1.Serum levels of 25-(OH)D(3) and total IgE in children with asthma.

OBJECTIVETo study the changes and clinical significance of serum levels of 25-(OH)D(3) and total IgE in children with asthma.
METHODSThirty children with asthma, 40 children with asthmatic bronchitis, and 40 healthy children were enrolled. Double-antibody radioimmunoassay was used to detect the levels of serum 25-(OH)D(3) and total IgE.
RESULTSSerum 25-(OH)D(3) levels (18±3 ng/Ml)decreased significantly in the asthmatic group compared with those in the asthmatic bronchitis group (43±3 ng/mL) and the control group (43±3 ng/mL) (P<0.01). In contrast, serum total IgE levels (192±16 IU/mL) increased significantly in the asthmatic group compared with those in the asthmatic bronchitis group (123±14 IU/mL) and the control group (118±15 IU/mL) (P<0.01). Serum 25-(OH)D(3) levels were negatively correlated with serum total IgE levels in asthmatic children (r=-0.783, P<0.01). There were no correlation between serum 25-(OH)D(3) levels and serum total IgE level in the asthmatic bronchitis and the control groups.
CONCLUSIONS25-(OH)D(3) may play an important role in the pathogenesis of asthma. The increased serum 25-(OH)D(3) level may inhibit total IgE expression, suggesting that increasing serum 25-(OH)D(3) level might be a new option for the prevention and treatment of asthma.

Objective To explore protective effect of Heme oxygenase-1(HO-1) on irradiation-induced endothelial cell apoptosis.Methods Human endothelial cell line EA.hy926 were administered with or without HO-1 activator Copp and/or HO-1 inhibitor Znpp,respectively.Then,cells were treated with or without 8 Gy radiation.The HO-1 protein expression of cells were assessed with Western blotting and apoptosis of cells treated with irradiation were evaluated with flow cytometry.Moreover,cytochrome C releasing into cytosol were also determined by Western blotting. Results In PBS+R group,HO-1 protein expression of EA.hy926 was low posterior to irradiation.When cells were preconditioned with Copp and/or Znpp,then recieved with 8Gy irradiation,the HO-1 protein expression of EA.hy926 increased significantly in comparision with the PBS+R group(P
5.Observation and analysis the impact of different laparoscopic surgical positions on respiratory,circulatory functions and postoperative symptoms of patients

Objective To investigate the effect of different laparoscopic surgical positions on respiratory and circulatory functions,and postoperative symptoms of patients.Methods Totals of 120 consecutive cases with different laparoscopic surgery positions were included and were divided into three groups with 40 cases each group:reverse trendelenburg position group,trendelenburg position group,and were lateral position group.Heart rate,blood pressure,oxygen saturation,end-tidal carbon dioxide partial pressure,and peak airway pressure (Ppeak) were collected at three time points:5 mins before the start of surgery pneumoperitoneum (T1),10 mins after pneumoperitoneum (T2) and 5 mins after the end of pneumoperitoneum (T3).Postoperative symptoms were recorded as following up.Results In three groups,respectively,the differences in systolic blood pressure,diastolic blood pressure and respiratory carbon dioxide partial pressure (PETCO2) were statistically significant (P < 0.05).The blood pressure of T3 was significantly lower than that of T2 (P<0.01).The level of SBP and DBP of were significantly higher than that of T3 and T1 (P<0.01).No significant difference was detected in the heart rate change,and SpO2 of T2,T3 and T1 (P > 0.05).In reverse trendelenburg position group,the level of Ppeak of T2 was significantly higher than that of T1,while lower than that of T3 (P <0.01).In trendelenburg position group,the levels of Ppeak of T2 and T3 were significantly higher than that of T1 (P < 0.01).No significant difference was found in the levels of Ppeak of lateral position group (P > 0.05).And no significant difference was found in the incidence of abdominal discomfort symptoms between three groups (P > 0.05).The incidence of discomfortable symptoms of reverse trendelenburg position group,trendelenburg position group and ateral position group,respectively was 15.0%,42.5%,15.0%,and the incidence of shoulder paìn after surgery of trendelenburg position group was significantly higher than that of other groups (P < 0.01).Conclusions Pneumoperitoneum pressure and different positions can affect both the respiratory and circulation function of patients that undergoing laparoscopic surgery.And positions may affect the incidence of postoperative shoulder pain.
6.Effect of dietary categories on the intestinal cleanliness before the colonoscopy

Objective To explore the effect of dietary categories on effectiveness of the intestinal cleanliness before the colonoscopy,patients' acceptance of dietary changes and adverse reactions in bowel preparation.Methods Totals of 94 patients who underwent colonoscopy were divided into normal diet group (n =30),semi-liquid food group (n =32) and fasting group (n =32) according to the dinner diet styles the day before the colonoscopy.Observed three groups of adverse reactions,patients' acceptance and intestinal cleanliness.Results No significant difference was found in the medicine dosage,water intake,intestinal cleanliness among the three groups (P > 0.05).The cases of fatigue phenomenon and discomfort on dietary changes in normal diet group were 4 and 1,in semi-liquid food group were 14 and 8,in fasting group were 13 and 5,and the difference was statistically significant (x2 =7.765,20.623,respectively; P < 0.05).While no significant difference was found in the incidence of other adverse reaction among the three groups (P > 0.05).Conclusions Dietary categories are no effect on the intestinal cleanliness of the colonoscopy.However,according to the conditions of the patients before taking laxatives poly would increase their comfort under certain dietary restrictions.
7.The prevalence of type 2 diabetes mellitus and prediabetes in children.

OBJECTIVETo investigate the incident and prevalence of type 2 diabetes mellitus (T2DM) and prediabetes in obese children in the last ten years.
METHODSThe clinical data of hospitalized children with newly diagnosed diabetes mellitus (DM) or obesity between October 2000 and September 2011 were retrospectively studied.
RESULTSA total of 503 newly onset cases were diagnosed as DM in the past ten years, of which 31 were diagnosed as T2DM. The prevalence of T2DM in the second five-year duration increased significantly compared with that in the first five-year duration (0.18‰ vs 0.05‰; P<0.01). The number of cases of type 1 diabetes mellitus (T1DM) and T2DM increased by 1.35 fold and 4.20 fold, respectively in the second five-year duration. A total of 1301 obese patients received oral glucose tolerance tests, and 29 cases were diagnosed with T2DM and 255 cases with prediabetes. Of the 255 cases of prediabetes, 133 had dyslipidemia, 138 had non-alcoholic fatty liver disease and 53 had hypertension.
CONCLUSIONSThe prevalence rates of T1DM and T2DM increased significantly in the last 5 years. The prevalence of T2DM increased more significantly than T1DM. There was a higher prevalence of prediabetes in obese children. Childhood obesity predicts a higher risk of T2DM and cardiovascular disease in the future.

9.Characterization of a bioflocculant from a newly isolated Vagococcus sp. W31.

Screening of microorganisms producing flocculating substances was carried out. A strain secreting a large amount of bioflocculant was isolated from wastewater samples collected from the Little Moon River in Beijing. Based on the morphological properties and 16S rDNA sequence analysis, the isolate (designated W31) was classified as Vagococcus sp. A bioflocculant (named MBFW31) produced by W31 was extracted from the culture broth by ethanol precipitation and purified by gel chromatography. MBFW31 was heat-stable and had strong flocculating activity in a wide range of pH with relatively low dosage requirement. MBFW31 was identified as a polysaccharide with molecular weight over 2 x 10(6). It contained neutral sugar and uronic acid as its major and minor components, respectively. Infrared spectra showed the presence of hydroxyl, carboxyl and methoxyl group in its molecules. The present results suggested that MBFW31 had potential application in wastewater treatment.

10.Methylation of FHIT gene promoter region in DNA from plasma of patients with myelodysplastic syndromes and demethylating effect of decitabine.

This study was aimed to detect the methylation status of FHIT gene promoter region in the DNA from plasma of patients with myelodysplastic syndrome (MDS), and to investigate the demethylating effect of decitabine. Methylation-specific PCR method was used to detect the methylation status of FHIT gene promoter region in the DNA from plasma of 4 patients with MDS before and after treatment with decitabine plus semis CAG therapy (among them, 1 case of newly diagnosed MDS, 3 cases progressed into acute leukemia). The results indicated that 3 cases were found to have an increased methylation in the promoter region. After treatment with decitabine plus semis CAG, increased methylation was reversed in 2 cases. In 4 cases, 2 cases displayed clinical response. It is concluded that FHIT gene hypermethylation is associated with MDS pathogenesis. Decitabine has demethylating effect on the FHIT gene hypermethylation of plasma from MDS patients. Detecting the methylation status of FHIT gene in DNA from plasma may play a role in MDS auxiliary diagnosis or prognosis.
