1.Study on haptoglobin polymorphism and plasma concentration in patients with polycystic ovarian syndrome

Objective·To investigate the correlation between the serum haptoglobin (Hp) level and the metabolic parameters in patients with polycystic ovarian syndrome (PCOS), and explore the distribution of Hp genotype and allele frequency in PCOS patients and their correlations with PCOS. Methods·A total of 106 patients with PCOS and 86 cases of patients with non-hyperandrogenic and regular menstruation (control group) were enrolled. Serum Hp level was measured by enzyme-linked immunosorbent assay (ELISA); Hp genotype and allele frequency were determined by polymerase chain reaction (PCR), respectively. Results·The serum Hp level in PCOS group was significant higher than that in the control group (P=0.000). Correlation analysis reveals that serum Hp level was positively related to body mass index (BMI) (P=0.000) and homeostatic model assessment insulin resistance (HOMA-IR) (P=0.000). Compared with control group, proportion of Hp2-2 genotype (Hp2/Hp2) increased significantly in PCOS group (P=0.006), and the frequency of Hp2 allele was also higher (P=0.002). Conclusion·Serum Hp level was positively related to BMI and HOMA-IR, which maybe associated with the occurrence and development of obesity and insulin resistance. Patients with Hp2-2 genotype might have genetic predisposition to PCOS, and high frequency Hp2 allele was found in them.
3.Clinical effect of gonadotropin releasing hormone-analogue and marvelon in the treatment of ovarian endometriosis after laparoscopy

Objective To study the clinical effect of gonadotropin releasing hormone-analogue (GnRH-a) and marvelon in the treatment of ovarian endometriesis after laparoscopy.Method The postoperative women with ovarian endometriosis were divided into three groups:19 treated with GnRH-a and marvelon(greup A),20 treated with GnRH-a(group B),20 without endocrinal therapy(group C).The symptoms scores,the recurrence and side effects were compared.ResultsThree years after laparescopy,the recurrent rate and symptoms scores in group C was the highest (P<0.05),the recurrent cyst size [(12 + 1)mm] and symptoms scores [(1.2 + 0.9)scores] in group A was the lowest among three groups.Conclusions Marvelon and GnRH-a can be used in the treatment of ovarian endometriesis after laparoseopy.The effect of marvelon combined with GnRH-a is prior to GnRH-a.
4.Epidermal growth factor improves in vitro maturation of immature mouse oocytes

Objective To investigate the significance of EGF in the in vitro maturation, fertilization, and pre-implantation development of the immature oocytes. Methods The immature oocytes were isolated from the ovaries of the rats. The germinal vesicle (GV) oocytes were cultured in the EGF solutions at concentrations of 0(control), 0.5, 1, 2.5, 5 and 10 ng/ml Ham’s F10 culture media. The status of germinal vesicle break-down(GVBD) and the extrusion of the first polar body (PB1) were observed. The achieved MⅡoocytes were then inseminated in vitro; and the pronuclei (PN) and the pre-implantaon development were observed. The gradeⅠ-Ⅱ4-cell embryos were selected and implanted into the fallopian tube of the rats with the 1 day pseudogestation. Follow-up observations were made till the delivery. 5.Effects of 1,25-dihydroxyvitamin D_3 on regulatory T cells and airway inflammation in asthmatic rats

Objective To investigate the effects of 1,25-dihydroxyvitamin D3[1,25(OH)2D3] on regulatory T cells(CD4+CD25+ Foxp3+ T cell)and airway inflammation in asthmatic rats. Methods Asthmatic models were established in 28 rats with ovalbumin as sensitinogen,and four groups were randomly divided(n=7).1,25(OH)2D3 was orally administered 1 h before challenge in treatment group 1,dexamethasone was subcutaneously injected 1 h before challenge in treatment group 2,both 1,25(OH)2D3 and dexamethasone were given 1 h before challenge in treatment group 3,while no medicine was administered 1 h before challenge in group 4(asthmatic control group).The levels of IL-4 and IL-10 in bronchoalveolar lavage fluid(BALF) were detected,the total cell numbers were obtained and the eosinophil count was performed.The ratios of Treg to CD4+ T cells in peripheral blood and spleen mononuclear cells were measured with flow cytometry,and the mRNA expression of IL-10 and Foxp3 in lung tissues was detected.Wistar rats without asthma were served as normal controls(n=7). Results There were significant differences between asthmatic control group and normal control group in each parameter.Compared with asthmatic control group,the total cell number and eosinophil count decreased,the level of IL-4 decreased and the level of IL-10 increased in BALF in treatment groups.The ratios of Treg to CD4+ T cells in peripheral blood and spleen mononuclear cells significantly increased,and the mRNA expression of IL-10 and Foxp3 in lung tissues increased in treatment groups compared with asthmatic control group.There was no significant difference between treatment groups and normal control group in each parameter. Conclusion Administration of 1,25(OH)2D3 increases the proportion of Treg and expression of Foxp3 mRNA in peripheral blood and spleen in asthmatic rats.1,25(OH)2D3 has a regulatory effect on Treg,and may serve as a potential therapeutic approach against asthma.
6.Analysis on macular choroidal thickness of patients with myopic choroidal neovascularization

? AlM: To examine choroidal thickness and its relationship with axial length and spherical equivalent by spectral domain optical coherence tomography ( SD -OCT ) in patients with myopic choroidal neovascularization ( mCNV) .
?METHODS: We prospectively recruited 30 cases of mCNV in Beijing Anzhen Hospital from October 2010 to December 2014. SD-OCT was used to measure choroidal thickness subfoveally and at 1. 5mm superiorly, inferiorly, nasally and temporally ( SFCT, SCT1. 5mm , lCT1. 5mm , NCT1. 5mm , TCT1. 5mm ) . Paired t- test was conducted to assess the difference of spherical equivalent, axial length and choroidal thickness between mCNV group and controls. Logistic regression analysis was performed to evaluate the correlation between spherical equivalent, axial length, choroidal thickness and mCNV.
?RESULTS: The difference of spherical equivalent and axial length between mCNV group and controls was not significant (P>0. 05). lnferior was decreased in mCNV group compared with controls (140. 85±33. 46μm vs 168. 95±45. 36μm). The difference was significant (P=0. 008). Logistic regression demonstrated that decreased choroidal thickness ( lCT1. 5mm ) at inferior location was associated with mCNV (OR=2. 12;95%CI 1. 35~3. 28;P=0. 02).
?CONCLUSlON: The decrease of lCT1. 5MM may be one of the risk factors of mCNV.

9.Nosocomial Infection in Neonatal Intensive Care Unit in Two Years

OBJECTIVE To perform a retrospective analysis to determine the prevalence of hospital infection and associated risk factors and offer strategies in our neonatal intensive care unit(NICU).METHODS The study enrolled 1082 neonatal patients hospitalized between Jan 1,2006 to Dec 31,2007.RESULTS There were 58 nosocomial infections(NI) in 1082 neonatal patients,and the infection rate was 5.36%.The most common infection was superficial(62.07%).The mortality of NI was 6.90%.The lower gestational age and birth weight,were risk factors of NI.Logistic regression analysis indicated that mechanical ventilation and birth weight ≤1500g were independent risk ractors for NI(P

Objective To prepare nanobubbles and analysis its application for enhanced ultrasound imaging of small-cell lung cancer (SCLC). Methods Nanobubbles were prepared using a thin-film hydration-sonication method. MTT assay was used to evaluate the cytotoxicity of the nanobubbles for SCLC H446 cell line. The contrast-enhanced ultrasound (CEUS) of xenografted SCLC tumors in 10 nude mice was performed using nanobubbles and micro-scale microbubbles, and compared with livers. The time-intensity curve (TIC) was obtained using the Gamma variate and the following parameters were calculated, including area under the curve, time to peak, arrival time, peak intensity, and half-peak time. Results Nanobubbles with spherical shape distributed homogeneously, without obvious aggregation, the mean diameters was (392.1 ±48.6) nm and average zeta potential was (-16.8 ±2.9) mV. The MTT results indicated that the nanobubbles had no obvious cytotoxicity toward H446 cell line within the concentrations used for in vivo ultrasound imaging with nanobubbles (5 μg/ml). CEUS with the nanobubbles showed significantly higher peak intensity, and half-peak time [(18.14 ±0.62) s, (141.55 ±8.21) s] in comparison with the micro-scale microbubbles [(14.82 ±0.51) s, (120.43 ±8.73) s] (P= 0.033, 0.040). There was no significant difference in time to arrival, area under the curve and time to peak (all P>0.05). Compared with livers, the nanobubbles in xenografted SCLC tumors showed significantly shorter time to peak, lower peak intensity and area under the curve, and higher half-peak time (all P < 0.05). There was no significant difference in time to arrival (P > 0.05). Conclusion Nanobubbles ultrasound enhanced contrast agent shows good stability and contrast-enhancement effect in vitro, and provides an experimental basis for targeting ultrasound imaging and therapeutics of SCLC.
