1.Effect of botulinum toxin A combined with delivery autologous platelet rich plasma into skin with microneedle for facial rejuvenation

Objective To observe the effect of botulinum toxin A combined with delivery autologous platelet rich plasma into skin with microneedle for facial rejuvenation.Methods 82 patients from our hospital treated for facial rejuvenation were randomly divided into observation group(41 cases)and control group(41 cases).The control group was given delivery autologous platelet rich plasma into skin with microneedle therapy.The observation group was treated with botulinum toxin A on the base of control group.After the treatment,the effect was observed.The facial skin moisture,skin elasticity,grease and spots were detected before and after treatment in the two groups.Before and after treatment,the role function,emotional function,social function,body function and the overall quality of life score were detected by the quality of life table.Observe all of the patients reaction duration of therapy.Results The total effective rate was 95.1% in observation group,significantly higher than the control group(P<0.05).After the treatment,the facial skin moisture and skin elasticity scores were(51.24±8.31)and(62.73±8.59)in observation group,significantly higher than the control group(P<0.05),while the grease and spots scores were(41.87±7.47)and(42.13±7.62)in observation group,significantly lower than the control group(P<0.05).After the treatment,the role function,emotional function,social function and the overall quality of life score were(71.08±8.75),(66.21±7.04),(81.80±13.42)and(75.18±7.97)in observation group,significantly higher than the control group(P<0.05),while the body function score was similar in the two groups(P>0.05).And there was no adverse reactions happened in the two groups.Conclusion Botulinum toxin A combined with delivery autologous platelet rich plasma into skin with microneedle for facial rejuvenation has a good effect and high safety.

3.Co-stimulatory molecule CTLA-4 and the potential application in treating rheumatic diseases

Rheumatic diseases are characterized by immunological dysfunction.T-cell medicated immune response plays a very important role in the development and persistence of rheumatic disease.CTLA-4 is an important co-stimulatory molecules expressed on T cells.It has inhibitory effect on T cell-mediated immune response.Plenty of evidences have demonstrated abnormalities in CTLA-4 alleles and proteins in patients with rheumatic diseases.Therefore,CTLA-4 has become an target in treating rheumatic diseases.CTLA-4Ig has been developed as an fusion protein which is composed of extrA-cellular domain of human CTLA-4 and the Fc fragment of human IgG1.So far,only data about CTLA-4Ig in treating rheumatoid arthritis are available.These data showed that CTLA-4Ig could relieve the symptoms of rheumatoid arthritis and retard the development of the disease.It is safe and effective.In addition,effectiveness was demonstrated even in patients who failed to response to anti-TNF monoclonal antibodies.Therefore,CTLA-4Ig is an encouraging therapy for rheumatic diseases.However,because of very short history,long-term study is needed to fully understand its role in the treatment of rheumatoid arthritis.
4.Research advances in monitoring platelet function in patients with coronary artery disease

Antiplatelet therapy and stent implantation have been the dominant treatment to reduce the mortality of patients with coronary artery disease.Recently,studies have showed that adverse cardiac events still occur in part of patients with coronary artery disease after the antiplatelet treatment with aspirin and/or clopidogrel.Thus,resistance to aspirin and clopidogrel has attracted increasing attention.It will be great benefit to these patients who were identified resistance and made tailoring antiplatelet therapy So far many platelet function tests has been used in clinical to monitor the reaction of the antiplatelet drugs for prevention and treatment of thrombosis in patients with coronary artery disease.These monitoring tests may be chosen based on different antiplatelet drugs including aspirin,clopidogrel and GP Ⅱ b/Ⅲ a antagonist.The results of antiplatelet drug resistance may be different due to different platelet function methods,thus the related clinical adverse events needs further verification.

6.Expression and function of plasminogen activator inhibitor-3 in normal adult skin

Objective To explore the expression of plasminogen activator inhibitor-3 (PAI-3) in normal adult skin. Methods Immunohistochemistry and RT-PCR were used to detect the expressions of PAI-3 and uPA. Results mRNA of PAI-3 and uPA were detected. PAI-3 was mainly expressed in stratum basale, stratum spinosum and stratum granulosum, and its expression enhanced in the well differentiated keratinocytes of the epidermis. uPA was located in basal layer. Conclusion PAI-3 exists in normal adult skin and is related with the differentiation of epidermal keratinocytes.
7.Expressions of GLP-1R and CK17 in papillary thyroid cancer and clinical significance

Objective To investigate the expressions of glucagon-like peptide-1 receptors (GLP-1R) and cytokeratin17 (CK17) in papillary thyroid cancer tissue and clinicopathological significance.Methods The immunohistochemistry and immunoblotting method were employed to detect the expressions of GLP-1R and CK17 in normal thyroid tissues (control group,50 cases) and papillary thyroid cancer tissue (observation group,80 cases).Then the correlation between the expressions of GLP-1R and CK17 with pathological factors of thyroid cancer was analyzed.Results Low expression of GLP-1R was found in 22 cases (44.00 %) of the control group,while GLP-1R was expressed in 77 cases(96.25 %) of the observation group,and the difference between both groups was statistically significant(P<0.01).The expression of CK17 was positive in 8 cases(16.00%) of the control group,while 77 cases(96.25 %) in the observation group were CK17 expression positive,and the difference between both groups was statistically significant(P<0.01).The difference between GLP-1 positive expression and clinicopathologic factors had no statistical significance(P>0.05).The difference between CK17 expression with sex and age had no statistical significance (P>0.05),and was obviously related with the tumor diameter,clinical stages and cervical node metastasis of thyroid neoplasms(P<0.05).Conclusion GLP-1R and CK17 all are highly expressed in papillary thyroid cancer tissue and CK17 expression is obviously related with the tumor diameter,clinical stages and cervical node metastasis of papillary thyroid cancer.
8.Diagnostic of serum tumor markers for colon cancer assessed with logistic regression analysis and PLS-DA model

Objective To investigate the diagnostic value of serum tumors M2 pyruvate kinase (TuM2-PK),carcino-embryonic antigen (CEA),CA199,CA724,CA125 and CA242 in colon cancer.Methods Serum levels of TuM2-PK,CEA,CA199,CA724,CA125 and CA242 in 231 patients with colon cancer,105 patients with colon benign lesion diseases and 166 normal controls were measured by the ELISA and electrochemiluminescence assays.The operation working characteristic curve (ROC) and the logistic regression-ROC were used to evaluate the diagnostic value of tumor markers for colon cancer individually and in combination.The models of logistic regression analysis and partial least-squares discriminant analysis (PLS-DA) were established to diagnose patients with colon cancer based on the optimal panel of serum tumor markers.Resalts Concentrations of serum TuM2-PK,CEA,CA199,CA125 and CA724 in the colon cancer group are higher than those in the colon benign diseases group and the normal controls (P<0.05).The area under the operation working characteristic curve (AUC) of CA199 is the highest 0.79 [95% confidence interval (95%CI),0.75-0.83] at the cutoff value of 69.5 U/L with 64.1% of sensitivity and 89.7% of specificity.The AUC of combined serum tumor markers based on logistic regression analysis is higher than those in individuals,of which serum (CEA+ CA199+TuM2-PK) is the optimal [AUC=0.89,95% confidence interval(95%CI),0.86-0.92].The diagnostic accuracy of logistic analysis and PLS-DA model for colon cancer is 82.7% and 77.5%,and for colon cancer is 93.7% and 95.6%,respectively.Conclusion The combination of serum tumor markers CEA,CA199 and TuM2-PK is more suitable as a diagnostic model for the screening of colon cancer.
9.An Analysis on the Differential Expressing Characters of Metastasis-Relating Genes in the Nasopharyngeal Primary Cancerous Tissues of Untreated NPC Cases with Different Patterns

Objective To investigate the relationship between the differential expressing characters of metastasis-relating genes in the nasopharyngeal primary cancerous tissues of untreated nasopharyngeal carcinoma (NPC) cases and the patterns in the terms of Traditional Chinese Medicine (TCM) manifested by these patients. Methods Forty one untreated NPC cases included were differentiated for their own patterns respectively at first and biopsys were made to take tissue samples from the nasopharyngeal mass of them for histopathological diagnosis. Furthermore, expressing level determination of metastasis-relating genes nm23-H1 and VEGF165 was carried out among these samples to analyze the correlation of expressing activity of these genes with their firstly differentiated patterns respectively. Results The expressing level of nm23-H1 was significantly lower in the biopsied tissue samples of nasopharyngeal primary focus among the untreated NPC cases with a pattern of dual deficiency of Qi and Yin than those with the other two kinds of pattern, while the expressing level of VEGF165 was in a reverse way to that of nm23-H1 in its activity expression among these cases with the same kind of pattern (P
10.Expression of nNOS in the hippocampus in vascular dementia rats

Objective To explore the expression of neuronal nitric oxide synthase (nNOS) in the hippocampus in vascular dementia rats.Methods 60 rats were randomly divided into five groups: control group, model 12 h group,model 1 d group, model 3 d group and model 7 d group.An animal model of vascular dementia (VD) was established through repeated cerebral ischemia-reperfusion in rats with repeated bilateral common carotid arteries occlusion. The number of neurons in CA 1 area of hippocampus in every group was observed using HE staining. The expression of nNOS in the hippocampus in VD rats was detected by immunohistochemical staining and western blot method.Results The number of neurons in CA 1 area of hippocampus decreased significantly in 12 h, 1 d, 3 d and 7 d group. The expression of nNOS in CA 1 area of hippocampus was weak in control group and up-regulated in 12 h group and increased further in 1 d group. However, its expression decreased gradually in 3 d and 7 d group.Conclusion nNOS may be related to the injury of hippocampus at early phase in VD rats. It might be regarded as one of mechanisms to cause VD.
