1.Repair and reconstruction mechanism of stromal cell derived factor-1 on rat abdominal aorta grafts

Objective To investigate the repair and reconstruction mechanism of stromal cell derived factor-1(SDF-1) on rat abdominal aorta grafts.Methods Male Sprague-Darley rats received abdominal aorta grafts from male Wistar abdominal arteries.Immunohistochemical staining(IHC) was performed to detect the SDF-1 expression.The expression of CXCR4,the receptor of SDF-1,was demonstrated by RT-PCR.(Results) SDF-1 was expressed on the endothelium all the time during the course of the abdominal aorta grafts in the bodies of recipients.Furthermore,it was found that there was a significant relationship between the expression of SDF-1 on the endothelia and the thickness of the neointima of the rat abdominal aorta grafts.On the other hand,CXCR4 was detected within the abdominal aorta grafts.Conclusions Stem cells could be mobilized by SDF-1 to the abdominal aorta grafts and differentiated into smooth-muscle like cells.Furthermore,SDF-1 is predictive of grafts arteriosclerosis and the subsequent development of chronic graft dysfunction(CGD).
2.Preliminary Exploration on Time Limits of Quality Guarantee for Barrelled Purified Water Preservation

Objective To study the variations of microorganism indexes of barrelled purified water during its preservation period of 90 days. Methods The total count of bacteria, the mould and yeast count, the conductance rate and pH value in water samples collected from the fresh products of barrelled purified water at the same day of production and then at the 5th, 10th, 20th, 40th, 60th and 90th days after the production date respectively were determined. Results The total count of bacteria of barrelled purified water reached peak at the 5th and 20th days during the observed preservation periods, and then decreased gradually. No significant variations of mould and yeast count, decrease of pH value (6.3 to 6.0), increase of conductance rate (4.2 to 5.8 ?s/cm) were observed in the barrelled purified water during the whole 90_day preservation period. Conclusion The total count of bacteria of barrelled purified water was fluctuated obviously in the observation period. A suggestion was made that the time limit of quality guarantee for preservation period of barrelled purified water must be within 30 days.
3.Comparative genomic hybridization of primary lung cancer

Objective To analyze the unbalanced state of hereditary material of squamous carcinoma, adenocarcinoma and other lung cancers. Methods Fifty-five patients suffered from lung cancer were included in this study, including 24 of squamous carcinoma, 13 of adenocarcinoma, 5 of adenosquanous carcinoma, 8 of bronchioalveolar carcinoma, 4 of small cell lung cancer and 1 of atypical carcinoma. The technology of comparative genomic hybridization (CGH) was used. The normal DNA was obtained from 20 healthy male volunteers. Results The common extension region of squamous carcinoma was 2q, 5p, 11q and 22q, and the common deletion region was 1p, 4q, 5q, 6q, 8p, 9p, 10q, 11p, 13q, 18q and 21q. The common extension region of adenocarcinoma was 5p, 8q and 11q, and the common deletion region was 10p and 19. Conclusion The hereditary material of squamous carcinoma, adenocarcinoma and other lung cancers was unbalanced. The extension and deletion of chromatosome were the base of the occurrence of different lung cancer.
4.Biologic characteristics and chromosome karyotype analysis of lung adenocarcinoma cell strain induced by cis-diaminodichloroplatin

Objective To establish lung adenocarcinoma drug resistance cell strain induced by cis-diaminodichloroplatin and investigate the biologic characteristics and chromosome karyotype of cell strain.Methods Large dosage impact and step by step inducing method were used to establish A549 and SPC-A-1 drug resistance cells: A549/CDDP and SPC-A-1/CDDP.MTT was used to detect the cell drug resistance index. Bioluminescence was used to detect the energy metabolism.Cell cycle was analysed by flow cytometer.The differential staining technique and SKY were used to analyze the chromatosome of A549/CDDP and SPC-A-1/CDDP.Results The drug resistance index of A549/CDDP and SPC-A-1/CDDP were 14.0 and 12.0. The content of ATP,ADP and AMP decreased significantly.The cell cycle of A549/CDDP and SPC-A-1/CDDP were of no notable changes.The results of the differential staining technique and SKY showed that the chromatosome of A549/CDDP and SPC-A-1/CDDP had several derivative chromosomes.Both cells had the same derivative chromosome: der(21)t(21;22).Conclusion CDDP can induce lung adenocarcinoma cell strain drug resistance.The derivative chromosome,including der(21)t(21;22) may have relationship with the drug resistance of cell strains.
5.Comparison of field molluscicidal effects of four molluscicides

Objective To compare the field molluscicidal effects of 25%suspension concentrate of nielosamide ethanolamine salt(SCNE),50% wettable powder of niclosamide ethanolamine salt(WPN),40% META-Li and 50% Rongbao powder,so as to provide scientific basis for choosing the safe and highly effective molluscicide in field.Methods Four snail habitats with the similar snail status were selected and sprayed with the four molluscicides mentioned above,respectively.Then the snail status in the four fields was surveyed 3,7 and 17 d after the spraying and the moiluscicidal effects of difierent molluscieides were cornpared.Results The corrected mortalities of snails in the field sprayed with SCNE 3,7 and 17 d afterthe spraying were the hiishest among the four fields,with the rates of 93.13%,91.85% and 85.28%,respectively.Conclusion SCNE has a high molluseieidal effect in the field and can be further used in snail-infested areas.
6.Establishment of a multidrug resistance cell line from human lung adenocarcinoma cells and its biologic features

Objective To establish a multidrug resistance cell line from human lung adenocarcinoma. Methods The human lung adenocarcinoma cell line SPC-A-1 was exposed to cisplatin of a high and then increasing concentration for 192 d to establish multidrug resistance cell line (SPC-A-1/CDDP). The relative resistance was tested with MTT assay. The morphology of the cells was observed with transmission and scanning electron microscopy and the chromosome of them was analyzed with Giemsa stained specimens. Results The resistance index of SPC-A-1/CDDP cells to cisplatin was 11.2 and the cells showed various cross-resistance to 5-Fu, doxorubicin, mitomycin, vincristine and etoposide, but not to hydroxycamptothecine. Electron microscopy showed the cells with irregular and enlarged nuclei and abundant microvilli. Conclusion A multidrug resistance cell line (SPC-A-1/CDDP) from human lung adenocarcinoma is established. It can be used to downstream experiment.
7.Synthetic process of telavancin

New process reported the synthesis of telavancin using decyl alcohol as the starting material.After methanesulfonylation,nucleophilic substitution with ethanolamine,Fmoc protection,and Parikh-Doering oxidation,the intermdiate N-(9-fluorenyl-9-methoxycarbonyl) decylaminoethyl acetaldehyde (5) was obtained.Vancomycin hydrochloride and intermediate 5 were then subject to reductive amination reaction,Fmoc deprotection,and Mannich reaction to give the telavancin product.The process have been deeply studied on the two key reactions of reductive amination and Mannich reaction,and the reaction conditions have been optimized.The overall yield is 46% based on starting material vancomycin.
8.Safety and feasibility of low?energy X?ray radiotherapy in breast conserving surgery for early?stage breast cancer

Objective To investigate the short?term complications and cosmetic outcomes of radiotherapy in breast conserving surgery for early?stage breast cancer. Methods A retrospective analysis was performed on clinical data from 30 patients with early?stage breast cancer from 2013 to 2015. All patients underwent breast conserving surgery combined with intraoperative low?energy X?ray radiotherapy. The prescribed dose was 20 Gy at the surface of the applicator. Local complications, radiation injury, and cosmetic outcomes were observed after surgery. Results No grade 3?4 adverse reactions were found in patients. In terms of short?term complications, four patients (13%) had seroma, two of whom needed suction treatment;three patients ( 10%) had grade 1?2 skin erythema. Half of the patients had excellent cosmetic outcomes. None of the patients had local recurrence or distant metastasis. Conclusions Intraoperative low?energy X?ray radiotherapy is safe and feasible in breast conserving surgery for breast cancer. It can be considered as an optional approach for tumor bed boost in some patients with early?stage low?risk breast cancer.
9.Clinical Analysis of 20 Children with Ganglioneuroma

Objective To increase the preoperative diagnosis rate,reform the operation and improve the prognosis in children with ganglio -nauroma.Methods Clinical data of 20 children with ganglioneuroma hospitalized in Tongji hospital of Huazhong university of science and technology from Nov.1986 to May.2006 were reviewed(male 9,female 11).The biological,clinical and pathological characteristics of patients were analyzed.Results Among these 20 patients,15 cases were discovered to have no clinical symtom.The B ultrasound showed low-echo in all 16 cases.Under CT scan,20 cases showed low to moderate density shadows,and the distinct enhancement in these tumors was noted with enhanced CT scan.Fourteen cases undergoing MRI all showed low signals in T1W1 and inhomogeneous high signals in T2W1.All patients underwent surgical operations,completely removed in 14 cases,partly in 4 cases, and biopsy was performed only in 2 cases.Fifteen cases were followed up from 7 months to 6 years,tumor recurrence happened only in 1 case.Conclusions Ganglioneuroma is mostly found in chest-abdominal sympathetic chain,it grows up slowly and seems to be innocent.The imaging data of B ultrasound,CT and MRI can give helpful information in the diagnosis of ganglioneuroma.We shall pay attention to the choice of incision and avoidance of injuring the important blood vessels during the operation.Children who have neuroblastoma cells should receive prophylactic chemoprophylaxis treatment and the result of long-time follow up is satisfactory.
10.The expression of Twist protein in adenocarcinoma of prostate and its relationship with prognosis

ObjectiveTo investigate the expression of Twist protein in adenocarcinoma of prostate and clinicopathological features and explore its roles in the carcinogenesis and progression of adenocarcinoma of prostate and its relationship with prognosis.MethodsThe expression of Twist was detected in 70 cases with adenocarcinoma of prostate and 30 cases with side cancer tissue by immunohistochemistry (EnlivionTM).ResultsThe expression of Twist protein in adenocarcinoma of prostate was higher than side cancer tissue.The expression of twist was related to Gleason grade,clinical stage and bony metastasis (P<0.05),but it was not related to the age (P>0.05).The survival rate of Twist protein positive patients was significant lower than negative patients(x2=9.52,P<0.01).ConclusionsTwist might be an important biological marker for malignant transformation and invasion and metastasis adenocarcinoma of prostate,and the up-regulation of the expression of Twist reflects more invasion and poor prognosis.
