVD) levels before and after treatment were compared between the 2 groups.VEGF-A, the positive rate of VEGF-C and estrogen receptor(ER), progesterone receptor(PR), human epidermal growth factor receptor-2(HER2), and P53 before and after treatment were com-pared between the 2 groups.Results The total effective rate was obviously higher in the observation group than in the control group(77.7%vs 53.7%,χ2 =18.6, P<0.05).The adverse reaction rate had no statistical sig-nificance between the 2 groups(9.7% vs 5.3%, χ2 =2.57, P>0.05).s-VEGF and MVD had no statistical difference between the 2 groups before treatment(P>0.05)while they were significantly lower in the observation group than those in the control group after treatment (P<0.05).The positive rate of VEGF-A, VEGF-C, ER, PR, HER-2, and P53 had no statistical difference while they were obviously lower in the observation group than those in the control group ( P<0.05 ) .Conclusions NAC can help to improve the operation efficiency in pa-tients with breast cancer , reduce s-VEGF and MVD levels and play a role in reducing the positive rate of VEGF-A, VEGF-CER, PR, HER-2 and P53.It is an effective adjuvant treatment for breast cancer with less adverse re-actions, which is worthy of clinical application .
3.Effects of high-fat diet on insulin sensitivity in rats

Objective To observe the effects of high-fat diet on insulin sensitivity in rats. Methods Normal rats were fed with equicalorical high-fat diet and then treated with troglitazone. Glucose-insulin tolerance test was used to measure insulin sensitivity, blood pressure, blood glucose, insulin, endothelin, lipid, tissue glycogen and triglyceride levels. Results high-fat diet had no effect on body weight gain and blood pressure. Fasting and postprandial blood glucose levels were similar among the different groups. 4.The association of polymorphism of endothelial nitric-oxide synthase gene in pregnancy induced hypertension

Objective To study the variable number tandem repeats (VNTR) polymorphism in intron 4 of the endothelial nitric-oxide synthase (eNOS) gene and its relationship with plasma NO in pregnancy induced hypertension (PIH). Method The VNTR polymorphism in intron 4 of the eNOS genes of PIH and healthy control pregnant women were detected by PCR. The plasma level of NO~(3-)/NO~(2-) was measured by spectrophotometer, which was used as the index of NO synthesis.Data were analyzed according to the different type of genotypes and allelic gene frequency. 5.Determination of Berberine Hydrochloride in San Yu Ointment by HPLC

[Objective] To establish a content determination method of Berberine hydrochloride in San Yu Ointment by HPLC.[Methods] RP-HPLC was performed to determine Berberine hydrochloride on a column of Agilent Zorbax Eclipse XDB-C8(4.6?150mm,5?m)at 20℃; Acetonitrile-0.1 % H3PO4 (50∶50)(Adding 0.1 g C12H25NaO4S to every 100 mL)was used as mobile phase,the flow rate was 1.0 mL?min-1.The detection wavelength was at 265 nm.[Results]A good linearity can be found in the range of 39.40 ~394.0 ng,and the regression equation was Y = 4.073?103x+2.248 ? 103 (r=0.9992).The recovery was between 97.9 %~100.9 %,and the average recovery rate was 99.4 %,RSD was 1.5 %.[Conclusion]The method is simple,rapid,accurate and available,which can be used to control the quality of San Yu Ointment.
6.Surgical treatment for patients with postpancreatic vascular invasion by pancreatic head carcinoma

Objective To study the method of surgical treatment for patients with postpancreatic vascular invasion by pancreatic head carcinoma. Methods 25 patients with pancreatic head cancer and 1 with recurrent duodenal carcinoma with postpancreatic vessels involved by malignancy were treated by radical pancreatoduodenectomy with involved vascular resection(RPDIVR). Results There was no operation death in the 26 patients.Of the 26 cases, 4 had pancreatic fistula and 2 bile fistula postoperatively, but no vascular complication occurred. All of the 26 patients were followed up for 2.5-6.5 years( median time 4.5 years) and the average survival time was 56 months with 96.1%,88.4%, and 57.5% of 1,3,5 year survival rate. Coclusions (1)Colour Dopper imaging is a more useful and practical method in the preoperative examination of the vessels involved by malignancy. (2)When the postpancreatic vessels are involved by pancreatic head carcinoma, the rational treatment is RPDIVR. RPDIVR is a useful, safe, and an effective method and can improve the 5 year survival.
7.The association between polymorphism of TNF-alpha gene and hypertensive disorder complicating pregnancy.

To study whether the development of hypertensive disorder complicating pregnancy is associated with -308G-->A, -850C-->T mutation at promoter of TNF-alpha gene, the -308G-->A, -850C-->T polymorphism was examined in patients and healthy pregnant women by PCR-RFLP technique. The frequencies of genotype and allele were compared between the two groups. The results showed that with -308G-->A polymorphism distribution, the allele frequency of TNF2 and the frequency of the genotype TNF2/1 in the patient group was significantly higher in the patient group than in control group (P<0.05). A significant difference in genotype distribution of -850C-->T polymorphism was observed between the two groups. The allele frequencies of T in patient group was higher in the control group as compared with the patient group. The frequencies of CT and TT genotypes were lower in the patient group. It is concluded that the TNF2 allele of -308 is associated with the occurrence of hypertensive disorder complicating pregnancy, while T allele of -850 may be the protective factor against the development of the disease. TNF2/1 CC may be susceptibility genotype of hypertensive disorder complicating pregnancy.

8.Echocardiogram evaluation of cardiovascular functions of acute mountain sickness patient before and after returning plain

Objective To investigate the values of echocardiogram for evaluating the changes of left ventricular structure and cardiovascular function of the acute mountain sickness (AMS) patient before and after returning the plain.Methods A total of 33 light AMS patients returning to the plain in time underwent echocardiogram examinations on some cardiovascular indexes before and after returning the plain,including stroke volume (SV),cardiac output (CO),left atrial volume index (LAVI),left ventricular end-diastolic volume (LVEDV),early diastolic mitral flow velocity (E),late diastolic mitral flow velocity (A) and so on.Results The values of heart rate (HR),systolic blood pressure (SBP),diastolic blood pressure (DBP),CO,A and LAVI after going to the plateau were all significantly higher than those before going to the plateau (P<0.05),while the values of SV,E,E/A and LVEDV were obviously lower (P<0.05).The values of HR,SBP,DBP,CO,A,E and E/A recovered respectively 1 month after going to the plateau when compared with those before going to the plateau (P<0.05),while did not restore to the ones before going to the plateau (P<0.05),and the values of LVEDV,LAVI and SV reached the level before going to the plateau (P<0.05).The values of LVEDV,SV,SBP,LAVI and E 1 week after returning to the plain came to the level before going to the plateau (P<0.05),and the values of CO,HR,DBP and A were significantly higher (P<0.05) while the value of E/A was statistically lower (P<0.05).Conclusion Echocardiogram can be used to evaluate the left ventricular structure and cardiovascular function of the AMS patient,especially for those light AMS patients in acclimatization period or before and after returning to the plain.

99Mo is the main raw material of 99Mo-99Tcm generator,its production stability directly determines the supply of 99Tcm.There exists very limited number of irradiation and processing facilities for fission 99Mo all over the world and most of them are aging.Different facets of problems are emerging,such as those related to supply chain withdrawal,reaction target changes,and maintenance shutdowns.This paper focuses on the discussion of the current supply status of fission 99Mo worldwide,its related problems and possible solutions offered by the corresponding agencies of different countries,operational parties,and the authors themselves.
10.Report of one patient of cerebral infarction associated with methamphetamine and a review of pertinent literature

A case of methamphetamine-related cerebral infarction was reported and the relevant databases of CBMdisc and Medline were searched.Among 14 cases of ischemic stroke , the ratio of male:female was 9∶5 and the age range 19-45 years.There were anterior circulation lesions ( n=13,92.8%) and 10 of them (71.3%) had cerebral vascular occlusion or stenosis.And the predominant manifestation was hemiplegia (85.7%);among 34 cases of hemorrhagic stroke ,the ratio of male: female was 12∶5 and the age range 16-60 years.And 28 cases (82%) were≤45 years.Cerebral hemorrhage was the most common (85%).The major manifestations were headache ( n =26, 76%) and disturbance of consciousness (n=21,62%).The methamphetamine-related stroke occurs frequently in young males.And ischemic stroke tends to involve anterior circulation and cause hemiplegia.Yet hemorrhagic stroke has extensive attack sites and headache and disturbance of consciousness are quite common.
