Protective effects of chlorpromazine on ischemic heart and its mechanisms were investigated in rats with acute myocardial infarction (MI). 30 SD female rats were divided into three groups. the ischemia group, being subjected MI by coronary ligation, the MI+chlorpromazine group and the control. All samples were taken at 2 hours after operation. The ischemia group, as compared to the control, showed remarkable ultrastructural damage in ischemic myocardium and significant increases in serum CPKmb by 77%, serum cortisol by 67% and serum malondialdehyde (MDA) by 25%(P

3.Experimental study of in vitro construction of engineering myocardial tissue based on rat BMSCs induced by Ang II and accellular bovine pericardium

Objective To investigate the feasibility of inducing rat bone marrow mesenchymal stem cells ( BMSCs ) into myocardial cells by angiotensin II ( Ang II ) and the approach of applying the induced BMSCs to construct myocardial tissue in vitro. Methods BMSCs from rats were isolated and cultured. The third generation of BMSCs were incubated in complete culture medium, in which 0.1 μmol/L Ang II was added, for 24 h. The BMSCs were switched to complete medium and cultured. The control group was cultured by complete medium without Ang II . The protein cTnT and β-MHC were tested by immunofluorescence staining and the structure of induced cells was observed by transmission electron microscopy. The bovine pericardiums were managed by the detergent-enzyme digestion method. The induced BMSCs were planted onto the acellular bovine pericardium and cultured for 3 d before being tested. Results The cells of induced group were induced into cardiomyocytes by Ang II, which could express cTnT andβ-MHC, while cells in the control group couldn't. The result of electron microscopy showed that desmosome could be observed in the induction group. The surface of acellular bovine pericardiums was detected by scanning electron microscopy and no cells remaining on the surface except 2 μm-aperture were observed in the acellular bovine pericardiums. The induced BMSCs adhered to acellular bovine pericardium and grew well. Conclusions BMSCs can be induced into cardiomyocyte in vitro and express cTnT and β-MHC. The cells can adhere well on the surface and the inside of acellular bovine pericardium. BMSCs have the potential to form engineered cardiac muscle tissue with blood vessel.
4.Research progress of calcification and anti-calcification of implanted silicone rubber

Calcification of silicone rubber is a pathologic mineralization phenomenon which occurs on the surface after it is implanted in vivo,and there is no effective prevention method.The important process of calcification including these steps:phosphate ions combined with calcium ions to promote nucleating aggregation,slow increasing,fusion,and gradually form a larger accumulation of hydroxyapatite calcium.The research had been developed to the level of cells and genes,such as matrix Gla protein clusters is a calcification inhibitor of the tissues around silastic prosthesis.It is necessary to obtain a theoretical breakthrough in the aspect of organic template modulation of inorganic crystals.Silicone elastomer having its own local anti-calcification property had been paid more attention.Fully revealing the mechanism of calcification and calcification-resistant of silicone rubber will probably provide the basis for building a new anti-calcification material.
5. Application of cook MOB-15 system in guiding wire insertion during endoscopic retrograde cholangiopancreatography

Objective: To evaluate the value of Cook MOB-15 system in guiding wire insertion during endoscopic retrograde cholangtopancreatography (ERCP). Methods: The clinical data of 51 patients who received Cook MOB-15 system-guided wire insertion during ERCP between Jan. 2005 to Dec. 2007 were retrospectively analyzed. Forty patients who received conventional ERCP catheter for malignant jaundice between Jan. 2002 and Dec. 2004 were taken as control. The successful insertion rates were compared between the 2 groups. Results: The successful insertion rate was 90.2% (46/51) in the Cook MOB-15 system group and 72.5% (29/40) in the conventional group; there was significant difference between the 2 groups (P<0. 05). Conclusion: The Cook MOB-15 system can significantly raise the successful insertion rate in ERCP.
6.Clinical observation of glaucoma trabeculectomy with mitomycin C treatment

AlM:To explore the clinical outcome of trabeculectomy with mitomycin C ( MMC) on glaucoma.
METHODS: A total of 57 patients (95 eyes) of primary glaucoma were randomly divided into two groups, 31 patients ( 54 eyes ) in trabeculectomy with MMC ( T+MMC ) group, which received trabeculectomy with 0. 2mg/mL MMC in surgical sites, and 26 patients ( 41 eyes ) in trabeculectomy ( T ) group. The anterior chamber, bleb, intraocular pressure ( lOP ) and complications were observed. The post-operative follow-up periods ranged between 4 and 6mo.
RESULTS:The mean lOP was 11. 24 ± 3. 73mmHg on 1d in group T+MMC. There was the significant difference compared with preoperative lOP ( P<0. 01 ), while no difference with the group T (P>0. 05). At final follow-up, the lOP was significantly different between group T+MMC and group T (16. 15±3. 62mmHg vs 18. 79±5. 27mmHg, P<0. 05). The rate of bleb formation was 94. 44% and 80. 48%in group T+MMC and T, respectively (P<0. 01). The postoperative complications were seldom, hyphema and corneal edema, and were cured finally.
CONCLUSlON:Trabeculectomy with MMC for glaucoma can effectively reduce postoperative scar formation of the filtration passage and lower the lOP to a target level with fewer complications.
7. Influence of trichostatin A on apoptosis of colon carcinoma cells and expression of HIF-1α under hypoxia in vitro

Objective: To observe the influence of trichostatin A (TSA) on the apoptosis of colonic cancer cell line HT-29 cells and expression of HIF-1α in HT-29 cells under hypoxia in vitro. Methods: HT-29 cells were treated with TSA(100, 200, 400, 600, 800 nmol/ L for 12, 24, 48, and 72 h. Cells in the control group were treated with the same volume of DMSO. MTT assay was used to investigate the survival of cells in each group. Annexin V and TUNEL were used to determine the apoptosis rate of HT-29 cells. RT-PCR and Western blotting were used to study the expression of HIF-1α mRNA and protein under hypoxia (37°C, 1% O2). Results: MTT assay demonstrated that cell viability decreased significantly in TSA-treated group compared to control group(P<0.05), and the inhibitory effect of TSA was in a dose- and time-dependent manner. Annexin V and TUNEL showed that TSA induced early apoptosis of HI-29 cells; the apoptosis rates in TSA treatment groups (200, 400, 600, and 800 nmol/L) were significantly higher than that of control group(P<0.05). TSA suppressed protein expression of HIF-1α under hypoxia in a dose-dependent manner but had no influence on mRNA expression of HIF-1α. Conclusion: TSA can induce apoptosis of HI-29 cells and inhibit their proliferation through suppressing expression of HIF-1α at protein level under hypoxia condition.
8.Application of Intra-Operative Cholangiography

Objective To evaluate the application of intra-operative cholangiography in cholecystectomy. Methods A total of 442 patients with biliary calculi underwent cholecystotomy were reviewed. Results Of the 442 cases, 3 were false positive and 1 was false negative radiologically. The accuracy rate was 99.1%. Conclusion The results suggested that the application of intra-operative cholangiography could decrease the incidence of residual calculi and avoid unnessary exploration and trauma of the common bile duct.
9.Prevention and treatment for early complications of posterior lumbar spinal surgery

objective To analyze the causes of early complications following posterior lumbar spinal operations and to find out about the way for prevention and treatment of those complications. Methods From 1998 to 2002, 903 patients underwent posterior lumbar spinal operations ,there were 587 males and 316 females , with an average age of 36.7years (range, 18 to 78 years ). The diagnosis of patients included lumbar disc prolapse in 483, lumbar stenosis in 145, lumbar spondylolisthesis in 96, lumbar fracture in 94, lumbar instability in 27, lumbar tuberculosis in 15 spinal tumors in 24 and reoperation in 19. All patients were evaluated by the medical history, clinical examination and review of the imaging data, and then the surgical plan was made respectively according to clinical evaluation. Results Early complications of 78 complications within 2 weeks post-operatively occurred in 76 cases with the incidence of 8.6%(78/903), in-cluding of 49 males and 27 females with an average age of 44.6 years. The complications consisted of 50 nerve root irritations(64.1%, 50/78), most of which relieved at 4 to 30 days after treatment, 9 gastrointesti-nal symptoms(11.5%, 9/78), all of which disappeared at 3 to 10 days but 2 cases needed enema, 4 hematomas of incisional wound(5.1%, 4/78), 4 cerebrospinal fluid leakages(5.1%, 4/78), 3 urinary reten-tions(3.9%, 3/78), 2 deep infections(2.6%, 2/78), 2 deep vein thrombosis(2.6%, 2/78), 2 pulmonary em-bolisms(2.6%, 2/78),1 infection of urinary tract(1.3%) and 1 fatty liquidization(1.3%, 1/78). The opera-tional procedure possible leading to surgical complication were laminectomy, nerve root dissection and in-strumentation. Conclusion Early complications may determine the clinical result of the posterior lumbar spinal operations. Besides the skilled technique, thorough pre-operative planning, close observation and proper management after operation, and early post-operative rehabilitations are of great benefit to the pre-vention and treatment of the complications.
10.Biomechanical evaluation of the ped icle screw system in the treatment of lumbar spondylolisthesis

Objective To evaluate the biomechanical featu res of the HOIST in the treatment of lumbar spondylolisthesis.Methods Seven fresh cadaver specimens from L 2 to sacrum was used in the study.Each specimen was tested in five states:intact state as the control,spon dylolisthesis model by Panjabi meth od without fixation,spondylolisthesis model fixed with HOIST device,fatigued test of the HOIST and fixed with DICK system.During the experiment,the flexion,extension,bilateral bending and axial rotation loading we re applied to the specimens and measure d with three dimensional analysis ap paratus.A fatigue process of HOIST device was carried out with multifunctional test system(MTS 858,American).The ranges of motion(ROM)of spinal segments were determined with two special cameras and t test was used in statistical analysis.Results The ROM in spondylolisthesis state was significantly greater than that o f the others(P0.05).The difference among two HOIST grou ps and DICK group was not significant at the0.05level except the ROM of left axia l rotation(P
