1.EFFECTS OF TRH AND INTERLEUKIN-1? ON CYTOSOLIC FREE CALCIUM MOBILIZATION IN RAT GH_3 PITUITARY CULTURED IN VITRO

The effects of TRH and recombinant human IL-1? on cytosolic free calcium mobilization in rat GH, pituitary cells cultured in vitro was studied spectrofluorometrically using Fura-2 as Ga++ indicator. The results showed that TRH 10-7~10-9mol/L stimulated the cytosolic Ca++mobilization in a dose-dependent manner. IL-1 alone in the concentration ranging from 0.01 to 10 U/ml did not increase the intracellular Ca++ level. 1- and 3-minute pretreatment of the GH3 cells with IL-1? 0.1 U/ml significantly inhibited TRH-induced Ca++ mobilization, whereas such suppressive effects were significantly diminished when IL-1/3 and TRH were added at the same time. These findings suggest that Ca++ may signal a TRH effect in pituitary cells and IL-1? influences pituitary function by inhibiting Ca++ mobilization induced by TRH.
2.Clinical Study of Dexmedetomidine Sedation in the ICU

Objective To observe the therapeutic effect and safety of dexmedetomidine for sedation in ICU. Methods We selected 40 patients who transferred to the ICU with retained endotracheal intubation after abdominal surgery under general anesthesia in this study. 40 patients were randomly divided into the dexmedetomidine group (D group,20 cases) and midazolam group (M group,20 cases) . Patients in group D were treated with loading dose 1 μg/kg from central vein pump injection 20 min, 1h according to Ramsay score for each adjustment of drug dose is pumped into 0.2 ~ 0.7 μg/(kg.h). Patients in M group were given a loading dose of midazolam 0.05mg/kg intravenous injection 1 min, depending on the degree of sedation maintenance dose of 0.02 ~ 0.1 mg/(kg.h). Pain scores every 2 h (NRS),when the NRS score>4 sharing,plus fentanyl 1μg/kg. When the patients achieved the offline condition, the endotracheal tube was pulled out. The sedation efficiency, dosage of fentanyl, after discontinuation wake-up time, extubation time, duration of mechanical ventilation, the incidence of cardiovascular events and delirium were compared between two groups.Results The sedation efficacy in Group D sedation was significantly higher than in group M ( <0.05) . Compared with group M, the dosage of fentanyl in patients in D group was significantly reduced ( <0.05), the wake time after stopping, extubation time were significantly shorter ( <0.05) . The incidence of cardiovascular events has no significant difference between two groups during the treatment. The incidence of postoperative delirium in D group was significantly lower than in that in group M ( <0.05) . Conclusions Dexmedetomidine has good sedative effect, and can shorten the time of extubation with hemodynamic stability and low incidence of delirium for patients in the ICU. It is an ideal ICU sedative.
3.DNA Sequence Features of Transfusion Transmitted Virus in Neonatal Infection

Objective To analyze the mutations in nucleotide sequences of transfusion transmitted virus(TTV) in neonatal infection.Methods Neonatal serum TTV-DNA was detected by a nested PCR technique.Fifteen Chinese neonates with positive TTV-DNA were diagnosed as TTV infection.ORF1 sequences of TTV-DNA from these neonates were determined.Results Homology of Chinese TTV(C01-C15) and Japanese TTV(N22)isolated ranged from 87.1%-97.7% at nucleotide level,but there were point mutations in Chinese TTV,such as GG→TT in locus 112 and 113,TTATC→CCTAT in locus 236-240.Conclusions Chinese and Japanese TTV isolated had the same genotype.Some gene mutations may increase the TTV pathogen,and result in neonatal hepatitis syndrome or hyperbilirubinemia.

Objective To investigate the protective effects of recombinant human erythropoietin(rhEPO)treatment on histopathologic changes seen in hyperoxia induced lung injury.Methods Rat pups were randomly divided into four groups:Ⅰ:air-exposed control group,Ⅱ: air-exposed+rhEPO-treated group,Ⅲ:hyperoxia-exposed control group,Ⅳ:hyperoxia-exposed+rhEPO-treated group.GroupⅢ and Ⅳ rats were exposed to 85% oxygen.GroupⅡand Ⅳ rats were received rhEPO (1 200 U/kg) subcutaneously on postnatal 0 day and 2 day.On postnatal 14 day,survival curve,measurement of body weight and lung weight,radical alveolar counts(RAC),microvessel count were compared,CD_ 31 and vascular endothelial growth factor(VEGF) were performed by immunostaining to assess hyperoxia-induced changes in lung morphology.Results Treatment of hyperoxia-exposed rats with rhEPO prolonged the survival and resulted in a significant increase in the weight gain of body and lung[(25.88?2.59) vs(18.8?3.93) P
5. Pharmaceutical care on a patient with lung cancer and trigeminal neuralgia

OBJECTIVE: To found appropriate approaches for a clinical pharmacist to carry out pharmaceutical care on patients with cancer pain. METHODS: The pharmaceutical care of a patient with lung cancer and trigeminal neuralgia was analyzed to explore how to make rational analgesia therapeutic schedule for patient with cancer pain. RESULTS: Clinical pharmacist actively participated in pharmacotherapy including evaluating analgesia therapeutic schedule according to guidelines, inquiring medication history, emphasizing compliance, providing medication education and rational medication recommendation. CONCLUSION: Pain can make patient anxious, depressed and somnipathy. Clinical pharmacist could help physicians to select rational drug, rational administration route and rational drug combination, pay attention to drug interaction, thus to make individual therapeutic schedule and ensure the rational use of analgesic drugs.
6.A clinical study of autologous tumor vaccine in patients with malignant tumors

Objective To evaluate the clinical significance and the mechanism of immunotherapy with autologous tumor vaccine. Methods 400 patients with pathological diagnosis of malignant tumor were enrolled in this study. 5 days after operation, the patients received vaccination of autologous tumor vaccine every 2-3 days for 5 times. 3 days before and 7 days after administrations, the peripheral blood mononuclear cells (PBMC) were isolated to assay the alteration in the proportions of CD3,CD4,CD8,CD4/CD8, and NK cells. Meanwhile, serum IL-2 and IFN-? were measured. Results After active immunotherapy with autologous tumor vaccine, the proportions of CD4 and CD4/CD8 were significantly elevated, while the proportion of CD8 and NK showed no difference. All the patients tolerated the vaccine well without serious side effects, such as skin ulceration. Conclusion The autologous tumor vaccine can elicit specific cellular immune response, and may be a potential approach to treat the cancer.

30 ?g/min were found to have retinopathy, but those with slightly increased UAE (15-30 ?g/min) showed a lower incidence of retinopathy similar to that of non-albuminuric diabetics. It indicates that UAE with 30 ?g/min may signal an imperative stage in preventing micro-vascular complications in diabetes.

Two parental strains BY-14 and BY-6,with high leavening ability in lean and sweet dough respectively,were selected.Through spore production and separation,two haploids with opposition types were selected for cross-breeding.At last one hybridization strain was obtained,with good fermentation ability as BY-14 in lean dough and better than BY-6 by 25%in sweet dough.
10.Different scoring systems to evaluate the prognosis of Fournier's gangrene: A comparative study.

OBJECTIVETo sum up the experience in diagnosis and treatment of Fournier's gangrene and find an optimal evaluation tool for its prognosis by comparing currently used prognostic scoring systems.
METHODSWe retrospectively analyzed 16 cases of Fournier's gangrene diagnosed and surgically treated in our hospital between 2004 and 2012. Using Fournier's Gangrene Severity Index (FGSI), Uludag Fournier's Gangrene Severity Index (UFGSI), Age-Adjusted Charlson Comorbidity Index (ACCI), and Surgical Apgar Score (sAPGAR) , we obtained the prognostic scores of the patients and made comparisons among different scoring systems.
RESULTSFGSI, UFGSI, ACCI, and sAPGAR were all clinically used scoring systems. Statistically significant differences were found in the scores of ACCI and UFGSI but not in those of FGSI and sAPGAR between the death and survival groups, with the maximum area under the ROC curve and minimum standard error for the ACCI score.
CONCLUSIONBoth ACCI and UFGSI are useful for evaluating the prognosis of Fournier's gangrene. However, ACCI is even better for its higher sensitivity and specificity and easier clinical collection.
