1.Clinical value of susceptibility weighted imaging in the diagnosis of Parkinson disease

Objective To explore the application value of susceptibility weighted imaging (SWD) in the Parkinson disease (PD) diagnosis by analysis the changes of brain iron content of the basal ganglia and red nucleus.Methods Totally 56 cases of patients with PD and 40 cases of normal control group were diagnosed by SWI with the 3.0 T magnetic resonance imaging system,the corresponding phase image by using SWI processing software was obtained,the signal values of the various regions of the brain basal ganglia and red nucleus were measured and compared.Results In comparison of PD group with normal control group,the signal values showed differences in caudate (0.05±0.01 vs.0.04±0.01,t=1.991),putamen(-0.42±0.13 vs.-0.25±0.07,t=2.544) and globus pallidus(-0.23± 0.07 vs.-0.11±0.02,t=2.893) (all P<0.05).Conclusions Determining the iron deposition signal of the various regions of the brain basal ganglia by SWI is valuable in the clinical diagnosis of Parkinson's disease.
2.Effect of Wenyang Huoxue Lishul Recipe Containing Serum on Expression of Cathepsin L in Puromycin Aminonucleoside-induced Injury of Mouse Glomerular Podocytes.

OBJECTIVETo observe the effect of Wenyang Huoxue Lishui Recipe (WHLR) containing serum on the expression of cathepsin L (CatL) in puromycin aminonucleoside-induced injury of mouse glomerular podocytes.
METHODSMouse podocyte cells (MPCs) in vitro cultured were divided into the normal control group, the model group, the dexamethasone (DEX) group, 10% WHLR containing serum group, 20% WHLR containing serum group, the vehicle serum control group. MPCs in the normal control group were cultured at 37 degrees C culture solution for 24 h. 45 mg/L puromycin was acted on MPCs in the model group for 24 h. On the basis of puromycin intervention, 1 limol/L DEX was co-incubated in MPCs of the DEX group for 24 h; 10% or 20% WHLR containing serum was co-incubated in MPCs of the 10% WHLR containing serum group and 20% WHLR containing serum group for 24 h. The vehicle serum control group was also set up by incubating with WHLR containing serum alone for 24 h. The expression of CatL and its substrate Synaptopodin in podocytes were detected by cell immunofluorescence staining. FITC-conjugated phalloidin was used to stain F-actin. A cortical F-actin score index (CFS index) was designed to quantify the degree of cytoskeletal reorganization in cultured podocytes.
RESULTSCompared with the normal control group, the expression of synaptopodin significantly decreased and the expression of CatL significantly-increased in the model group. F-actin arranged in disorder, gradually forming pericellular F-actin ring. CFS index was obviously elevated (P < 0.01). Compared with the model group, the epression of synaptopodin increased, the expression of CatL decreased, and CFS index also decreased in the DEX group, 10% WHLR containing serum group, and 20% WHLR containing serum group (P < 0.05, P < 0.01). Compared with the DEX group, the expression of synaptopodin decreased in 10% WHLR containing serum group, CFS index also decreased in 20% WHLR containing serum group (P < 0.05).
CONCLUSIONSWHLR could up-regulate the expression of synaptopodin, down-regulate the expression of CatL, and alleviate cytoskeletal reorganization of F-actin. It was helpful to stabilize the cytoskeleton of F-actin and improve the merging of podocytes.

4. Effect of tempol, a free radical scavenger, on p38 activation in rats with renal ischemia reperfusion injury

Objective: To investigate the activation of p38 signaling transduction cascade in renal ischemia reperfusion injury (IRI) and to study the effect of tempol, a free oxygen radical scavenger, on p38 activation. Methods: Male Sprague-Dawley rats were randomly divided into sham-operation group (n=10), IRI group (n=45) and IRI + tempol group(n=10). Animal IRI model was created by renal pedicle ligation (50 min) of the left kidney along with a contralateral nephrectomy followed by 2 h reperfusion. Rats were sacrificed on 0, 5, 10, 15, 30, 45 min, 1 and 2 h after renal reperfusion. Animals in IRI + tempol group were pretreated with tempol (100 mg/kg) 1 h before undergoing the same protocol as in IRI group; the kidney was harvested after 45 min of reperfusion. Animals in the sham-operation group were subjected to contralateral nephrectomy without renal pedicle ligation and were sacrificed 45 min later. The renal p38 activities of the 3 groups were determined by Western blotting analysis. Malondialdehyde (MDA) content was detected and pro-inflammatory cytokine TNF-α, IL-1β levels were analyzed by ELISA. Results: Activation of p38 was observed in the kidney as early as 5 min after reperfusion and reached its peak 45 min after reperfusion and remained to be activated until 2 h after reperfusion(P<0.05). The activities of renal p38 in IRI and IRI + tempol group were markedly increased compared with that of the sham-operation group(both P<0.05). Pretreatment with tempol significantly inhibited IRI-induced p38 activation(P<0.05); it also decreased MDA activity and TNF-α and IL-1β levels (both P<0.05). Conclusion: Our results demonstrate that reactive oxygen species-mediated p38 activation plays an essential role in IRI-induced renal inflammatory damage in rats, suggesting that inhibition of p38 activation by tempol may be used for prophylaxis and treatment of IRI.
5.Study Progress of Cluster A in Homeobox Gene Family and Its Relationship with Leukemia

Homeobox(HOX) gene is a kind of regulatory gene,which can regulate the embryonic development and cell differentiation,and participate in the regulation and control of hematopoietic stem/progenitor cell proliferation,differentiation and maturation,etc.Cluster A in HOX gene family is the master gene for proliferation and differentiation of hematopoietic stem/progenitor cells,which has an influence on the number of hematopoietic stem cells and the differentiation of hematopoietic stem cell into erythroid,myeloid,megakaryocytic and lymphatic system,and has a relationship with the generation of leukemia.In this article,study progress of Cluster A in HOX gene family and its relationship with leukemia were briefly stated.
6.Purification of rat dental follicle cells by differential passage

Objective:To develop a simple culture and purifying method for rat dental follicle cells.Methods:The upper and lower first and second intact molar germs of SD rat were separated. Then, dental follicle and enamel organ were stripped together, minced into little pieces, digested with collagenase and cultured. Dental follicular cells were purified by differential passage and indentified by immunohistochemical staining of vimentin and cytokeratin. Results:The primary cells were mixed, consisting of dental follicle cells and enamel organ cells. After differential passage, the cells of fourth passage became purified dental follicle cells. Purified dental follicle cells were elongated spindle or triangle in shape, positive for vimentin and negative for cytokeratin.Conclusion:Dental follicle cells can be purified by several differential passages from the mixed primarily cultured cells.
7.Role of ?-endorphin in the regulation of ConA-induced IL-2R expression and IL-2 production in rat splenic cells following traumatic hemorrhagic shock

Objective To investigate the role of plasma ? endorphin(? EP) in the regulation of cellular immunity, mainly IL 2R expression and IL 2 production in rat splenic cells, following traumatic hemorrhagic shock(T HS). Methods ①Wistar rats with T HS were used and sacrificed at 0, 1, 3, 6, 12 and 24 h after T HS. Plasma specimens were collected and ? EP levels in plasma were detected. Rats with sham operation were served as the controls. ②Models of in vitro experiment were used. Spenic cells were isolated and mixed from four normal rats and cocultured with shock plasma or shock plasma+? EP antiserum. ConA induced splenic cell IL 2 production and IL 2R expression were observed. Results ①Level of plasma ? EP increased remarkably after T HS immediately, peaked at 1 h, showed decreasing tendency and restored to pre shock level 24 h after T HS. ②Shock plasma significantly suppressed ConA induced splenic cell function; Levels of plasma ? EP were negatively correlated with IL 2 production and IL 2R expression in spenic cells; Compared with that in SP group, splenic cell function elevated markedly in SP+? EP antiserum group, but was still lower than that in controls. Conclusion The increased plasma ? EP following T HS is involved in the suppression of cellular immunity.
8.Analysis of the cause and strategy for air quantity signaling in old patients using mechanic air

Objective: To explore the cause and the strategy for air quantity signaling in old patients using mechanic air. Method: Summary and analysis of the causes for air quantity signaling in 187 cases of old patients using respiratory machine. Results: One hundred fifty-one cases showed low-limited air quantity signaling per minute whereas 36 cases presented high-limited air quantity signaling per minute. Conclusion: Judging and removing the obstacle in respiratory machine on time and accurately are the key point raising both the survival rate of severe patients and the successful rate of mechanic air.
9.Computer-assisted full planning system for long bone fracture treatment with intramedullary nailing:an experimental study

Objective To evaluate the efficacy and clinical feasibility of the computer-assisted full plan- ning system for tibial fracture treatment with intramedullary nailing.Methods After analyzing the functional structure and operative procedures of the system,nine plastic tibia and 12 cadaver lower limbs were used for image mosaicing based on C-arm (PHILIPS BV Libra) fluoroscopic images in the operation room to assess the correctness of the mosaicing and planning algorithms.The plastic tibial model was used for analysis of the mosaicing precision.The cadaver tibial bone was used for reduction experiment with the reduction mechanism to analyze the operation feasi- bility.Results Only 7 to 10 [fluoroscopy time:(19.75?0.61)s] valid C-ann projection images were needed to produce a long bone panorama of the lower limb.The total time for image acquisition and mosaicing was within (4.17?0.86)minutes and the mosaicing precision in the plastic tibial model was (1.26?0.76)mm.The opera- tion of the reduction mechanism was very simple and could be controlled by a surgeon automatically or free of hand. An integrated reduction strategy could be produced for rough positioning in general and elaborate operations in de- tails.Conclusion The computer-assisted full planning system can be used for anatomical analysis based on the C-arm panorama,full surgical planning,virtual simulation,selection of proper intramedullary nails and fracture reduction in treatment of long bone fractures.
