Objective To investigate the effects of lipid microspheres prostaglandin E1(Lipo PGE1) on subarachnoid hemorrhage(SAH).Methods Ninty-three patients with SAH were randomly divided into control group(treated with nomal drugs) and Lipo PGE1 treatment group(treated with nomal drugs and Lipo PGE1).Changes in neuroimaging,biochemical indexes and incidence of cerebral vasospasm were measured. Results A lower incidence rate of cerebral vasospasm was observed in Lipo PGE1 treatment group(P0.05).The levels of vWF and GMP-140 were significant lower in the Lipo PGE1 treatment group than those in the control group after treatment for 3 and 7 d(P

3.Effects of bFGF on cognitive ability of rat with diffuse axonal injury

Objective To study the alterations in neurobehavior of rats with diffuse axonal injury(DAI) and to evaluate the potential role of basic fibroblast growth factor(bFGF). Methods A weight-drop device was employed to build the DAI model.An injection of bFGF subdurally and subcortically was given to the bFGF therapeutic group(n=60).Besides,normal control(n=20) and injured control group(n=60) were established.The elevated walkway test,prehensile traction test,sensorimotor integration test and the Morris' water maze test were adopted to examine the motor and memory abilities.After the implantation of skull electrodes,P3-like potential was explored in rats before and after injury. Results After DAI,the scales of the elevated walkway test,prehensile traction test,sensorimotor integration test and the Morris' water maze test were decreased,and the rats in the bFGF therapeutic group presented a better behaviour in the early stage.The latency of P3-like potential prolonged significantly in rats with DAI,with the P3-like potential in the injured control group longer than that in the bFGF therapeutic group(P
4.Effects of bFGF on alterations in pathology and immunohistochemistry in rats with diffuse axonal injury

Objective To study the alterations in pathology and immunohistochemistry in rats with diffuse axonal injury(DAI) and the effects from basic fibroblast growth factor(bFGF). Methods A weight-drop device was employed to produce DAI in rats.In the treatment group(n=60),bFGF was injected subdurally and subcortically.Besides,normal control group(n=20) and injury-control group(n=60) were also established.The pathological changes were observed by light microscopy and electromicroscopy,and the expression of brain-derived neurotrophic factor(BDNF),growth associated protein-43(GAP-43) and glial fibrillary acidic protein(GFAP) were also examined by immunohistochemistry. Results Typical pathological changes were observed in the basal portion of pons,corpus callosum and white matter of cerebral hemisphere in the rats with DAI.And an upregulation of GFAP,GAP-43 and BDNF was also found.In the treatment group,better outcomes of pathological changes were observed.bFGF increased the expression of BDNF and GAP-43,while inhibited the immunoreactivity of GFAP. Conclusion Topical application of bFGF can improve brain tissue regeneration and speed function recovery in rats with DAI,though its long-term effect warrants further study.
7.Analysis of size,aspect ratio and configuration in ruptured aneurysms

Objective To evaluate the size,aspect ratio and configuration in ruptured aneurysms. Methods Three-dimensional CT angiogram of 61 confirmed ruptured saccular aneurysms were analyzed.The maximum diameter and aspect ratio were measured,and the configuration of the aneurysms were observed. Results The average maximum diameter was 8.54 mm,and 78.8% of the aneurysms were less than 10 mm in maximum diameter.The average aspect ratio was 2.15,and more than 65% of the aneurysms had an aspect ratio over 1.6.45.9% of the aneurysms harbored a bleb or were multi-lobular in shape. Conclusion Aneurysms less than 10 mm in maximum diameter are also easy to rupture.More than 60% of the ruptured aneurysms had an aspect ratio over 1.6.Bleb and multi-lobular change are common in ruptured aneurysms.
8.Establishment and application of a high-throughput screening assay for premature activation of HIV-1 precursors.

Strict regulation of HIV-1 PR function is critical for efficient production of mature viral particles. During viral protein expression and viral assembly, HIV-1 PR located within Gag-Pol precursor must be inactive to prevent premature cytoplasmic processing of the viral Gag and Gag-Pol precursors. Premature activation of HIV-1 precursors leads to major defects in viral assembly and production of viral particles. A cell-level premature activation of HIV-1 precursors assay using bioluminescence resonance energy transfer (BRET) was established. Three thousand compounds were screened to evaluate this assay. The results showed that the assay is sensitive, specific and stable (Z' factor is 0.905).
9.Establishment and application of a screening anti-HIV-1 drug model targeted nuclear trafficking of virus RNA.

The HIV-1 Rev protein facilitates nuclear export of unspliced and singly spliced viral transcripts containing RRE RNA through the CRM1 export pathway. Inhibition of Rev-mediated RNA nuclear export can arrest HIV-1 transcriptional process, which clearly, reveals a target for anti-HIV drug development. In this work, a cell-based assay has been established for screening anti-HIV compounds targeting the Rev-mediated RNA nuclear export. This assay utilized a codon-optimized green fluorescent protein (GFP) as reporter gene, which expression is in a Rev-dependent manner. Any compound that inhibits the Rev-mediated RNA nuclear export is identified by reducing emission of GFP. The Z' score of this model is 0.8220. Three thousands compounds were screened and the positive rate was 9.3% with a cutoff at 50% inhibition. IMB7C7, one of the positive compounds, efficiently inhibits viral production from HIV-1 infected cells.
10.Applied value of 320-slice CT perfusion examination in ischemic-type biliary lesions after liver transplantation

Objective Using CT perfusion (CTP) technique,to investigate the graft perfusion changes in patients with hepatic artery stenosis (HAS) with or without ischemic-type biliary lesions (ITBL) after orthotopic liver transplantation (OLT).Methods Thirteen recipients with HAS received CTP scan of the liver,including 8 with ITBL and 5 without ITBL.For all patients,the diagnosis of HAS was made by CTA,and the diagnosis of ITBL by percutaneous transhepatic cholangiography.CT perfusion indices were obtained,including hepatic artery perfusion (HAP),portal vein perfusion (PVP),total liver perfusion (TLP) and hepatic perfusion index (HPI).Results Of the 13 patients with HAS,mean HAP in patients with and without ITBL was 59.8 and 35.1 ml·min-1 ·100 ml-1 (P =0.021,two-tailed paired Student t test) ; mean PVP was 125.4 and 166.2 ml·min-1·100 m1-1 (P =0.016) ; mean TLP was 185.2 and 201.3 ml· min-1 · 100 ml-1 (P =0.306) ; and mean HPI was 33.6 and 18.2 (P =0.005),respectively.Conclusion Using CTP technique,liver perfusion changes were reflected by measuring CTP indices noninvasively.Compared to those without ITBL in this study,HAP and HPI in patients with ITBL were higher and PVP was lower,which may be contributed to biliary inflammation.
