Objective To detect the changes of serum complement 3(C_3) and C_4 in neonates with hypoxic-ischemic encephalopathy(HIE) and its effects on non-specific immunological function.Methods Sixty-seven neonates with HIE and 30 normal full-term infants were recruited in the study.According to characteristic of immunological function and severity of HIE,the neonates were divided into 1-3 day group(n=32)and 4-6 day group(n=35).In each group,it was divided into mild group and moderate-severe group.The complement in serum was detected by radial immunodiffusion assay with Behring Nephelometer 100 Analyzer.Results In 1-3 day group,levels of C_3,C_4 in serum were lower than those in control group(P

3.Sixteen Cases of Primary Small Vessel Vasculitis with Involvement of Lungs

Objective To understand the clinical characteristics of primary small vessel vasculitis with involvement of lungs in children and analyze the causes of misdiagnosis.Methods Clinical data from 16 patients with primary small vessel vasculitis were collected and their pathological,antineutrophil cytoplasmic antibody(ANCA),pulmonary lesion manifestations,image and misdiagnosis were further analyzed.Results Among 16 cases,9 cases were microscopic polyarateritis(MPA) and 4 cases were Wegeners granulomatosis(WG) and 3 cases were Churg-Strause syndrome(CSS).The ages of onset were from 3 to 14 years old with average age of 7.5 years old.Seventy-five percent(12/16) were ANCA positive.The patients with lung involvement had cough,chest pain,dyspnea,hemoptasis and asthma.Findings on lung X-ray films were variable.Some showed multiple patch shadows,others showed single or multiple masses or nodular shadows with or without cavity formation.After the treatments with corticosteroid and cyclophosphamide,10 cases turned better;including 8 cases with normal on lung X-ray films.Three cases given up;3 cases died.Conclusion It is very difficult to make diagnosis of primary small vessel vasculitis with involvement of lungs and should be paid more attentions to this issue.
4.Determination of Tetrahydropalmatine in Anwei Tablets by Dual Wavelength TLC-scanning

Objective: To establish a method for determination of tetrahydropalmatine in Anwei tablets. Methods: tetrahydropalmatine in Anwei Tablets was extracted by chloroform and determined by dual wavelength TLC scanning (? s=275nm,? R=340nm).Results: The average recovery was 95%.17%, RSD was 2.50% (n=5). The contents of tetrahydropalmatine in three batches were 7.45,7.17 and 7.90?g/tablet, respectively.Conclusions: The method is stable and feasible.
5.Different Effects of Triptergium.reglii On T and B cell Function

The suppressive effects of T. reglii on mouse cellular and humoral immune responses were studied. Lymphocyte proliferation effect induced with ConA,and PFC and OHS reactions with TD Ag(TNP-SRBC)were effectively suppressed by the drug. But the similar effects on LPS and TI Ag (TNP-SRBC) have not been found. The results shown that target cells of T. reglii effects may be T lymphocytes and the drug may not have a direct effect on B cells. The drug also shown a suppressive effect on IL-2 production by mouse spleen cells. The phenomenon suggests that the suppressive effects of T. reglii on humoral immune response may be indirectly mediated through suppressing helper T cells.
6.Photoelastic Stress Analysis of Two Kinds of Rigid Fixed Bridge Design after Hemisection of Mandibular First Molar

ObjectiveTo observe the effects of the two kinds of rigid fixed bridge design on the stress of the alveolar bone of mandibular first molar after hemisection.MethodsThe normal mandibular first molars were selected as control group. The two-abutment teeth and three-abutment teeth rigidly fixed bridge designs were selected as trial group. Stress distributions in the remaining sections of the alveolar bone models were evaluated with photoelastic stress analysis method.ResultsThe stress values of two kinds of rigid fixed bridge design were higher than the control group but still below the twice. There was no significantly difference between two-abutment teeth rigidly fixed bridge design and three-abutment teeth rigidly fixed bridge design (P>0.05).ConclusionTwo-abutment teeth rigidly fixed bridge of mandibular first molar should be chosen after hemisection.
7.Development of the Photoelastic Model for the Tooth and Periodontal Tissue of Mandibular First Molar after Hemisection

ObjectiveTo discuss the design and fabrication of the photoelastic model of mandibular first molar after hemisection.MethodsAccording to principles of similarity, different materials were used to manufacture the photoelastic model of mandibular first molar with hemisection, which included teeth, alveolar bone and pericementum.ResultsThe photoelastic modle of mandibular first molar with hemisection was manufactured, which was as similar as noumenon in appearance, parameter and boundary condition.ConclusionThe photoelastic model can be applied in qualitative analysis of the stress distribution in the periodontal supporting tissues of mandibular first molar with hemisection.
8.The relationship of resistance mechanism of two strains of Enterobacteriaceae resistant to carbapenems isolated from a single specimen

Objective To investigate the relationship of resistance mechanisms of a Klebsiella pneumoniae strain and a Morganella morganii strain resistance to carbapenems isolated from a single specimen. Methods Sensibility of antimicrobial agents was detected by agar dilution method. Specific PCR and DNA sequence analysis were performed to detect resistance genes. Plasmid feature was detected by plasmid conjugation and electrophoresis analysis. Genetic environment around blaKPC was analyzed with sequencing. The changes of outer membrane permeability were analyzed with electrophoresis of outer membrane proteins. Results blaKPC-2 was detected in 2 original isolates strains and their transconjugants. Carbapenem-resistance was successfully transfered by conjugation experiments. blaKPC-2 was located on dissimilar plasmids, but genetic environment around blaKPC-2 was the same sequence. The Morganella morganii isolate showed a loss of 38 ×103 OMPs and an additional 36 ×103 OMPs appearance, while the Klebsiella pneumoniae isolate showed a loss of OMPK36. Conclusion blaKPC-2 was detected in 2 isolates. This gene encoded by two plasmids with different sizes was located on the same composite transposon. The lack of outer membrane proteins could also play an important role causing isolates to exhibite resistance to carbapenems.
9.Effect of integrinβ1 on multidrug resistance in gastric cancer SGC-7901 cells

AIM:To study the effect of integrin β1 on multidrug resistance in gastric cancer and its possible mechanisms .METHODS:The expression of integrin β1 at mRNA and protein levels in the SGC-7901 cells and SGC-7901/DDP cells was determined by qPCR and Western blot .The expression of integrin β1 in the SGC-7901/DDP cells was silenced by antisense oligodeoxynucleotide .The cell viability was detected by the CCK-8 assay, the cell apoptosis were ana-lyzed by flow cytometry, and the protein levels of integrin β1, Bcl-2/Bax, cleaved caspase-3/caspase-3, cytochrome C ( Cyt-C) and p-AKT/AKT were determined by Western blot .RESULTS:The expression of integrin β1 at both mRNA and protein levels was significantly upregulated in SGC-7901/DDP cells.The expression of integrin β1 was increased in SGC-7901 cells treated with chemotherapeutic agents such as cisplatin , paclitaxel and 5-fluorouracil .Knockdown of integrin β1 induced apoptosis of SGC-7901/DDP cells with an increased sensitivity to the chemotherapeutic agents .Meanwhile , knock-down of integrin β1 downregulated the protein levels of Bcl-2/Bax, p-AKTSer473 and p-AKTThr308 , while promoted the release of Cyt-C and upregulated the protein level of cleaved caspase-3.CONCLUSION:Knockdown of integrin β1 increases the sensitivity of SGC-7901/DDP cells to the chemotherapeutic agents , and promotes the cell apoptosis via mitochondrial apop-tosis pathway .The mechanism may be related to the attenuation of AKT pathway by inhibiting phosphorylations of AKT at Ser473 and Thr308.
10.Determination of the related substances of benzyl hydroxybenzoate by RP-HPLC

An RP-HPLC method was established to separate the related substances of benzyl hydroxybenzoate.The separation was carried out on a Agilent ZORBAX Eclipse Plus Phenyl-Hexyl column.The mobile phase was methonal-0.1% glacial acetic acid,using linear gradient elution,and the detection wavelength was 254 nm.There was a good linear relationship between 0.051-101.88 μg/mL (r =1.00) and 0.050-99.48 μg/mL(r =0.999 8) for benzyl hydroxybenzoate and p-hydroxybenzoic acid,repectively.The average recovery of p-hydroxybenzoic acid was 100.3% and the RSD was 0.95%.The LOQ of p-hydroxybenzoic acid was 0.24 ng.The detected impurities were also identified by UPLC-Q-TOF.The established method is accurate and reproducible,and could be used for the quality control of benzyl hydroxybenzoate.
