1.Ionizing radiation activates the HIF-lα-Survivin pathway in human lymphoma cells

Objective To determine the levels of radiation-induced expressions of HIF-1α and Survivin in three human lymphoma cell lines with or without HIF-1a specific antagonist or HIF-1a siRNA transfection and to explore the possible mechanism of cell radioresistance.Methods Namalwa,Ramos,Raji cells were irradiated with X-rays with or without the combination of echinomycin (EC),a HIF-1α specific inhibitor,or transfection of HIF-1α siRNA.Then the expression levels of HIF-1α and Survivin proteins and mRNAs were determined by Western blot and real-time PCR,respectively.Results After irradiation,the expression level of HIF-1α protein was up-regulated at 10-20 h followed by up-regulation of survivin protein at 24 h (t =7.53-31.31,P < 0.01 ) in three lymphoma cell lines.Radiation-induced up-expression of survivin protein was significantly reduced by either HIF-1α inhibition or EC treatment (t =7.21-32.81,P <0.01 ).However,radiation failed to induce up-erpression of survivin at both mRNA and protein levels after transfection with HIF-1α siRNA.Conclusion HIF-1α-Survivin pathway is activated in human lymphoma cells after radiation,which might be involved in the cell radioresistance.
2.Effect of recombinant human erythropoietin injection on the nutritional quality and cardiac function in the patients with maintenance hemodialysis

Objective To investigate the effect of recombinant human erythropoietin (rhEPO)injection on the nutritional quality and cardiac function in the patients with maintenance hemodialysis.Methods Sixty-two patients with chronic kidney failure were divided into two groups by random digits table:treatment group(46 patients, rhEPO injection therapy was given) and control group( 16 patients, without rhEPO injection therapy). The serum biochemical indexes and color sonography was respectively performed before and after the treatment. Results In treatment group, the cardiac function and the serum biochemical indexes improved obviously after 6 months' treatment (P < 0.05 ). But there was no change in control group.Conclusion The treatment with rhEPO injection can effectively improve the nutritional quality and cardiac function in the patients with maintenance hemodialysis, and improve life quality.
3.Inhibition of NF-kB enhanced X-ray irradiation induced apoptosis in human non-Hodgkin lymphoma

Objective To investigate the role of NF-kB in radiation-induced apoptosis ot human non-Hodgkin lymphoma (NHL) cells and the mechanism involved.Methods Three human non-Hodgkin lymphoma cell lines,Namalwa,Ramos,and Raji cells were divided into control,IR and IR + QNZ ( 10nmol/L) groups,respectively.Annexin-V kit was used to determine cell apoptosis. Protein expression levels of Survivin,Bax,Bcl-2 and cleaved Caspase-3 were evaluated by Western blot.Survivin mRNA was quantified by real-time PCR.Results Inhibition of NF-kB by QNZ pretreatment significantly enhanced X-ray induced apoptosis in human NHL cells in a dose-dependent manner (t =2.93 - 12.52,P < 0.05).At the same time,QNZ significantly reversed the expression levels of Survivin protein and mRNA that were upregulated by radiation(t =3.29 - 16.72,P < 0.05).QNZ also increased the levels of Caspase-3 and proapoptotic protein Bax,but reduced anti-apoptotic protein Bcl-2 expression level and hence the ratio of Bcl-2/Bax.Conclusions Inhibition of NF-kB could enhance radiation-induced cell apoptosis in human NHL cells through down-regulating Survivin expression and decreasing Bcl-2/Bax ratio.
4.Activation of NF-κB-HIF-1α pathway induced by X-ray irradiation in human lymphoma cells

Objective To observe the effect of X-ray radiation on NF-κB-HIF-1α pathway activation in three malignant lymphoma and to explore the mechanism of radioresistance mediated by this pathway.Methods Expression levels of p-P65,IKK and HIF-1α in Namalwa,Ramos and Raji cells were determined by Western blot 1,4,10 and 20 h after 5 Gy X-ray irradiation.The effect of QNZ on HIF-1α expression was also observed.Results Expression level of IKK protein was up-regulated 4 h after irradiation in the 3 lymphoma cell strains( t = 8.01,5.14,5.42,P < 0.01 ),followed by up-regulation of p-P65 protein expression level at 10 h and HIF-1α at 10-20 h after X-ray irradiation (t = 11.25,17.43,22.09,P < 0.01 ).Pretreatment of QNZ 24 h before X-ray irradiation significantly reduced the upregulation of HIF-1α protein expression induced by simple ion radiation ( t = 18.69,19.35,12.26,P <0.01 ).Conclusion NF-κB - HIF-1α pathway was activated in human lymphoma cells after ion radiation and may be involved in the mechanism of radioresistance.

6.Treatment of post-stroke depression

The post-stroke depression refers to a affective disorders within 2 to 3 years after stroke in patients with stroke.Its main features are depressed mood,decreased interest,irritability,pessimism,and slow thinking,etc.Usually the symptoms will last for more than 2 weeks.It is one of the most common complications of stroke and may seriously affect the rehabilitation and quality of life of patients.Some patients even have suicidal tendency.Early intervention is very important for late neurological recovery in patients with stroke.It can not only improve cognitive function,but also reduce the morbidity and mortality.This article reviews the treatment of post-stroke depression.
7.Advances in Neuroprotective Effects of Estrogen

Most epidemiological investigations and basic studies have demonstrated that estrogen has neuroprotective effects,and the application of estrogen in cerebrovascular disease has a positive attitude.However,large clinical trials of the application of estrogen have indicated that harms outweigh benefits,and even have the risks of stroke.This article reviews the related studies about the neuroprotective effects of estrogen from the positive and negative aspects.
8.Application of oral food challenge in the diagnosis of gastrointestinal food allergy in the children

Food allergy is a common illness in children,which is divided into IgE-mediated,non-IgE mediated and mixed-mediated food allergy.Gastrointestinal tract is the mainly affected organ,mostly mediated by non-IgE mechanism.Gastrointestinal food allergy are lack of unified diagnostic criteria currently,whose mechanism is also elusive.Moreover,the double-blind placebo-controlled food challenge is the gold standard for the diagnosis of food allergy.This review focuses on the procedure for the oral food challenge and its application in the diagnosis of gastrointestinal food allergy.
10.Expression of RhoA in placenta of preeclampsia.

In order to detect the expression of RhoA in placenta from normal pregnancy and preeclampsia and evaluate the role of RhoA in preeclampsia, the expression of RhoA in placenta collected from 40 preeclampsia patients and 20 normotensive controls was determined by immunohistochemistry and RT-PCR. RhoA was found in syncytiotrophoblasts and cytotrophoblasts. The mean density of RhoA protein in placental tissues of mild and severe preeclampsia groups was significantly higher than that in normal pregnancy. The expression level of RhoA mRNA in mild and severe preeclampsia groups was significantly higher than that of normal pregnancy. Increased expression of RhoA in placental tissues might play an important role in the pathogenesis of preeclampsia.
