2.Interfering Remedial Effect of Adenosine on Rats with Acute Renal Failure

Objective To investigate the effect of adenosine on rats with acute renal failure(ARF) resulting from ischemia-reperfusion(IR).Methods The model of renal IR injury rats was induced by nipping bilateral renal pedicels of rats with non-traumatic forceps for 40 minutes.The first adenosine group was given adenosine 10 mg/kg intraabdominally at 2 hours before IR.The second adenosine group was given the same adenosine at 40 minutes after IR.Serum creatinine(Scr) levels,the proportion of apoptosis of renal tubular cells(RTCs) and the pathological changes of renal tissue were observed separately in every group at 2,6,12 hours after IR.Results 1.At 12 hours after IR,in IR group Scr levels and apoptosis of RTCs count were(198.56?13.35) ?mol/L and 26.50?2.07,which were increased significantly compared with sham group.In the first and second adenosine groups,they were(113.85?(12.90) ?mol/L),14.67?3.44 and(218.79?37.65) ?mol/L,20.83?5.27,respectively,and all of them were higher than those in sham group(all P0.05).Conclusions Adenosine which is given to rats with IR before IR may be an effective pharmacological agent for prevention and treatment of renal IR injury.The mechanism may be relevant to saving energy and reducing cellular apoptosis.
3.Inhibitive Effect of Interleukin-4,-10 on Cell Apoptosis in Rats with Renal Ischemic-Reperfusion Injury

Objective To study the inhibitive effect of interleukin-4(IL-4) and (or) IL-10 on cell apoptosis of renal ischemic-reperfusion injury(IRI) in rats and its mechanism.Methods Thirty female Sprague-Dawley rats were divided into five groups(n=6): sham-operated group,operation+9 g/L NaCl control (NaCl group),operation+IL-4(IL-4 group),operation+IL-10(IL-10 group),operation+ IL-4+IL-10 (IL-4+IL-10 group). The animal model of IRI was set up by clamping bilateral renal pedicles for 45 min.When the clamp was loosed, relevant agents was injected in every group. Apoptosis of renal epithelial cells (TUNEL method) was obserued.Results IL-4 could not reduce level of apoptosis of renal epithelial cells.To some extent,IL-10 could reduce level of renal epithelial cells. The combination of IL-4 and IL-10 was more effective in inhibiting apoptosis of renal epithelial cells.Conclusion The combination of IL-4 and IL-10 is effective in inhibiting apoptosis of renal epithelial cells, indicating that it has a relationship in preventing renal histological and functional damage.
4.Research on the Curative Effects of Integrated Intervention on Children with Learning Disability

Objective:To probe the curative effects and functional mechanism of integrated interference on children with learning disability.Methods:31children with learning disability have been treated for a year with a combination of methods such as cognitive training,behavior interference and training of all thesenses.During the different periods of treatment,results were recorded and measured. Resrlts:After one year’s integrated treatment of interference,the resultsacquired through Raven Standard Reasoning Test and Beery’s integrated-skill test of children’s vision and movements have greatly improved; errors made in thevisional attention test were dropped dramatically; and the scope of hearing and memory were obviously improved. The excessive exponent measured by Coroner’sForm has decreased; the results obtained by means of the nine factors provided by CBCL to examine psychological behavior have dropped respectively during thedifferent periods of treatment; and 87% of the children have made progress in their studies.Conclusion:The integrated interference by means of cognitive training,behavioral interference and the training of all the senses have produced a good and persistent effect on children with learning disability. The methods of integrated interference, having take into consideration the characteristics of children’s cognition, psychology and emotions, have worked together to bring about the results.
5.Establishment of Microbial Limit Test for Dahuang Zhechong Pill

objective Microbial limit test(MLT)for Dahuang Zhechong pill was established and verified to supply scientific foundation for the establishment of MLT for antibacterial and anti-inflammatory Chinese patent medicine,and to supply principle guidance for the verification of the method issued in Chinese pharmacopoeia(2005 edition);meanwhile the antibiogram of Dahuang Zhechong pill was probed,which can provide evidence for its clinical administration.Methods With centrifugal sedimentation method and culture medium diluted method,the recovery test was done on each tested bacterial strain to validate the accuracy and feasibility of MLT.Results Dahuang Zhechong pill had a stronger inhibitory effect on staphylococcus aureus and bacillus subtilis,had a weak inhibitory effect on escherichia coli,and had no effect on candida albicans and aspergillus niger,indicating that the antimicrobial spectrum of Dahuang Zhechong pill is Gram-positive bacteria.Conclusion Centrifugal sedimentation method combined with culture medium diluted method can eliminate the anti-bacterial effect of Dahuang Zhechong pill during the experiment,thus the microorganisms in the polluted pill can be detected successfully.

Objective:To investigate the effects of microgroove surface morphology on the adhesion and proliferation of the human gin-gival fibroblasts(HGFs).Methods:Microgroove surfaces of titanium were fabricated by photolithography with parallel grooves of 15,30 or 60 μm in width and 5 μm or 10 μm in depth.The groups of the samples were denoted as T15 /5,T15 /10,T30 /5,T30 /10,T60 /5 and T60 /10.Smooth titanium surface(T0)was used as the control.The surface topography was observed by enviroment SEM(ES-EM).HGFs were cultured on the topographically modified surfaces.Morphology was observed by SEM.Cell proliferation was examined by CCK-8 kit.Results:HGFs on the microgroove surfaces had “contact guidance”parallel to the microgrooves,whereas the cells on T0 were oriented randomly.Cell proliferation was promoted and kept for longer period on T60 /10 surface.Conclusion:Surfaces of mi-crogrooves with increasing groove width and depth may achieve “contact guidance”for HGFs and promote cell proliferation.
8.The effects of microgroove surface form of titanium on the adhesion and cell cycle progression of human gingival fibroblasts

Objective:To investigate the effect of microgroove surfaces of titanium on the adhesion and cell cycle progression of human gingival fibroblasts(HGFs).Methods:Microgroove titanium surfaces were fabricated on silicon plate by photolithography with parallel grooves:1 5,30 and 60 μm in width and 5 μm and 1 0 μm in depth,the groups were denoted as T1 5 /5,T1 5 /1 0,T30 /5,T30 /1 0, T60 /5 and T60 /1 0,respectively.Smooth titanium surfaces (T0)were used as the controls.Surface topography were observed by ES-EM.HGFs were cultured on the microgroove surfaces.Morphology of “contact guidance”was observed by immunofluorescence tech-nique.Cell cycle distribution was analyzed by Flow cytometry.Results:HGFs on the microgroove surfaces had “contact guidance”par-allel to the microgrooves,whereas the cells on T0 were oriented randomly.T60 /1 0 group had the highest percentage of S phase cells, followed by T30 group and T1 5 group,but still higher than that in the control group.In groups with higher groove width (T60 group and T30 group),the increase of groove depth benefited the increase of S phase percentage,while in T1 5 group,the increase of groove width decreased the S phase percentage.Conclusion:Surfaces of microgrooves with different dimensions achieved “contact guidance”for the cultured HGFs.The surfaces with increasing groove width and depth benefit the cell cycle progression.
10.Therapeutic effect assessment of integrated intervention for learning difficulties in children

BACKGROUND:Children with learning difficulty often present with cognitive impairments and imbalanced development of intelligence accompanied by emotional and behavioral problems.OBJECTIVE:To evaluate the therapeutic effects of integrated interventions in multiple aspects for children with learning difficulties.DESIGN:A case-control study of children with learning difficulties.SETTING:Department of Pediatric Internal Medicine, Union Hospital Affiliated to Fujian Medical University.PARTICIPANTS:This study involved school-age children of 6 to 12years old with learning difficulties who came to the department specialized in pediatric psychology for consultation from July 1998 to July 2001. The diagnosis of learning difficulties conformed to the diagnostic criteria for special learning disability in ICD-10 with IQ > 70 as an essential condition. Children with emotional disorders, mental deficiency, hyperactivity,visual and auditory dysfunctions and organic cerebral diseases were excluded. The diagnosis was further verified by a professor specialized in neuropsychology. The children voluntarily participated in the cognitive training, behavioral intervention and comprehensive trainings of senses and those who fulfilled the one-year treatment were included in this study. The ratio of therapists to the children was 1:2, and each session of treatment lasted for 90-120 minutes given 2-3 times a week, and a treatment course consisted of 24 sessions. The children were supposed to complete 4 consecutive treatment courses. Totally 31 children completed the treatment courses, including 24 boys and 7 girls with the mean age of(7.7±1.2)years who had an average IQ of 87.5±8.6.grated interventions for one year consisting of cognitive training, behavioral intervention and comprehensive training of the senses. Raven's Deductive Reasoning test and Beery Visual-Motor Integration Test were use to examine the deductive reasoning ability and visual-motor integration of the children, and visual attention test and auditory attention test, as well as sequential and reverse-sequence memory tests were employed to test their attentions. Achenbach children behavioral scale was applied to evaluate the children's behavioral problems, and Conners' questionnaire for children's behavioral problem was used to measure their attention-deficit/hyperactivity disorder (ADHD) index.MAIN OUTCOME MEASURES:Changes in the deductive reasoning ability, capability of visual-motor integration, attention and psychological behaviors after one year of intervention.RESULTS:All the 31 patients children entered the result analysis with no loss. After one year of integrated intervention, the children had significantly improved scores for Raven's Deductive Reasoning Test (87.51 ±8.64 vs 90.03±8.70, t=-9.610, P < 0.001) and Beery Visual-Motor Integration Test (6.75±1.74 vs 8.27±1.94, t=-9.002, P < 0.001). Obvious improvement was also seen in the visual attention test (15.90±20.11 vs 2.74±3.67), scope of auditory attention and memory (7.70±1.60 vs 11.23±1.23for sequential memory and 3.52±0.93 vs 6.97±0.16 for reversed-sequence memory, P < 0.01). The total scores for ADHD assessment declined to different extents at 3 and 6 months during the treatment and upon the completion of the treatment (22.65±3.53 vs 18.87±4.14 at month 3, 5.65±3.92at month 6 and 12.65±3.36 at one year, t=7.35-15.36, P < 0.01). The scores for 9 psychobehavioral items in Achenbach children behavioral scale obviously declined were also reduced significantly during the therapy,and 87% (26/31) of the children had obviously improved academic performance.CONCLUSION:Interventions integrating cognitive training, behavioral intervention and comprehensive training of the senses may produce good and persistent effect on the cognitive and emotional symptoms of children with learning difficulties, resulting in obviously improved visual, auditory and brain functions.
