1.Clinical comparison and analysis of laparoscopic and open operation for colorectal neoplasms

Objective To evaluate the feasibility and short-term outcome of laparoscopic operation for colorectal neoplasm in comparison with open operation procedure for colorectal neoplasm, laparoscopic operation for colorectal neoplasm may be critical in the evolving practice of medieine. Methods 234 cases of colorectal neoplasm were divided into two groups and was analyzed. 103 patients with colorectal neoplasm underwent laparoscopic operation (LCS), while 131 cases received open operation (OCS). Results The mean operating time in laparoscopic operation group was significantly longer than in open operation group (P <0.05). The blood loss was less. Time to pass flatus, time to ambulate and return of bowel function were restored earlier in laparoscopic operation group than that in open operation group (P <0.05). Duration of hospital stay were longer in open operation group than that in laparoscopic operation group (P <0.05). There was not significant difference in the length, distal margin, size of tumors and number of removed lymphatic nodes(P > 0.05) between the two groups. Laparoscopic colorectal resection is better in pathologic sample compared with the traditional open operation procedure. Conclusion Laparoscopic colorectal resection is secure and effective for colorectal neoplasm, and it is with lower blood loss during operation,earlier return of bowel function and shorter hospitalization, and provide better short-term outcome compared with the traditional open procedure.
2.Concentration changes and clinical significance of hepatocyte growth factor in acute kidney injury ICU pa-tients

Objective To investigate the concentration changes and clinical significance of hepatocyte growth factor (HGF) during the development of acute kidney injury (AKI). Methods The serum concentrations of HGF in 62 patients with AKI were measured every two days, and the relation between HGF and glomerular filtration rate (eGFR) ,the renal failure index,course of diseases,numbers of viscera injury,and short-term prognosis were an-alyzed. Results The serum concentrations of HGF were progressively rising along with the course of diseases and the amount of viscera injury (P=0.000), reached peak at the 9th day, and then decreased, which was was elevated in the death group than renal function failure group, but which in the latter group was elevated than in renal function recovery group (P<0.05 for each). AKI with liver injury patients were obviously higher than without liver injury pa-tients(P<0.05 for each). The serum concentrations of HGF were positively correlated with the renal failure index (r=0.580~0.821) but negatively correlated with eGFR(r=-0.943~-0.703) (P<0.05 for each). Conclu-sion The serum concentrations of HGF are abnormally rising in the patients of AKI ,which reflects the state of dis-ease and short-term prognosis of AKI.
4.Immunity mechanism of vertical transmission blocking for HBV immunoglobulin injection to pregnant women

Objective:To discuss the passive immunity effect and mechanism of HBIG injections to matrix to block the mother-to-child vertical transmission of HBV. Methods:94 cases of patients with chronic viral hepatitis were selected and divided into 3 groups randomly. 31 cases of control group were given no HBIG intervention,while 31 cases of baby intervention ( BBI) group were given HBIG injection in 6h of birth,and 32 cases of infant & mom intervention ( IMI) group were given HBIG injection respectively in 28,32,36 weeks of gestation and 6h of birth. Further more,all newborns were vaccinated against hepatitis B in 0,1 and 6 months,after the last vaccination,peripheral blood of the children were extracted and detected for HBV markers,HBV-DNA and immune function. Results:There were significant difference (P<0. 05) in neonatal HBeAg,HBsAg and HBV-DNA positive rate for the three groups,with Control group got the highest while IMI group got the lowest;and there are also significant differences (P<0. 05) HBeAb positive rate,with Control group got the lowest while IMI group got the highest. We also found that the complement (C3,C4) levels and T cell subtypes (CD3+,CD4+,CD8+) count of the three groups of newborns had significant differences too(P<0. 05),with Control group got the lowest while IMI group got the highest;in terms of immunoglobulin,both the IMI and BBI group were higher in IgG and IgM level (P<0. 05), while there was no obvious difference in IgA between groups (P<0. 05). Conclusion:Maternal HBIG injections can effectively activate the maternal humoral immunity and cellular immunity,resulting in the decrease of HBV. It can also improve newborn′s antigen-antibody response and relieve T lymphocytes loss induced by antiviral consumption through placenta,which may play great role in the passive im-munity mechanism of blocking mother-to-fetus transmission.
5.Determination of Phosphorus and Silicon in Edible Vegetable Oil by Inductively Coupled Plasma-Tandem Mass Spectrometry

An analytical method was developed for determination of P and Si in edible vegetable oil using inductively coupled plasma-tandem mass spectrometry (ICP-MS/MS).The microwave-assisted acid digestion of vegetable oil using HNO3 + H2O2 was carried out in closed vessels.The mass spectral interferences were eliminated by O2 mass shift when promoting reaction with O2 inside the collision reaction cell (CRC), and the monitoring of P as 31P16O+ product ion significantly improved the accuracy of the analysis.H2 was added into the CRC for H2 on-mass reaction.The interferences were eliminated by the quadrupole analyzer to accurately identify 28Si+.The effects of the flow rate of O2 and H2 in ORS3 on the signal intensities and BECs of 31P16O+ and 28Si+ were investigated.The optimum O2 and H2 flow rate was determined.Under the optimized conditions, the limits of detection were 0.043 and 0.66 μg/L for 31P16O+ and 28Si+, respectively.The accuracy of the analytical method was assessed by the analysis of the standard reference materials lubricant oil (SRM 1848) from the National Institute of Standard and Technology.No significant differences were observed between the certified values and measured values.This method was used to analyze 5 kinds of edible vegetable oils (rapeseed oil, sunflower oil, peanut oil, corn oil and soybean oil) from different regions of China, and it was found that the content of P was the highest in peanut oil, and Si showed the highest content in soybean oil.
6.Study of non-linearity relation of MTS cytotoxicity assay

AIM We found in experiment that the relation of cell number-OD in MTS cytotoxicity assay is non-linearity. In this paper we probe into the relations of cell number-OD and drug dose-inhibition rate. METHODS Cells HL-60 and Raji were used in this experiment. Assay on the relation of cell number-OD: Cells of different concentrations were seeded into 96 well plates. OD was read at 490 nm after incubating with MTS for 1~5 hour. The regression curves were estimated from the data with the software SPSS 11.0. Cytotoxicity assay: Cells were seeded into 96 well plate and incubated with drug of different doses for 72 hours. OD was read at 490 nm after incubating with MTS for 3 hour. The regression curves were estimated from the data with the software SPSS 11.0. RESULTS The cubic curves fit well the relations of cell number-OD of the 2 strains of cells at 5 time points and the coefficient R2 of these cubic curves is 0.997~1.000, greater than the R2 0.938~0.993 of linear model. For the relations of dose-inhibition rate, the cubic curves also have best fitness and their R2 is 0.998~1.000 greater than the R2 0.948~0.987 of linear model of logarithm dose-probit. CONCLUSION For the relations of cell number-OD in MTS cytotoxicity assay, the fitness of cubic curves are better than the linear model. A more accurate IC 50 would be obtained by the cubic curve equation than by probit regression that is in common use.
8.Study on application of amphotericin B in the perforated whole-cell patch clamp technique.

OBJECTIVEEstablished with amphotericin B perforated patch-clamp technique, to study the electrophysiological properties of calyx synapses.
METHODSIn the present experiments, we studied the application of perforated patch clamp technique on the calyx synapses of mice with Amphotericin B.
RESULTSThe use of Amphotericin B significantly slowed down the decay of channel currents and the optimum concentration was 400 microg/ml.
CONCLUSIONThe syudy developed a stable of perforated patch clamp whole cell recording technique, could be more effecitve, more real reaction neurons electrophysiological characteristics of the channel current. Our work might provide the basic information to future users studying the signal transmission and regulation of auditory system of rodents.

10.Assessment of the results of an investigation on eliminating iodine deficiency disorders in Ya'an city of Sichuan province in 2009

Objective To evaluate the investigation outcome of eliminating iodine deficiency disorders(IDD)in Ya'an city of Sichuan province,and to master the current situation of IDD in the whole city.Methods In 2009,according toCounties'Assessment Guideline in Sichuan for Implementing the Measure to Eliminate Iodine Deficiency Disorders,assessments were conducted.At county level,composite score was given to collected data of the eight counties(districts)and monitoring data of household iodized salt were statistically analyzed.A township (town)was randomly selected by east,west,south,north and center locations,respectively.One primary school was randomly selected in each selected township(town),eighty children aged 8 to 10 in each selected school were checked of their thyroid by palpation(WS 276-2007)and urinary iodine level of20 children among them was measured by As3+_Ce4+ catalytic spectrophotometry(WS/T 107-2006).Thirty fifth-grade students and 10 housewives lived around school were asked to take a questionnaire survey.Results Average score of the 8 counties(districts)was 89.96,ranged from 85.42 to 92.90.The coverage rate of iodized salt of household was 100.00%(2328/2328),rate of qualified iodized salt was 97.98%(2281/2328).and consumption rate of qualified iodized salt reached 97.98%(2281/2328).The goiter rate of children aged 8-10 was 0.91%(33/3620),median urinary iodine was 235.85μg/L,and 1.50%(12/800)of child's urinary iodine was less than 50μg/L.The rate of passing a knowledge questionnaire survey of the fifth-grade students and women was 92.33%(1 131/1225)and 9413%(397/421),respectively.Conclusions Ya'an city in Sichuan province has reached the goal of eliminating iodine deficiency.disorders,but there is a coexist trend of IDD and iodine excess.They should pay attention to the daily consumption of iodized salt in the future;sustained elimination of IDD can be reached only by iodine supplementation scientifically.
