Cell viability of Pichia pastoris was detected by flow cytometry (FCM) with two reagents fluorescein diacetate (FDA) and propidium iodide (PI). Compared with FDA/PI double-stained dot plots and PI single-stained dot plots,the latter could divide dead and living cells into two separate zones,and get the correct proportion. Then PI single-stained method was used to detect the change of cell viability in Pichia patoris fermentation. At glycerol batch and fed-batch phase,little dead cells were detected. At methanol fed-batch phase,cell viability decreased when cell weight increased,and was only 73.8% at 88 h.

Microbial is a renewable resource which can produce polysaccharide. Its unique physiological activities and broad applications are attracting increasing attention. In this article, the source and the fermenta- tion conditions of microbial polysaccharide was reviewed, with a view to provide a scientific basis for the production of the microbial polysaccharide.

Objective To explore the risk factors and protective factors for stomach cancer in Ganyu county.Methods Data of 319 pairs of new stomach cancer cases and health controls(1:1 matched)were studied by conditional Logistic regression method on the influential factors.Results The main risk factors for stomach cancer were the habits of eating hot foods(OR= 2.21,95%CI:1.57～3.12),the history of upper gastrointestinal inflammation(4.26,2.59～7.03),family history of cancer(2.29, 1.37～3.82);The main protective factors were often eating raw garlic(OR=0.64,95%CI:0.42～0.96),regular drinking tea(0.37, 0.25～0.54).Conclusion Unhealthy dietary habits,the history of upper gastrointestinal inflammation and family history of cancer are related to stomach cancer,while eating raw garlic and regular drinking tea may be effective in preventing the occurrence of stomach cancer.The results could be applied in stomach cancer prevention activities in Ganyu county.

Objective To construct and evaluate the effects of 4C characteristic home health care services model on the health of puerperas and fetuses.Methods A total of 162 puerperas and neonates were recruited,assigned by random nember table to routine community care condition and 4C characteristic home health care condition matched on the type and degree of health condition as well as the puerperas' culture background.The health condition of puerperas and neonates were investigated and compared between the two groups 42 days after delivery.Results The incidence of breast bilges,cracked nipple,re-visit the doctor,re-admission,anxiety and depression were significantly lower in the home health care group than routine community care group [3.7％ (3/81) vs.23.5％ (19/81),2.4％ (2/81) vs.17.3％ (14/81),2.5％ (2/81) vs.16.0％ (13/81),1.2％ (1/81) vs.16.0％ (13/81),(37.35±10.72) scores vs.(41.73±10.55) scores,(39.53±13.19) scores vs.(43.94±13.44) scores,x2=21.753,18.378,8.890,11.259,t=2.621,2.108,P ＜ 0.05 or 0.01].The success breastfeeding rates,correct rate of related knowledge and master degree of newborn,self-nursing newborn skills and puerperas and relatives' satisfaction scores were significantly higher in the home health care group than routine community care group [71.6％ (58/81) vs.37.0％ (30/81),95.1％ (77/81) vs.59.3％ (48/81),91.4％ (74/81) vs.53.1％ (43/81),（92.31±3.61) scores vs.(89.83±5.43) scores,x2=21.202,29.506,29.569,t=3.423,P＜ 0.01].In additon,the rate of re-visit the doctor,re-admission and omphalitis were significantly lower in home health care group than the routine community care group [2.5％ (2/81) vs.13.6％ (11/81),6.2％ (5/81) vs.23.5％ (19/81),4.9％ (4/81) vs.22.2％ (18/81),x2=6.774,9.172,9.288,P＜0.01 or 0.05].Conclusions 4C characteristic home health care reduced the probability of breast problems and unhealthy emotions for the puerperas,reducing the severity of jaundice as well as omphalitis for fetuses,reducing the probability of re-visit the doctor and re-admission for the puerperas and neonates,which can increase patients' satisfactory and promoting the mothers and their families to master self-nursing and newborn nursing skills and the other related knowledge.

Aim To explore the pharmacological mechanism of arsenic trioxide (ATO) on endoplasmic reticulum(ER) stress and provide indirect pharmacological indicators for the therapeutic drugs monitoring of acute promyelocytic leukemia.Methods The ATO toxicology database was utilized to associate gene data and establish protein-protein interaction networks.MCODE algorithm was used to analyze clustering of network topology relationship in order to find the critical functional gene group.The HL-60 cells were used as the model in this study.The cytotoxicity of ATO to HL-60 was evaluated by MTT assay.Real-time PCR assay was involved in detecting the expression of stress-related genes in ER stress.Western blot was used to detect the expression of stress-related proteins in ER stress.ER-specific staining was conducted by ER-Tracker Red.Cell differentiation and apoptosis was tested by flow cytometry.Results 206 ATO-related genes had 3 794 pairs of relationships,and ER stress-related module was outstanding among the top 4 gene cluster.ATO inhibited HL-60 cell proliferation in a dose-dependent manner;4 h after administration ATO could induce ER stress.The expressions of GRP78 and GRP94 were significant and the gene expression level changed greatly over time.The expression level of GRP78,GRP94 and Nrf2 was induced by 1 ～4 μmol · L-1 ATO,and the expression of CHOP was significant by 8 μmol · L-1 ATO.ATO could induce the phosphorylation level of eIF2α protein at 1 ～2 μmol · L-1 as well as a large expression of CHOP protein markedly at 8 μmol · L-1.As shown in ER-specific staining,ATO at 4,8 μmol · L-1 induced ER swelling.Flow cytometry showed that ATO could induce cell differentiation at 1 μmol · L-1,and the differentiation group was obvious at 2 μmol · L-1.So as to the concentration of 4 and 8 μmol · L-1,but the apoptotic group emerged at 8 μmol · L-1.Conclusion ATO can inhibit the proliferation of HL-60 and in this process,ER stress is involved,which shows a concentration-dependent phaseswitching effect.

OBJECTIVETo investigate the impact of 1, 25-(OH)2D3 on left ventricular hypertrophy (LVH) in type 2 diabetic rats.

METHODSType 2 diabetic mellitus (DM) model rats were established by intraperitoneally injecting with 30 mg/kg streptozotocin. After 8 weeks, 19 male rats were identified as diabetic with left ventricular hypertrophy (LVH) by ultrasound examination, and randomly assigned into three groups: untreated (DM-LVH, n=7), treated with insulin (DM-LVH+INS, n=6), and treated with 1, 25-(OH)2D3 (DM-LVH+VD, n=6). Healthy male rats were used as the controls group (n=6). The fasting blood glucose and the insulin level were determined weekly. The left ventricular mass index, myocardial collagen content, collagen volume fraction, and 1, 25-(OH)2D3-receptor level were determined by 4 weeks later.

RESULTSIn the DM-LVH model group, the insulin level was significantly decreased compared with the non-diabetic control group (P<0.05), whereas the blood glucose, left ventricular mass index, myocardial collagen content, collagen volume fraction, and 1, 25-(OH)2D3-receptor expression were significantly increased (all P<0.05). In the DM-LVH+INS and DM-LVH+VD groups, the insulin levels were significantly increased compared with the DM-LVH model group (P<0.05), whereas the other parameters were significantly decreased (all P<0.05).

CONCLUSION1, 25-(OH)2D3 could reverse LVH in diabetic rats and that the mechanism may involve stimulating insulin secretion and reducing blood glucose via direct up-regulation of 1, 25-(OH)2D3-receptor expression.

OBJECTIVETo analyse the correlation between the cellular immunity and TCM Syndrome typing of patients with minimal residual leukemia (MRI).

METHODSThe analysis was performed by detecting cellular immunological parameters in 30 MRI patients, 55 healthy persons and 36 patients with acute leukemia (AL) using three fluoresceine-conjugated monoclonal antibodies and flow cytometer.

RESULTSAs compared with those in the healthy persons, universal reductions of various parameters were shown in the MRI patients, including WBC count, absolute value of total T-lymphocytes, T-helper lymphocyte (P < 0.05) and total lymphocytes; the percentage and absolute value of NK cells (P < 0.01); and the percentages of total T-lymphocytes, CD4+ CD29+, T-suppressor cells and T-memory cells (P < 0.05 or P < 0.01), but without any rising of absolute value. As compared with those in the patients with AL, parameters were similar in the two groups with insignificant difference. The disturbances, including the lowering on ratio of T-helper/T-suppressor lymphocytes, in MRI patients of Qi-blood insufficiency type was the severest, that in the patients of Qi-Yin deficiency type was the mildest, and that in patients of Yin-deficiency with excess Fire type located between them.

CONCLUSIONThe immune function of MRI patients is low, belonging to the TCM Syndrome of vital energy deficiency with evil-lingering. Since the degree of cellular immune disturbance is different in various TCM Syndrome types, therefore, they should be treated with different dosages of different drugs.
