2.Optimization technique in separation of the total flavonoid from Radix Puerariae with macroporous resin

AIM: To explore the optimal separation of the total flavonoid from Radix Puerariae by selecting appropriate macroporous resins and to study systematically the factors which affect separation. METHODS: Static and dynamic adsoption desorption methods were adopted, and evaluated for separating efficiency by measuring the concentration of total flavonoid in Radix Pueraria with UV spectrophotometer. RESULTS: The SP70 macroporous resin had the best separating efficiency when the flavonoid content in the liquid was 0.5g per 1mL equivalemt to raw material. pH5~6. the volume of drug 60BV (resin bed volume) with the adsorption power 2BV/h. and the volume of 70%(mL?mL -1 ) ethanol as eluant 4BV with desorption power 2BV/h. In result. total flavonoid's purity could reach 80%. CONCLUSION: This method is simple and feasible with good effect of separation, which can meet industrial requirements.
3.Correlation analysis of the severity of EEG and secondary epilepsy in elderly patients with cerebral infarction

Objective To investigate the correlation between EEG severity and secondary epilepsy in elderly patients with cerebral infarction.Methods 128 cases with cerebral infarction were selected as the research subjects. According to whether the patients happened secondary epilepsy,they were divided into the observation group (65 cases) and the control group (63 cases).The difference of EEG severity between the two groups was compared,and its correlation with secondary epilepsy was analyzed.Results There were 18 cases (27.69%)in the observation group graded as moderate differences,which was significantly higher than that in the control group of 12 cases (19.05%), the difference was statistically significant (χ2 =4.43,P =0.04).There were 23 cases (35.38%)in the observation group graded as severe differences,which was significantly higher than that in the control group of 9 cases (13.85%),the difference was statistically significant (χ2 =6.51,P =0.01 ).There were 21 cases(32.31%)of EEG changes of observation group being only diffuse abnormalities forms,which was significantly higher than that of the control group of 11 cases(17.46%),the difference was statistically significant (χ2 =4.18,P =0.04).There were 28 cases(43.08%)of EEG changes of the observation group being both focal abnormalities and diffuse abnor-malities forms,which was significantly higher than that of the control group of 14 cases(22.22%),the difference was statistically significant (χ2 =5.40,P =0.02).Compared open -closed test,hyperventilation provocation test results of the observation group being abnormal or not fit with the control group,the difference was not statistically significant (P >0.05).Conclusion According to the classification of EEG,EEG changes and EEG related tests to determine the severity of EEG.The more severe EEG,the greater the possibility of secondary epilepsy happen in patients with cerebral infarction.

6.Piwi-interacting RNA(piRNA):a Novel Class of Noncoding Small RNA

piRNA(Piwi-interacting RNA) is a novel class of small single strand RNA that were recently isolated from testes of the mammals, associate with PIWI proteins, and are organized into distinct genomic clusters. These RNAs are typically 30 nt long, strikingly different from microRNAs in their length, expression pattern, and genomic organization. piRNA has a role in RNA silencing via the formation of an RNA-induced silencing complex (RISC) with Piwi proteins, these piRNA complexes (piRCs) have been linked to transcriptional gene silencing of retrotransposons and other genetic elements in germ line cells, particularly those in spermatogenesis.Recent researches and progresses of piRNAs are reviewed.
7.Survey of root canal curvature of anterior teeth from Chuang population.

OBJECTIVETo investigate the root canal curvature of permanent anterior teeth from Chuang population.
METHODS245 anterior teeth from Chuang population were collected and examined by X-ray radiography both from labiolingual and mesiodistal directions. For 218 type I anterior teeth, degree of root canal curvature, radius of curvature and length of the curved part of root canal were measured by a special electronic vernier caliper according to Schneider's and Schäfer's method and the data obtained were analyzed.
RESULTSRoot canals of anterior teeth from Chuang population were mainly of type I. The number of type II, III, IV were about 13 in mandibular central and 12 in mandibular lateral incisors. The incidence of curvature in maxillary central incisors, lateral incisors, canines and mandibular central incisors, lateral incisors, canines were 40%, 80%, 77%, 65%, 66%, 73% in mesiodistal directions, 62%, 69%, 70%, 62%, 41%, 61% in labiolingual directions respectively. The most curvature was moderate and happened in apical third. The heaviest curvature occurred in maxillary canines in mesiodistal direction and mandibular canines in labiolingual direction. The shortest radius and length of curvature occurred in maxillary lateral incisors.
CONCLUSIONRoot canal curvature of anterior tooth in Guangxi Chuang population is complex. The incidence of type II, III, IV is high in mandibular incisors.

8.Effect of L-arginine on expression of PKC mRNA in pulmonary injury induced by ischemia-reperfusion in rabbits

Objective To investigate the effect of L-arginine on expression of protein kinase C(PKC)mRNA during pulmonary ischemia and reperfusion injury(PIRI)in the rabbits.Methods Single lung ischemia and reperfusion animal model was used in vivo.The rabbits were randomly divided into three groups(n=9,in each),sham operated group (Sham),PIR group(I-R)and PIR+L-arginine group(L-Arg).Changes of several rariables including malondialdehyde (MDA),superoxide dismutase(SOD),malandialde hyde(MDA),nitril oxide(NO),wet to dry ratio of lung tissue weight(W/D)and index of quantitative assessment(IQA)of histolngic lung injury were recorded at 60 minutes after reperfusion in lung tissue.Meanwhile the location and expression of PKC mRNA were observed.Lung tissue was prepared for light microscopic and electron microscopic observation at 60 minutes after reperfusion.Results In comparison with group I-R,PKC mRNA strongly expressed in intima and extima of small pulmonary artery as well as thin-waU vessels (mostly small pulmonary veins).The average optical density values of PKC-?,?and?mRNA in small pulmonary veins in L-Arg group had significance(all P<0.01);SOD increased while MDA,W/D and IQA decreased at 60 minutes after reperfusion in lung tissue(P<0.01 and P<0.05).A morphologically abnormal changes of the lung tissue,were lessen markedly in L-Arg group.Conclusion L-arginine possess notably protective effects on PIRI in rabbits by activating PKC-?,?and?mRNA expression in lung tissue,raising NO level,dropping oxygen free radical level and decreasing lipid peroxidation.
9.Cytopathological scrape examination of superficial lesions of skin and mucosal membrane--a cytological and histological comparative study of 148 cases.

OBJECTIVETo investigate the reliability, practicability and limitations of cytopathological scrape method in the diagnosis of superficial lesions of skin and mucosal membrane.
METHODSThe quantity of cells harvested by different collecting methods were compared in 18 cases of malignant tumors of the skin and mucosal membrane. The scrape method was performed in 456 cases, of which 148 cases had corresponding histological specimens.
RESULTSGlass slide scraping provides the most satisfactory cell numbers from the lesions of skin and mucosal membrane. The specificity for diagnosing benign lesion in 37 patients was 100% and that for diagnosing malignant tumors in 111 patients was 92.8% (103). The overall diagnostic accuracy was 94.6% (140/148). The false negative rate, false positive rate and the overall misdiagnostic rates were 7.2% (8/111) , 0, and 5.4% (8/148) respectively.
CONCLUSIONSFor cytopathological examination of superficial skin and mucosal membrane lesions, the glass slide scraping methodprovides broad applications with high diagnostic accuracy and advantages of simplicity, safety and efficiency. Certain limitations do exist, but can be resolved by histological examination of the lesion.

10.Effect and mechanism of total flavonoids of bugloss on rats with myocardial ischemia and reperfusion injury.

This study is to investigate the effect of total flavonoids of Uygur medicine bugloss (BTF) on rats with myocardial ischemia/reperfusion injury, and to explore the mechanisms by which it acts. Left anterior descending (LAD) coronary artery in rats was occluded for 30 min followed by 4 h reperfusion. Meanwhile, BTF dissolved in saline was administered intraperitoneally at dosage of 10, 30 and 50 mg x kg(-1). Electrocardiograph, infarction index, serum myocardial enzymes and heart function were determined to evaluate the effect of BTF. Some other observations were carried out to explore whether inhibiting inflammation and apoptosis is involved in the mechanisms underlying BTF. Our results showed that in ischemia/reperfusion injured rats BTF could dose-dependently reduce myocardial infarction index and myocardial enzyme leakage, and enhance heart function, indicating that it possesses significant cardio protection. ELISA analysis showed that BTF could decrease the content of myocardial inflammatory cytokines such as IL-1beta, IL-6 and TNF-alpha. Western-blotting confirmed that BTF could increase the expression of anti-apoptotic protein Bcl-2 and reduce the expression of proapoptosis protein Bax. Further more, the phosphorylation level of PI3K and Akt was upregulated by BTF treatment. BTF can protect rat against myocardial ischemia/reperfusion injury. Anti-inflammation and inhibition of apoptosis through upregulating PI3K/Akt signal pathway may contribute to the protective effect of BTF.
