RESULTS: The patients with FD in two groups were matched well in respect of age, sex and index of dyspeptic symptoms. Routine therapy was achieved and adverse reaction wasn’t found. Curative efficacy and symptom score in treatment group were significantly better than in control group after 4 weeks of treatment, such as poor appetite, abdominal distension, early satiety, belching , except upper abdominal pain and burning sensation (P
8.Immunometabolism and regulatory T cells
Chinese Journal of Microbiology and Immunology 2017;37(5):396-399
Recent studies have shown that immune cells play an important role in the development of non-immune diseases.Regulatory T cells (Treg) are a group of T cell subsets which have immunoregulatory and immunosuppressive functions.Abnormalities in the number and function of Treg cells may cause immune imbalance, which leads to the development of diseases.Immunometabolic disorders may in turn affect the generation, proliferation and function of Treg cells.Clarification of the relationship and interaction between immunometabolism and Treg cells will make it possible to treat metabolic diseases with cellular immunotherapy.

Chinese Journal of Applied Clinical Pediatrics 2017;32(7):543-547
Objective To determine the therapeutic efficacy of curcumine on inflammatory bowel disease(IBD)and its underlying mechanisms. Methods The rat ulcerative colitis model was developed by using 30 g/L dextran sulfate sodium.Twenty-four young rats were divided into 4 groups:normal group, model group, curcumin 100 mg/kg group(curcumin 100 group),and curcumin 300 mg/kg group(curcumin 300 group).Curcumin was given through gastric gavage once a day for 7 days.General condition, disease activity index(DAI)and histopathological score(HPS)were evaluated. The proportions of CD4+ CD25+ Foxp3+ regulatory T cells(Treg)and CD4+ IL-17+ helper T lymphocyte 17(Th17)in CD4+ T cells in rats, and spleen cells were calculated by using flow cytometry.Immunohistochemical method was performed to determine the level of SOCS3 in colon tissues. Results After curcumin administered through gastric gavage for 5 days, compared with the DAI in the model group[(7.50±0.32)scores],HPS in the curcumin 100 group[(4.00±1.10)scores] and the curcumin 300 group [(5.00±0.89)scores] significantly reduced, and the difference was significant(F=12.469,P<0.05).After curcumin administered through gastric gavage for 7 days, compared with the HPS in the model group[(2.69±0.70)scores],histological score in the curcumin 100 group[(1.94±0.68)scores],the curcumin 300 group [(2.00±0.63)scores] and the normal group[(0.50±0.05)scores] significantly reduced, and the difference was significant(F=33.282,P<0.05).Compared with the model group[(0.015 1±0.000 7)D],the expression of SOCS3 in the curcumin 100 group[(0.002 0±0.000 5)D],the curcumin 300 group [(0.002 6±0.006 0)D] and the normal group[(0.002 2±0.001 0)D] was significantly inhibited(F=382.270,P<0.05),but there was no difference among the former 3 groups(F=0.828,P>0.05).Compared with the model group[(6.5±1.5)%],the propotion of CD4+ CD25+ Foxp3+ Treg cells in CD4+ T cells in the curcumin 100 group[(9.9±1.0)%],the curcumin 300 group [(9.3±0.7)%] and the normal group[(9.6±1.1)%]was obviously upregulated(F=16.635,P<0.05),but there was no difference among the former 3 group(F=1.564,P>0.05).Compared with the model group[(3.5±1.4)%],the propotion of CD4+ IL-17+ Th17 cells in CD4+ T cells in the curcumin 100 group[(2.0±0.9)%],the curcumin 300 group [(1.2±0.6)%] and the normal group[(2.0±0.9)%] was downregulated(F=5.155,P<0.05),but there was no difference among the former 3 group(F=2.073,P>0.05).There was no obvious difference between the curcumin 100 group and the curcumin 300 group in these indicators respectively(all P>0.05).Conclusions Curcumin probably attenuates IBD severity, reduces inflammation and regulates the balance of Treg/Th17 through the inhibition of SOCS3 expression.
10.Dendritic cells differentiated from 129/svj ES-D3 embryonic stem cells prolong syngeneic neural progenitor cells survival in ischemic rat brain
Chinese Journal of Immunology 2015;(3):318-322,328
Objective:To study if embryonic stem cell derived dendritic cells(esDCs) could induce transplant tolerance to syngeneic neural progenitor cells ( NPCs) in ischemic rat brain. Methods:Neural progenitor cells ( NPCs) were differentiated from 129/svj pCX-eGFP ES-D3 embryonic stem cells and dendritic cells were directly differentiated from 129/svj ES-D3 respectively. All of SD rats were accepted MCAo surgery and subdivided in two groups based on pretreatment with or without esDCs through tail vein injection 1 week after MCAo.pCX-eGFP NPCs were then injected into the lateral ventricle of animals 2 weeks after MCAo.A proliferation assay of lymphocytes dissociated from cervical lymph nodes by MTT method, counting of the survival of the grafted cells, histological evaluation of CD4,CD8 and ED1 positive cells in brain and detection of mRNA level of IL-10 and IFN-γin ischemic lesions by reverse transcriptase-polymerase chain reaction(RT-PCR) were performed 2 weeks after graft (4 weeks after MCAo).Results:Pre-treatment with esDCs decreased CD4 positive cells infiltration (134.7 ±36.2 vs.198.8 ±59.6,P<0.05) and enhance the survival of NPCs in ischemic striatum, but had no effect on ED1 (298.8 ±75.9 vs.302.2±88.5),CD8 positive cells (145.8 ±45.4 vs.134.3 ± 39.0) distribution (P>0.05).There was also no difference in lymphocytes proliferation (PI,1.245 ±0.211 vs.1.331 ±0.235) or mRNA expression level of IL-10 ( 1.147 ±0.260 vs.1.264 ±0.119 ) and IFN-γ( 1.697 ±0.273 vs.1.829 ±0.250 ) between two groups ( P>0.05).Conclusion:The results indicate that pretreatment with esDCs may prolong syngeneic NPCs survival though reducing CD4 positive cells reaction in ischemic striatum, which provides some evidence for the tolerogenic function of esDCs.However, there was lack of evidence for cytokine-dependent routine involving in this mode and further investigation was needed to make certain the cardinal principle.
