1.Analysis of Genotypes and Drug Resistance of Extended-Spectrum-Lactamases-Producing Strain in Pat-hogenic Gram-Negative Rod in Infection of Newborn in Guangzhou

Objective To investigate the distribution,drug resistance characteristics and genotypes of extended-spectrum-lactamases (ESBLs)-producing strain in pathogenic gram-negative rod in infection of newborn in Guangzhou.Methods The standard was performed by the production for ESBLs by phenotypic screening and confirmatory test provided by the National Committee for Clinical Laboratory Standards in 2001.The method of polymerase chain reaction(PCR) amplification was perbonmed and DNA sequences were analyzed by ESBLs gene sequencing.Results Total of 71 un-repicated and consecutive Gram-negative bacilli were isolated from 13 hospitals in Guangzhou,and the prevalence of ESBLs-producing clinical Gram-negative isolates was 59.2%(42/71).The PCR results showed that most pathogenic bacilli which infected newborn could be separated two or more genes of ESBLs.The type of TEM,SHV,CTX-M1,CTX-M9,OXA was 35.6%, 26.7% ,10.9%,24.8%,2.0%,respectively.The result of drug resistance monitoring showed that pathogenic gram-negative bacillui which infected newborn were Escherichia coli and Klebsiella pneumonia mostly.Most parts of them were drug fast and even multidrug resistant to the commonly used antibiotics.The sensitive drugs were lmipenem(the rate of sensitivty 100%),cefoperazone/sulbactam(87.3%),piperacillin/tazbatam(85.3%),ceftazidime(82%),aztreonam(82%),cefepime(81.8%).Conclusions In Guangzhou,the incidence rate of ESBLs-producing strain are very high inpathogenic bacilli which infected in newborn and is multidrug resistance.The genetypes of produced ESBLs are TEM,SHV,CTX-M1,CTX-M9,OXA.

It is to observe the therapeutic action of Guizhi Fuling capsule and the combination of active ingredients on model rats with uterine leiomyoma. The hysteromyoma rats models was established in rats by loading eatrogen, to observe the effect on pathological condition of uterus, uterus wet weight, the content of estradiol and progesterone. Guizhi Fuling capsule and the combination of active ingredients remarkably decreased uterus weight, restrained the excess proliferation of the smooth muscle of uterus, decreased the estraiol and progesterone in blood serum. Guizhi Fuling capsule and the combination of active ingredients can restrain the formation of hysteromyoma in a dose-dependent manner. Perhaps the combination of active ingredients is the material foundation of antihysteromyoma.

4.Research strategies and applications of functional metabolomics in anti-depressive mechanisms of traditional Chinese medicine

Depression is a common emotional disorder that seriously affects people's life and health all over the world. The pathogenesis of depression is complex, and traditional Chinese medicine (TCM) for antidepressants has a good therapeutic effect because of its multi-component, multi-pathway, and multi-target action mode. At present, the anti-depressive mechanism of TCM has not been fully clarified, but it is clear that depression is closely related to metabolic health. Therefore, in order to further explore the anti-depressive mechanism of TCM, this paper proposes research strategies on the anti-depressive mechanism of TCM based on functional metabolomics from the perspective of metabolism, the potential biomarkers of depression are analyzed with the help of multi-omics combined analysis technology, and the functional molecules of TCM for antidepressant are studied. Molecular biology techniques are used to accurately capture the molecular interactions between biomarkers of depression and functional compounds, which identify effective drug targets and further elucidate the biochemical functions and related mechanisms involved in depression metabolic disorders. This paper systematically reviews the research strategies and applications of functional metabolomics in the anti-depressive mechanisms of TCM, expounds on the core value of functional metabolomics, and summarizes the current research status and hot issues of TCM for antidepressants in recent years, providing new methods and new ideas for the study of mechanisms of TCM with the help of functional metabolomics.
5.Analysis of risk factors related with hyperuricemia among 1420 youth and middle-aged adult health check-up residents in Guangzhou

To survey the serum uric acid (SUA) levels and associated risk factors of hyperuricemia among youth and middle-aged residents in Guangzhou.A total of 1420 subjects,aged from 20 to 60 years,receiving health check-up at our hospital in 2010 were enrolled.The total prevalence of hyperuricemia was 22.04%,32.01% in males and 14.07% in females.The average SUA was (388 ±78) μmol/L in males and (288 ± 63) μ mol/L in females.The prevalence of hyperuricemia in males was 30.11% before 40 years of age and 33.81% between 40 and 60 years of age.The average level of SUA in males was significantly higher than that of females.logistic regression analysis showed that BUN,body mass index (BMI) and hypertriglyceridemia were the independent risk factors of disease while HDL-C and gender (females) the protective factors.
6.Effects of subanesthetic dose of ketamine on perioperative serum cytokines in orthotopic liver transplantation.

OBJECTIVETo evaluate the effects of ketamine on perioperative serum cytokine levels in patients undergoing orthotopic liver transplantation (OLT).
METHODSTwenty patients undergoing OLT were randomly divided into ketamine group (n=10) and control group (n=10). Patients in ketamine group were given intravenous bolus injection of ketamine at 0.25 mg/kg followed by ketamine infusion at 0.5 mg.kg(-1).h(-1) until the end of operation except in the anhepatic phase, whereas the control group received saline of the same amount. Arterial blood samples were obtained at the start of surgery (T(1)), 5 min before the anhepatic phase (T(2)), 5 min before recirculation (T(3)), 15 and 60 min after recirculation (T(4), T(5)), and 0, 4 and 24 h after operation (T(6), T(7), T(8)). Serum levels of tumor necrosis factor-alpha (TNF-alpha), interleukin-6 (IL-6) and IL-10 were measured by ELISA.
RESULTSSerum TNF-alpha, IL-6 and IL-10 levels increased significantly during anhepatic phase as compared with the baseline level (T(1)) (P<0.05), and the changes were especially obvious in IL-6 and IL-10. The levels of the cytokines kept rising after recirculation and reached the peak level at T(5)(P<0.05), followed then by rapid decline and still maintaining higher levels than the preoperative ones 24 h after operation. The levels of TNF-alpha in ketamine group between T(2) and T(7) were significantly lower than that in the control group, and the IL-6 level between T(2) and T(5) were also significantly lower in ketamine group. Serum IL-10 level did not show any significant difference between the two groups.
CONCLUSIONIschemia and reperfusion injury of the liver and surgical stress induce pro- and anti-inflammatory cytokine responses during liver transplantation, in which event IL-6 and IL-10 are more sensitive than TNF-alpha. Ketamine can inhibit the production of TNF-alpha and IL-6 but not IL-10.

7.Study on intra-retinal layers imaged with ultra - high resolution optical coherence tomography

To evaluate the automated segmentation algorithm for detection of intra - retinal layers to process images obtained from ultra- high resolution optical coherence tomography ( OCT ) . Graph theory and the shortest path search based on dynamic programming were applied to automatically segment the 8 intra - retinal layers. We experimentally verified the accuracy and reliability of the algorithm. The results showed that the intra-retinal layer boundaries between automated and manual segmentations matched well. The algorithm successfully segmented the intra- retinal layers in glaucoma, high myopia, and retinitis pigmentosa patients. The proposed automatic segmentation for intra-retinal layers provides a promising tool for quantitative analysis in clinical diagnosis and treatment.
8.Effects of total flavonoid of chrysanthemum on Fas and Fasl expression in lacrimal gland cells of male castrated rabbits with dry eye

AIM: To evaluate the effects of total flavonoid of chrysanthemum on the expressions of Fas and FasL in male rabbits with dry eye, and to investigate the therapeutic effects of the total flavonoid of chrysanthemum on dry eye.
METHODS: Totally 150 male Japanses white rabbits were divided into blank group ( group A ) , sham -operated group ( group B ) , model group ( group C ) , androgen control treatment group (group D), and total flavonoid of chrysanthemum treatment group ( group E ) . The dry eye model was established with orchiectomy on group C, D and E. Rabbits in group E were treated with total flavonoid of chrysanthemum. Rabbits in group D were treated with androgen intramuscular injection. Rabbits in the group A, group B, group C was treated with normal saline. All rabbits were detected with Schirmer's Ⅰ test and tear break-up time (BUT). Fas, FasL were checked on immunohistochemistry.
RESULTS:The Schirmer's I test values of group E was significantly higher than that of group C ( P<0. 01 ) and the BUT value of group E was significantly longer than that of group C ( P<0. 01 ). The quantity of positive expression of Fas in glandular tube cell and acinar epithelial celland apoptosis cells of group E after treatment at 1, 3, 5mo were significantly lower than that of group C, cell population of the positive expression of FasL was obviously higher than that of group C (P<0. 01).
CONCLUSION:The main component of chrysanthemum is flavonoid, which could significantly inhibit happening of dry eye in rabbit after androgen level lowered and lacrimal gland apoptosis and keep basic tears secretory volume and tear film stability.
9.Morphologic Localization and Purified Cultivation of Sinoatrial Nodes Isolated from Newborn Rabbits

Objective To localize the sinoatrial node (SAN) of the newborn rabbits in vivo and cut it for purifying cultivation and study the morophologic characters of primary cultured pacemaker cells of SAN under light microscope and transmissional electron microscope. Methods Hearts of the newborn rabbits were embedded in paraffin for HE-staining and observed the location, form of SAN under optical microscope; SAN cells isolated from neonatal rabbits cultured and purified with the method of differential attachment and BrdU-treatment.Results SAN localized in the anterior wall of the superior vena cava and the posterior-lateral atrial wall.There was about 0.32 mm between its lowest point and sulcus terminalis. Three distinctly different types of cells were observed among the cultured cells of SAN: spindle, araneiform and polygon. The spindle cells covered the greatest proportion of the cultured cells of SAN (59.6%?7.3%). The frequency of spontaneous contraction of spindle cells was the highest among the constrcting cells (145 ?9)time/min. The results of ultrastructure observation showed that myofibrils and other organelles in spindle cells were poorly organized and significantly decreased in number compared with araneiform cells. There was no significant difference between araneiform cells isolated from SAN and from atrial muscle.Conclusion Among the cultured cells from neonatal rabbits SAN, the spindle cells are the pacemaker cells of SAN.
10. Effects of DJ-1 gene silencing on proliferation and apoptosis of the endometrial carcinoma cell line Ishikawa

Objective: To investigate the effects of DJ-1 gene silencing on proliferation and apoptosis of endometrial cancer Ishikawa cells. Methods: After the specific interference plasmid pGPU6/GFP/neo-DJ-1-small hairpin RNA (shRNA) targeting DJ-1 gene was transfected into Ishikawa cells, the change of cell proliferation was detected by MTT method, the expressions of DJ-1 mRNA and protein were detected by real-time fluorescence quantitative PCR and Western blotting, as well as the apoptosis of Ishikawa cells was detected by Annexin-V/propidium iodide (PI) dual staining and flow cytometry, respectively. Results: The transfection of pGPU6/GFP/neo-DJ-1-shRNA significantly inhibited the proliferation of Ishikawa cells in a dose- and time-dependent manner (P < 0.05). The expressions of DJ-1 mRNA and protein in Ishikawa cells transfected with pGPU6/GFP/neo-DJ-1-shRNA were significantly lower than those in the control groups (P < 0.01). The apoptosis rate in pGPU6/GFP/neo-DJ-1-shRNA transfection group was significantly higher than that of the control groups (P < 0.01). Conclusion: DJ-1 gene silencing can inhibit the proliferation of endometrial cancer Ishikawa cells and promote the apoptosis.
