1.Determination of paeoniflorin and baicalin in Weichang Heji by HPLC

AIM: To establish a HPLC method for determing paeoniflorin and baicalin in Weichang Heji(Radix Paconiae alba,Radix Scutellariae,Radix et Rhizoma Khei,etc.). METHODS: The column was Ultimate XB-C_(18)(5 ?m,250 mm?4.6 mm),methanol-water-acetic acid(35∶65∶1) as the mobile phase at a flow rate of 1.0 mL/min,the detecting wavelength was at 244 nm. RESULTS: The linear range was within 25 ?g/mL-250 ?g/mL for paeoniflorin and baicalin. The average recovery rate was(98.12%) and 96.81%,respectively.RSD was 1.16% and 0.95%(n=6) respectively. CONCLUSION: The method is simple and accurate.It can be used to determine the contents of paeoniflorin and baicalin in Weichang Heji meantime.
2.Diagnosis Study Progress of Aortopulmonary Septal Defect

Aortopulmonary septal defect is a rare anomaly,it should be considered whenever the course of complex congenital heart di-sease includes early cardiac failure and pulmonary hypertension.So the importance of early correction to avoid irreversible pulmonary hypertension was emphasized.Noninvasive imaging with echocardiography and (or) 64MRSCT will adequately define the anatomy before surgery,it's necessary to perform cardiac catheterization to evaluate the reversability of pulmonary vessels when pulmonary hypertention is suspected.
3.Study on Clinical and Immunophenotype of Acute Leukemia in Children

Objective To study the clinical significance and characteristic of acute leukemia(AL) immunophenotype expression in children.Methods Immunofluoscence labeled with three and two color monoclonal antibodies directly were used to analyze the surface antigen and cytoplastic antigen.Cytoplastic antigen was detected by flow cytometry additionally.Results Specimens from 85 patients with childhood acute leukemia were detected.1.The undifferentiated phenotype account for 1 case(1.2%);Mixed leukemia 7 cases(8.2%);Acute myeloid leukemia 18 cases(21.2%);Acute lymphoid leukemia 59 cases(69.4%).The subtypes of acute lymphoid leukemia:(B-ALL) 50 cases(84.7%),T-ALL 6 cases(10.2%),T/B 3 cases(5.1%).2.Among 59 cases of acute lymphoid leukemia,there were 31 cases ALL expression myeloid-associated antigen;There was no difference on the clinical and prognosis between the group of myeloid-associated antigen positive and the negative group.3.Of 18 patients with AML,5 patients(27.8%) expressed lymphoid antigen,CD7 was the most commonly expressed(60%);The frequency of CD_(14) expression in childhood M4,M5 was 75%.CD_(13),CD_(33) presented in 83.3% and 94.4% cases of AML,respectively.4.Among 7 cases acute mixed leukemia,B/M 6 cases,TB/M 1 case.Five cases were misdiagnosed as ALL by FAB classification.Conclusions With the multi-color flow cytometry,every acute leukemia patient was diagnosed accurately,especially on acute mixed leukemia and cross-lineage expression leukemia.Those help us choice the best treatment protocol for childhood AL.
4.Identification of nuclear localization signals of pseudorabies virus gene UL49.

Tegument protein VP22 is encoded by Pseudorabies Virus (PRV) UL49. To identify the nuclear localization signals of UL49, it is necessary to determine the transport mechanism and biological functions of the VP22 protein. In this study, we identified two nuclear localization signals from UL49, NLS1 (5RKTRVA ADETASGARRR21) and NLS2 (241PGRKGKV247). The functional nuclear localization signal (NLS) of UL49 was identified by constructing truncated or site-specific UL49 mutants. The deletion of both NLS1 and NLS2 abrogated UL49 nuclear accumulation, whereas the deletion of NLS1 or NLS2 did not. Therefore, both NLS1 and NLS2 are critical for the nuclear localization of UL49. And our resuts showed that NLS2 is more important in this regard.

6.Binding mechanism of traditional Chinese medicine active component 5-hydroxymethyl-furfural and HSA or BSA.

A combination of spectral experiment and molecular modeling techniques has been used to characterize the binding mechanism between an active component 5-hydroxymethyl-furfural (5-HMF) of traditional Chinese medicine and human serum albumin (HSA) or bovine serum albumin (BSA). The interaction mechanism of 5-HMF binding with HSA/BSA is analyzed. Although the drug can bind with HSA/BSA to form stable complexes, there are some differences in the bond strength. The values of binding distances (r) are different and low, which indicated the occurrence of energy transfer. The drug had conformational effect on HSA/BSA, which resulted in different changes of hydrophobic environment of the binding domain in HSA/BSA. The 'phase diagram' of fluorescence revealed that the changes on the conformational pattern of proteins have been affected by drug conformed to the "all-or-none" pattern. The interactions between drug and protein influenced by Co(II) were also discussed. Its effects acting on 5-HMF-HSA/BSA interactions are different. The computational modeling method was used to study the interaction between 5-HMF and HSA/BSA. The results of molecular model studies revealed that the binding modes for drug-serum albumin systems are mainly hydrophobic interactions and hydrogen bonding. These results are in accordance with spectral results. The research results have given a better theoretical reference for the study of pharmacological mechanism of 5-hydroxymethyl-furfural.
7.Mechanism of Antiasthma by Inhaled Arsenic Trioxide

Objective To investigate the effect of eosinophils(EOS)in asthmatic guinea pigs airway and explore the mechanism of antiasthma by inhaled arsenic trioxide. Methods The guinea pigs asthma models were established with ovalbumin(OVA)challenge method,2 groups received inhaled different doses of arsenic trioxide,and three control groups respectively received normal saline inhaled and high dose of arsenic trioxide by intraperitoneal injection and dexamethasone(DEX)intraperitoneally. EOS invaded and apoptosis with all of the groups were assessed after 7 days intraperitoneal injection or inhalation. Results Compared with group of inhaled normal saline, both EOS invaded and apoptosis in bronchus submuconsa were significantly different(P0.05)among groups of inhaled low dose arsenic trioxide [2.0 mg(kg?d)]and intraperitoneal injection with higher dose arsenic trioxide[5.0 kg/(kg?d)]and DEX[10 mg/(kg?d)]for these two parameters. Conclusions The mechanism of arsenic trioxide antiasthma with arsenic trioxide can decrease EOS amount in bronchial submucosa and accelerate EOS apoptosis, and relieve bronchial inflammation in asthma. For inhaled lower dose arsenic trioxide or intraperitoneal injection with high dose arsenic trioxide ,the effect of EOS is equivalent. The antiasthma effect of inhaled lower dose arsenic trioxide[2.0 mg/(kg?d)]is equivalent to intraperitoneal injection higher dose[5.0 mg/(kg?d)],and may be safe comparatively. The mechanism of antiasthma by inhaled arsenic trioxide is the same with DEX.
8.HISTOLOGICAL EXAMINATION OF THE RELATION BETWEEN THE CENTRAL FIBROUS BODY AND THE CONDUCTION SYSTEM

The shape and position of the central fibrous body (CFB) and the relation between the CFB and the conduction system were observed in 30 human hearts (children: 25 cases, adult: 5 cases) in serial sections.The shape of the CFB was wedge-shaped. The CFB of each child was horizontal in position, but the CFB of each adult was nearly sagittal in position.A-V node was a long sagittal flat structure. Its left surface contacted with the CFB and the left surface of A-V node often extended processes into the CFB in the children. The processes were called archipelagos. About 80 percent of the children with superficial archipelagos, and 16 percent with deep archipelagos. The superficial archipelagos in the adults was only found in one case, but the deep archipelagos were not seen.His bundle more permanently penetrated into the right inferior part of the CFB.
9.The value of 3D TOF MRA in evaluation of anomalous vertebral artery in healthy adults

Objective: To describe 3D-TOF MRA features of anomalous vertebral artery(VA) and evaluate its clinical significance in healthy adults who were accepted physical examination. Methods: A total of 442 cases in which 3D-TOF MRA was performed were retrospectively reviewed to discuss the incidence and relationship between VA origin and course variation. Results: Among this 442 cases, the incidence of origin variation was 4.75%(21/442) in which all cases were unilateral(19 cases in the left and 2 cases in the right). In these cases, the anomalous VA originated from the aortic arch or the right common carotid artery. All of the 21 cases with origin variation had the aberrant entrance position into the transverse foramen (4th or 5th). The incidence of abnormal entrance position into the transverse foramen was 9.50%(42/442),of which most cases(95.24%, 40/42)were unilateral. Of 40 cases, the incidence of both sides was the same(50.00%(20/40)). Conclusion: The VA origin variation always happened in the left side, and often combined with the course variation. 3D-TOF MRA plays an important part in the detection of the anomalous vertebral artery in healthy adults. It can provide useful information for the surgery and the therapy of related diseases.
10.Investigation of morphologic variations of circle of Willis using 3D-TOF MR angiography in medical examination and its importance

Objective:To investigate morphologic variations of circle of Willis in healthy adults using 3D-TOF MR angiography in medical examination and its clinical significance. Methods:To analyze the characteristics of the vascular morphology in 442 healthy adults of head 3D-TOF MRA in the post processing workstation retrospectively. Results: All the 442 cases, the variations of circle of Willis were found in 152,the variations of A1, A2, ACoA, PCoA, P1 were found in 76(50.00%), 23(15.13%), 7(4.61%), 101(66.45%), 68(44.74%). The variation of A1 was positively correlated with PCoA, the variation of PCoA in female was more than in male. Conclusion:Medical examination-3D-TOF MRA has important value to display morphologic variations of circle of Willis in healthy adults, which can provide important guide value in the clinical.
