Objective To investigate the correlation of interleukin-1 family genotypes,including interleukin-1 (IL-1α,IL-1β) and interleukin-1 receptor antagonist (IL-1Ra),with coronary heart disease (CHD) and serum lipoprotein level in the elderly.Methods Interleukin-1 family genotypes were detected in 318 elderly controls and 329 elderly CHD patients by polymerase chain reaction and restriction fragment length polymorphisms method.Serum levels of lipoproteins were inspected simultaneously.Results The TT and Ⅰ / Ⅰ or Ⅰ/Ⅳ genotype frequency of IL-1Ra was 90.3％ in elderly CHD patients,but 82.4％ in controls.Carriers with TT,Ⅰ / Ⅰ or Ⅰ/Ⅳ genotype of IL-1Ra were at an increased risk with an odds ratio of 1.98 in elderly CHD patients as compared with controls (x2=8.55,95％ CI:1.25-3.16).The TT and Ⅰ/Ⅰ or Ⅰ/Ⅳ genotype frequency of IL-1Ra was 96.2％ in elderly CHD patients with acute coronary syndrome,but 84.8％ in elderly CHD patients with stable angina.Carriers with TT,Ⅰ / Ⅰ or Ⅰ/Ⅳ genotype of IL-1Ra were at an increased risk with an odds ratio of 4.54 in acute coronary syndrome group as compared with stable angina group (x2=12.17,95％CI:1.81-11.36).The CT or TT genotype frequency of IL-1α-889 was 22.8％ in acute coronary syndrome group,but 7.6 ％ in stable angina group.Carriers with CT or TT genotype of IL-1α-889 were at an increased risk with an odds ratio of 3.59 as compared with stable angina group (x2 =14.93,95％CI:1.82-7.03).There were no significant differences in levels of serum lipoproteins among the different genotypes (P＞0.05).Conclusions In elderly patients with coronary heart disease,IL-1α(-889) CT or TT genotype carriers are at high risk for acute coronary syndrome,but IL-1Ra CC,TC,Ⅰ / Ⅱ or Ⅱ / Ⅱ genotype carriers are at a low risk for CHD or severe CHD.

Objective:To analyze genetic diversity of Fluconazole(FLZ)-resistant and -susceptible Candida albicans(C. albicans) strains isolated from patients with Sjgren's Syndrome (SS). Methods:30 C.albicans strains were isolated from the patients with SS and identified using standard criteria. Microdilution method was performed to determine the minimum inhibitory concentrations (MICs) of C.albicans to FLZ. A composite genotype was generated for each strain through random amplification of polymorphic DNA (RAPD) using three different primers, RSD10, RSD11 and RSD12. Results:The DNA fingerprinting profiles indicated genetic diversity amongst both the FLZ-resistant as well as -susceptible isolates, and no specific features emerged distinguishing the drug-resistant and -susceptible groups. Conclusion:These observations cast doubt on the theory of a clonal origin of FLZ-resistant C. albicans isolates. The emergence of FLZ resistance in SS patients may be associated with continuous exposure to FLZ.

Two lipase active fractions Lip1 and Lip2 were purified from the cell extract of Rhizopus chinensis CCTCCM201021.Both gave a single band on SDS-PAGE after using ammonium sulfate precipitation、Phenyl-Sepharose FF、DEAE-Sepharose FF and Sephadex G100 gel filtration chromatographies.The molecular masses of two lipases were 59.2kD and 39.4kD respectively.Lip1 and Lip2 showed optimal pH at 8.0 and 8.5 and their optimal temperatures were 40℃ and 35℃ respectively.The substrate specificity of the two lipases was obviously different.Lip1 was more specific to long chain fatty acid of p-nitrophenyl esters while Lip2 had a preference for the hydrolysis of short chain fatty acid of p-nitrophenyl esters.Lip1 had 1,3-position specificity for triacylglycerols hydrolysis while Lip2 had nonspecific position.Both lipases were stimulated by Ca 2+、Mg 2+ while SDS had strong inhibition on their activities.Lip1 and Lip2 had good stability in cyclohexane、hexane、heptane and isooctane(30% V/V).

Objective To evaluate the effects of surgical treatment for severe ocular rupture with blood staining of cornea and non-light perception, and analyze the relative factors. Methods Seven severely ruptured eyes of 7 patients with blood staining of cornea and non-light perception underwent second-stage penetrating keratoplasty combined with vitrectomy using temporary keratoprosthesis. All injured eyes had blood staining of cornea, hemorrhage in anterior chamber and vitreous, and choroidal and retinal detachment before the second-stage surgery. The average interval of the two surgical stages was 18 days (12-21 days). The preoperative visual acuity was non-light perception in all injured eyes with the mean intraocular pressure of 3 mm Hg (1 mm Hg=0.133 kPa) (2- 5 mm Hg). The mean follow-up period was 12 months (6-30 months). Results The postoperative visual acuity was better than light perception in 5 eyes with the best corrected visual acuity of light perception to 0.06. The retina was attached in 5 eyes (5/7). The mean postoperative intraocular pressure was 12 mm Hg (5- 15 mm Hg) which was significantly higher than the preoperative one (P

The recent advances in the methods for the isolation and manipulation of multiple classes of polyketide synthase genes from fungi were introduced. It is useful for discovery of novel fungal polyketide synthase gene clusters. These methods can also be useful for revealing the genetic potential of fungi to produce multiple types of bioactive polyketide.
