[Objective]To detect the effect of constant magnetic field on metal ion Co2+,Cr3+ induced TNF-? secreted by human mononuclear cells, and to search a method for prevention and treatment of aseptic loosening. [Methods]CoCl2 powder and CrCl3 powder were dissolved in the asepsis injecting water. Mononuclear cells from human peripheral blood, were taken and cultured with Co2+,Cr3+ ions in different magnetic field of 10Gs,100 Gs,1000Gs for 12,24 and 48 hs. There were nine groups:control group,Co2+ group,Cr3+ group,Co2+andCr3+ with various intensities of constant magnetic field, respectively.ELISA method was applied to detect tumor necrosis factor (TNF-?) in serum via the absorbance (A).[Results]Co2+ and Cr3+ ions stimulated human mononuclear cell to secrete TNF-? (P

Journal of Environment and Health 1993;0(03):-
Objective To observe the effects of pentabrominated diphenyl ether (BED-99) on the histological structure and function of the thyroid of rats. Methods Forty SPF, SD rats aged 4 weeks, were randomly divided into 4 groups according to the body weight,10 in each group. The rats were treated with BED-99 at the doses of 30 mg/kg,60 mg/kg,120 mg/kg,respectively, through gavage. The rats in the control group were received the equal volume of corn oil. The levels of serum hormone were determined, the change of histopathology, histochemistry and follicular cells ultramicroscopic structure were observed at 15 days of exposure to BED-99. Results Serum levels of thyroxin (FT4),triiodothyronine (FT3),decreased gradually with dose increase at 15 days of exposure to BED-99.However,thyroid stimulating hormone (TSH) levels increased. The thyroid follicular epithelium hyperplasia was aggravated gradually with dose increase. Proliferating cell nuclear antigen (PCNA) positive cells increased constantly with dose increase. The activities of thyroperoxidase (TPO) and succinate dehydrogenase (SDH) of the treated rats increased significantly compared with the controls, and appeared color darkened. The rough endoplasmic reticulum of the thyroid follicular cells changed to saccate under the electron microscope, whereas nucleolus, cell membrane and mitochondria were not injured. Conclusion BDE-99 may damage the histological structure and function of the thyroid, the mechanism may be that BDE-99 down-regulates the thyroglobulin synthesis and then induces thyroxin level decrease which causes an increase of TSH through feedback.

Objective To analyze the impact of uterine septum on pregnancy and influencing factors on postopera-tive pregnancy. Methods 125 patients with septate uterus and bearing requirement who underwent TCRS were fol-lowed up to assess fertility outcome. The clinical data was retrospectively analyzed. Results Spontaneous abortion rate was 70.40%and 19.39%, live birth rates was 10.40%and 72.45 %in preoperation and postoperation respec-tively. The difference was statistically significant ( < 0.05). Spontaneous abortion rate in older than 35 years old group was significantly higher than that in younger than 35 years old group, but live birth rate was lower. There was no significant difference in different times of operation in uterine cavity, number of abortion and septum length and so on. Conclusions TCRS can significantly improve pregnancy outcome. The age has influence on postoperative pregnancy outcome. Abortion numbers, septum length, septal base width, intrauterine device (IUD) and hormone re-placement therapy (HRT) may have no effects.

Objective To explore the differences of bacterial culture results and time consumption between the two methods of holding the syringes to draw physic liquors,so as to find the objective and scientific evidences to raise the efficiency of the clinical nursing work and to ensure the medication safety at the same time. Methods Making bacterial cultures for the physic liquors respectively drawn by grasping the piston of syringes (30 cases in the experimental group) and grasping the piston holder of syringes(30cases in the control group).Recorded the time consumption by grasping the piston of syringes to draw the physic liquors (the experimental group) and grasping the piston holder of syringes to draw the physic liquors (the control group). The testing results were compared between the two groups. Results Bacterial cultures:There was no bacteria growth in both groups.Timing:The time spent by grasping the piston of syringes to draw physic liquors was obviously shorter than that by grasping the piston holder of syninges to draw physic liquors. Conclusions Grasping the piston of syringes to draw physic liquors can ensure the medication safety and raise the work efficiency.

Objective To study the effects of siltuximab on the interleukin-6 (IL-6)/signal transducer and activator of transcription 3 (Stat3) signaling pathway in ovarian epithelial carcinoma. Methods (1) Expressions of IL-6 in ovarian cancer patient specimens were assessed by immunohistochemistry.(2) Expression of phosphorylation Stat3 (pStat3) protein in siltuximab and IL-6 treated SKOV3 cell lines was determined by western blot, and expression levels of Stat3-induced bcl-XL,MCL-1, survivin, in siltuximab treated SKOV3/TR and CAOV3/TR cells lines were also determined by western blot.(3) Real-time image analysis was used to study the nuclear translocation of pEGFP-Stat3 fusion protein in ovarian cancer cell line SKOV3-pEGFP-Stat3 treated with siltuximab and IL-6.(4)Paclitaxel sensitivity in siltuximab treated SKOV3/TR and CAOV3/TR cell lines were assessed using the methyl thiazolyl tetrazolium (MTT).The 50% inhibiting concentration ( IC50 ) was defined as the paclitaxel concentration required to decrease the A490 value to 50%.Results ( 1 ) There were significantly difference in IL-6 staining density and the positive rate of IL-6 protein stained among the metastatic, and drug-resistant recurrent tumors, and matched primary tumors [69%(18/26)] vs.77% (20/26)vs.23% (6/26), P<0.05].(2)A clear increase in Stat3 phosphorylation levels was observed in the IL-6-treated SKOV3 cell lines as compared to the SKOV3 cell lines. When the IL-6-treated SKOV3 cells were incubated with siltuximab with a range of concentrations of 0.001,0.01,0.1, 1.0 and 10 μg/ml, there were trends toward reduced pStat3 expression in the treated cell lines.Compared without treatment with siltuximab, the expression of the anti-apoptotic proteins MCL-1, bcl-XL and survivin in SKOV3/TR and CAOV3/TR cell lines were significantly decreased after treated with siltuximab.(3) In resting cells, the majority of pEGFPStat3 was cytoplasmic until the addition of human IL-6, which promptly induced the translocation of fluorescent Stat3 molecules to the nucleus.Exposure of cells to siltuximab with a range of concentrations of 0.001, 0.01,0.1, 1.0 and 10 μg/ml, followed by an incubation in IL-6 significantly reduced pEGFP-Stat3 nucleocytoplasmic translocation.(4) MTT cytotoxicity assay demonstrated that siltuximab increased paclitaxel-induced cell death and partially overcame paclitaxel resistance.Treated with siltuximab ( 1 and 10 μg/ml) ,the paclitaxel IC50 value of siltuximab in SKOV3/TR (0.49, 0.19 μg/ml) and CAOV3/TR (0.0010, 0.0008 μg/ml) cells were significantly lower than those in untreated cells (0.71,0.0021 μg/ml;all P < 0.05).Conclusions These results demonstrated that siltuximab effectively block the IL-6 signaling pathways, which .Blockage of IL-6 signaling may provide benefits for the treatment of ovarian cancer.
