2.The dynamic change of Brn-3a expression in retina of chronic high intraocular pressure rat
Chinese Journal of Experimental Ophthalmology 2011;29(12):1077-1081
Background Brn-3a is a newly discovered specificity marker for retina ganglion cells(RGCs).It is well-known that RGCs damage is a important pathological basis of hypertension-visual disorder. But the study concerning expression of Brn-3a in RGCs in glaucoma eye is still rately. Objective The purpose of this work was to investigate the changes of Brn-3a expression in model eye with chronic high intraocular pressure(IOP) and its relation with morphology of retina and the expression of Brn-3a in chronic ocular hypertension rats. Methods Thirty-five clean adult SD rats were randomly divided into normal control group(5 rats) and model group(30 rats).Experimental chronic ocular hypertension models were induced unilaterally in the left eyes of 30 health adult SD rats by cauterizing super-scleral veins, and the conjunctival incision was made in the right eyes as sham operative group. The operated rats were subdivided into 6 groups according to the examination time points and 6 rats for each group.IOP was measured with Tono-Pen tonometer before and after 30 minutes,1,3,7,14,28 day after surgery respectively. The rat models were sacrificed in 1,3,5,7,14,28 days after operation by excessive anesthesia method, and retinal section was prepared for the histopathological examination and the RGCs were counted using Nissl staining method.Expression of Brn-3a in RGCs was detected by immunohistochemistry. This experimental complied with the Regulations for the Administration of Affair Concerning Experimental Animals by State Science and Technology Commission. Results The IOP was significantly raised from 30 minutes to 28 days after operation in model eyes with the top IOP( 34.46±4.65 )mmHg in the 30 minutes after operation, showing statistically significant differences in various time groups ( F =95.631,P =0.001 ) and different eyes ( F =287.473,P =0.001 ).Compared with sham operative group, the IOP were elevated from 1 day through 14 days after operation ( q =18.418,15.261,10.987,6.931,4.975,2.962,P < 0.05 ).The numbers of RGCs were ( 29.08 ± 1.98 ) in the normal control group and decreased gradually by 3.17%,7.84%,14.60%,22.40% in 1,7,14,28 day after surgery in the model eyes with the considerable differences in comparison with normal eyes ( t =5.943,8.034,15.023,17.004,19.371,P < 0.05 ).Immunochemistry revealed that Brn-3a was specially expressed in the RGCs layer and the positive RGCs for Brn-3a were evidently decreased as the prolong of high IOP duration ( F =127.583,P =0.000 ).Conclusions Chronic glaucoma model can be successfully established using Shareef-Sharma method. The loss of RGCs is associated with the high lOP duration. The expression level of Brn-3a is a reliable index of high-IOP-induced damage of optical nurve.

Objective To investigate the alteration of serotonin-producing gastric enterochromaffin (EC)cell in patients with functional dyspepsia(FD).Methods Fifteen healthy volunteers and 33 patients with FD were enrolled.Proximal gastric mucosal EC cells were countered after immunohisto- chemistry staining. The ultrastructure of EC cell was observed by electromicroscope. Results The EC cells in proximal gastric mucosa in patients with FD were significant higher than that in controls(12.5?2.1 vs 8.3?1.4,t=2.353,P<0.05),and the staining intensity of EC cell in patients with FD was also stronger than that in controls(3.72?0.42 vs 2.61?0.57,t=2.078,P<0.05).The more sever the gastric mucosal inflammation was,the more number of EC cells and the stronger staining intensity were.Under the electromicroscopy, more Golgi apparatus, mitochondria and endoplasmic reticulum were found in EC cells.Special secreting particles were also found in cytoplasm. Conclusions EC cells may be involved in the pathogenesis of FD.The number of EC cell is related with the severity of gastric mucosal inflammation.

Objective To investigate the CT features of pulmonary sarcoidosis. Methods Ninety patients with histologically proved pulmonary sarcoidosis were retrospectively studied by using CT scans and clinical recording. Results The main CT findings of pulmonary sarcoidosis were nodules which were seen in 69 cases(76.7%),and the nodules mostly distributed around the bronchovascular bundle(n=37, 41.1%).Other abnormalities included consolidation(n=31,34.4%),ground-grass(n=39,43.3 %), thickening of bronchovascular bundle(n=30,33.3%),interlobular septal lines(n=58,64.4%), fibrosis(n=17,18.9%)including bronchial distortion(n=8,8.9%),linear shadow(n=5,5.6%), and honeycombing shadow(n=4,4.4%),air-trapping(n=3,5.3%),bronchial straitness(n=8, 8.9%),pleural thickening(n=42,46.7%),and hilar and mediastinal adenopathy(n=76,84.4%). Two or more abnormal findings co-existed in 83 cases. The pulmonary lesions co-existed with hilar and mediastinal adenopathy in 76 cases. The nodules(n=25),consolidation(n=9),ground-grass(n=11), thickening of bronehovascular bundle(n=10)were improved after therapy.Ten cases of the interlobular septal(10/22),0 of bronchial distortion(0/4),1 case of diffuse linear(1/3),and 0 case of honeycombing(0/2)were improved. Conclusion CT manifestations of pulmonary sarcoidosis are varied, but has some specific radiographic features.A correct diagnosis can be made.combined with hilar and mediastinal adenopathy.

Objective To investigate the histological and biochemical responses associated with periprosthetic osteolysis, including the changes of tumor necrosis factor-?(TNF-?),osteoprotegerin(OPG) and os- teoprotegeria ligand (OPEL) in the interface membrane in rat models so as to understand the mechanisms of these interactions. Methods Twenty-four male SD rats were used in this study.A titanium alloy plug was inserted into each distal femur through the knee joint.Polyethylene particles were injected into the left knee at 2,4,6,8 and 10 weeks after implantation of the plugs.Saline solution was injected into the right knee as the control. The animals were killed 12 weeks after implantation. The distal femur was taken from each knee to make histological sections. The samples were made decalcified and stained with hematoxylin and eosin. The histopathological changes were observed under a light microscope. The concentrations of TNF-?in the membrane were assayed by a ELISA kit and the mRNA expressions of OPG and OPEL extracted from the tissue around the plug were detected by RT-PCR. Results The interface membrane taken from the experimental side was found to contain extensive fibrous tissues which extended deeply into the surrounding hone.TNF-?concentrations in the membrane of the experimental side were higher than in the control membrane. There was a significant decrease of OPG mRNA and a significant increase of OPGL mRNA in the membrane around the plug at the experimental side compared with the control side.Con- clusions Polyethylene particles can induce inflammatory responses around the titanium alloy plug. The increase of OPGL expression and reduction of OPG expression in membrane at the experimental side may be induced by TNF-?. Such changes might contribute to the differentiation of osteoclasts and result in the periprosthetic osteolysis.
