The research on regulation rules of traditional Chinese medicine (TCM) syndrome was the core of TCM syndrome information. It was thought that the constitution identification which provided the technology for evaluating health status was one of the bases in drawing the curve of conditional standard of syndrome. It was fitted and analyzed by the curve of conditional evolution of syndrome through establishing the curve of conditional standard of TCM constitution theory. It was the key technology of realizing the dynamical evaluation of TCM clinical effect.

OBJECTIVETo investigate the value of fractional nitric oxide concentration in exhaled breath (FeNO) in assessing the level of asthma control in children.

METHODSA total of 226 asthmatic children were divided into controlled asthma (n= 86), partially controlled asthma (n=63), and uncontrolled asthma groups (n=77). Ninety healthy children were enrolled as controls. FeNO was measured for both asthmatic and healthy children using the Swedish-designed NIOX system.

RESULTSThe control group had an FeNO of 14±6 ppb, the controlled asthma group had an FeNO of 29±26 ppb, the partially controlled asthma group had an FeNO of 32±30 ppb, and the uncontrolled asthma group had an FeNO of 40±32 ppb. The three asthma groups showed significantly higher FeNO than the control group (P<0.05). The uncontrolled asthma group showed significantly higher FeNO than the controlled asthma group (P<0.05), but there were no significant differences in FeNO between the partially controlled and uncontrolled asthma groups and between the partially controlled and controlled asthma groups (P>0.05).

CONCLUSIONSAsthmatic children have significantly higher FeNO than healthy children, and FeNO is correlated with the level of asthma control.

OBJECTIVETo investigate SiO2-induced EMT in human bronchial epithelial cells HBE in vitro.

METHODSHBE cells were cultured and then stimulated with indicated doses of SiO2 (0, 50, 100, 200, 300 µg/ml). The morphological changes were observed by microscope. In addition, Western blot was per-formed to detect the expression of E-cad, α-SMA and Vim. The changes of migration ability were examined by wound-healing assay in vitro.

RESULTS(1) After exposure to SiO2, HBE cells lost contact with their neighbor and displayed a spindle-shape, fibroblast-like morphology. (2) Compared with the control, the E-cad (300 µg/ml group) expression downregulated 2.98 fold (P < 0.05), and the Vim (300 µg/ml group) and α-SMA (200 µg/ml group) expression upregulated 4.46 fold and 3.55 fold (P < 0.05). There were significant differences between 100, 200, 300 µg/ml groups and the control group (P < 0.05). (3) In the test group, the percentage of wound-healing areas/wound areas were larger than those in control group (P < 0.05).

CONCLUSIONSSiO2 could induce EMT in human bronchial epithelial cells.

Objective To study the changes of permeability of blood brain barrier (BBB) in rats after acute traumatic brain injury and explore its mechanism. Methods Ninety-six male Wistar rats were randomly divided into sham-operated group (S group) and traumatic brain injury group (TBI group).Each group was divided into 6 subgroups (3,6,12,24,48 and 72 h after trauma).Brain injury animal models were established according to Feeney' s method.Measurement of Evans Blue (EB) was performed at different time points after injury to measure the changes of BBB permeability.Brain water content was tested by wet-dry weighting method. Expression of tight junction protein Claudin-5 was detected by Western blotting. Results As compared with that in rats of the S group,the brain water content and content of EB in rats of the TBI group were significantly increased at each time point (P＜0.05); and in TBI subgroups,the brain water content and content of EB were begun to increase 3 h after TBI,reaching its peak level 24 h after TBI.As compared with that in rats of the S group,the expression of Claudin-5 in rats of the TBI group was significantly decreased at 12,24,48 and 72 h after TBI (P＜0.05); and in TBI subgroups,the expression of Claudin-5 was begun to decrease 6 h after TBI,reaching its lowest level 24 h after TBI. Negative correlations were noted between protein expression level of Claudin-5 and both the brain water content and content of EB (r=-0.994,P=0.000; r=-0.846,P=0.036); positive correlation was observed between the brain water content and content of EB (r=0.863,P=0.027). Conclusion The degree of brain injury and changes of permeability of BBB may be time dependent in rats after acute brain traumatic injury; and Claudin-5 may be correlated with the changes of BBB.

Background/Aims: The involvement of long noncoding RNAs in the carcinogenesis of hepatocellular carcinoma (HCC) has been well documented by substantial evidence. However, whether cytoskeleton regulator RNA (CYTOR) could affect the progression of HCC remains unclear. Methods: The relative expression of CYTOR, miR-125a-5p and HS1-associated protein X-1 (HAX-1) mRNA in HCC cells were determined via quantitative real-time polymerase chain reaction. The viability of treated HCC cells was measured by Cell Counting Kit-8 assay. Cell apoptosis was estimated by flow cytometry analysis, assessment of caspase-9 activity and terminal deoxynucleotidyl transferase dUTP nick-end labeling staining, and Western blot of apoptosisrelated proteins. The interplay between CYTOR or HAX-1 and miR-125a-5p was validated by dual-luciferase reporter assay. Results: CYTOR was upregulated and miR-125a-5p was downregulated in HCC cells. CYTOR silencing inhibited cell proliferation and promoted cell apoptosis in HepG2 and SMMC-7721 cells.miR-125a-5p was sponged and negatively regulated by CYTOR, and HAX-1 was directly targeted and negatively modulated by miR-125a-5p. Overexpression of miR-125a-5p enhanced the repressive effects of CYTOR knockdown on HCC cells, and knockdown of HAX-1 enhanced the inhibitory effects of miR-125a-5p mimics on HCC cells. Conclusions CYTOR silencing facilitates HCC cell apoptosis in vitro via the miR-125a-5p/HAX-1 axis.

Based on the deletion information of high virulent PRRSV genome, 3 oligonucleotide primer were designed and synthesized. Specific and sensitive reverse transcription-PCR (RT-PCR) assays were de-veloped for the detection of high virulent PRRSV. The sensitivity and specificity of RT-PCR assays were evaluated, the results showing that the detection limit of the assay was found to be 0.265 pg of tissue total RNA, and the protocol have no cross-reaction with classical swine fever virus, porcine circovirus type 2,pseudorabies virus, streptococcus, haemophilus parasuis and Escherichia coli. Then 36 cell cultures, two PRRSV live vaccine strains and 184 clinical specimens from 52 farms were tested. Five PRRSV field iso-lates were the high virulent PRRSV; two PRRSV live vaccine strains from normal PRRSV, and 123 speci-mens from 42 farmer were positive (only 1 specimen was normal PRRSV). This RT-PCR method proved to be accurate differential diagnosis of the high virulent PRRSV and normal PRRSV with the characteristics of rapidity, sensitivity and specificity, and has a strong clinical relevance.

BACKGROUNDWilms' tumor (nephroblastoma) is a cancer of the kidneys that occurs typically in children and rarely in adults. Early diagnosis is very important for the treatment and prognosis of the disease. The aim of our study was to discover and identify potential non-invasive and convenient biomarkers for the diagnosis of Wilms' tumor.

METHODSNude mice were used to construct a Wilms' tumor model by injecting nephroblastoma cells into their bilateral abdomen. We collected 94 serum samples from mice consisting of 45 samples with Wilms' tumor and 49 controls. The serum proteomic profiles of the samples were analyzed via surface-enhanced laser desorption/ionization time-of-flight mass spectrometry. The candidate biomarkers were purified by high-performance liquid chromatography, identified by liquid chromatography-mass spectrometry, and validated using ProteinChip immunoassays.

RESULTSWe finally retrieved two differential proteins (m/z 4509.2; 6207.9), which were identified as apolipoprotein A-II and polyubiquitin, respectively. The expression of apolipoprotein A-II was higher in the Wilms' tumor group than in the control group (P < 0.01). By contrast, the expression of polyubiquitin was lower in the Wilms' tumor group than in the control group.

CONCLUSIONApolipoprotein A-II and polyubiquitin may be used as potential biomarkers for nephroblastoma in children, and the analysis of apolipoprotein A-II may help diagnose and treat Wilms' tumor.

OBJECTIVETo explore the relationship between occupational stress and blood glucose, Blood lipid and blood pressure.

METHODS108 video display terminals(VDT) operators who had the working experience were recruited to the study. The occupational stress indicator (OSI), the lever of blood glucose, cholesterol, triglyceride, lipoprotein of high density and lipoprotein of low density in serum were measured by using GOD-PAP, CHOD-PAP, GPO-PAP and PVS. The subjects were classified into three groups according to the score of occupational stress.

RESULTSThe contents of blood glucose of low, middle and high level of stress groups were (3.39 +/- 1.24), (3.59 +/- 1.26), (2.54 +/- 0.94) mmol/L respectively (F = 7.324, P < 0.01), and with the increase of level of stress, the content of blood glucose decreased significantly (r = -0.376, P < 0.01).

CONCLUSIONThe level of blood glucose of VDT operators is affected by occupational stress, among video display terminals and it can be used as the index for estimating occupational stress.
