Chinese Journal of Biochemical Pharmaceutics 2014;37(4):19-21
Objective To prepare transferrin and Arg-Gly-Asp polypeptide co-modified nanoparticles(TF/RGD-NPs)and evaluate its targeting efficiency to melanoma. Methods The co-modified nanoparticles were prepared by emulsion method and its appearance, particle size and Zeta potential were evaluated. The cellular uptake experiment and melanoma tumor spheroids penetration test were used to evaluate the affinity and ability to penetrate tumor tissues of TF/RGD-NPs to melanoma B16 cells. Results The particle diameter of co-modified nanoparticles was(113.4 ±12.5)nm and the Zeta potential was(4.53 ±2.15)mV.In vitro uptake test demonstrated that the efficacy of cellular uptaken TF/RGD-NPs by B16 cells were 2.7 times and 2.9 times to TF-NPs and RGD-NPs,respectively,the differences were all significant(P<0.05 ).Tumor spheroid penetration test results showed that TF/RGD-NPs has good affinity to melanoma cells. Conclusion TF/RGD-NPs can target to melanoma B16 cell efficiency in vitro,it may be serve as a potential drug delivery system for targeting melanoma.

Chinese Journal of Biochemical Pharmaceutics 2014;37(4):5-7,11
Objective To explore the effect of X-linked inhibitor of apoptosis protein(XIAP)combined with doxorubicin on the proliferation of MCF-7 cell and growth of MCF-7 bearing nude mice. Methods The anti-proliferation efficiency of doxorubicin and embelin were determined by MTT assay.MCF-7 cell apoptosis induced by embelin and doxorubicin were detected by Flow cytometry.Anti-tumor ability of embelin and doxorubicin were evalued with tumor spheroids test.MCF-7 cell were xenografted to mice to establish the animal model, which was used to evaluate the effect of anti-tumor. Results Compared with saline group, embelin group and doxorubicin group, the combination(doxorubicin +embelin)group inhibited the growth of MCF-7 cells effectively(P<0.01).The combination group induced the apoptosis of MCF-7 cells more effectively than doxorubicin alone(P<0.01), and significantly inhibit the growth of tumor in vitro and in vivo than other groups(P<0.01).Conclusion The combination of doxorubicin and embelin may be used as a potentially effective treatment method for breast cancer.

Objective To evaluate the effect of biofeedback therapy for the treatment of myogenic fecal incontinence. MethodsA general assessment about anorectal function was made on 17 cases receiving biofeedback therapy including muscle power training, sensory training and coordination training. Results The clinical scores before and after biofeedback therapy were 1.66?0.23,3.80?0.42 respectively, with an effective rate of 82%. The anus maximum contracting pressure elevated,(73?20) mm Hg vs. (123?30) mm Hg; myoelectric amplitude increased,(122?32) ?V vs. (230?41) ?V;Contracting time prolonged,(4.1?2.0) s vs. (9.4?3.0) s; The sensory threshold was lowered,(50?12) ml vs. ( 20? 10) ml;The feel-contract time increased,(3.1?0.4) s vs. (1.2?0.3) s. Positive rectal contraction reflex was seen in 41% patients before therapy compared with 82% after therapy. Conclusions Biofeedback therapy increases contractility of sphincter, decreases threshold of rectal sensory, and is a therapy of choice for myogenic fecal incontinence.

BACKGROUND:Previous studies have shown that nerve function may achieve different degrees of recovery in most patients after transvertebral transposition of the spinal cord for repair of spinal cord nerve compression and kyphoscoliosis. However, the effective mechanism of the proposed method to improve postoperative nerve function is stil not clear. OBJECTIVE:To investigate the improvement in neurological outcomes after transvertebral transposition of the spinal cord and posterior correction in patients suffering from neurologic deficit secondary to angular kyphoscoliosis. METHODS:Eighteen patients suffering from neurologic deficit secondary to angular kyphoscoliosis underwent transvertebral transposition of the spinal cord and posterior correction. Preoperatively and 1 week postoperatively, al patients were subjected to somatosensory evoked potential detection. During operation, motor evoked potentials and somatosensory evoked potentials were monitored. Using MRI, the distance from the inner edge of the spinal canal to the outer edge of the convex side of the spinal cord at apical region was measured, and the distance of spinal cord transposition was calculated. RESULTS AND CONCLUSION:(1) There were no significant differences in terms of the latency and amplitude of posterior tibial nerve P40 (preoperatively versus intraoperatively:amplitude:(1.66±0.29)μV vs. (1.68±0.28)μV, P>0.05;latency:(39.25±3.02) ms vs. (38.68±2.67) ms, P>0.05). (2) After surgery, the major curve was improved with (51.1±21.2)%correction rate, and the mean kyphosis was improved with (38.9±18.1)%. The distance between spinal cord and canal on the convex side at the apex was decreased by (2.4±1.7) mm on average. (3) The amplitude and latency of posterior tibial nerve P40 at 1 week postoperatively were (2.21±0.40)μV and (34.98±2.83) ms, respectively (P<0.05). (4) These results suggest that posterior correction and internal fixation can remarkably improve neurophysiological index and nerve function in patients with neurologic deficit secondary to angular kyphoscoliosis.

Objective To investigate the effect of intrathecal (IT) administration of L-NAME , a nonselective nitric oxide synthase (NOS) inhibitor, on c-fos expression in spinal cord induced by neuropathic pain. Methods Eighty-four female SD rats weighing 220-310g, which showed no signs of never injury 3 to 4 days after IT catheterization with the tip of catheter reaching the lumbar region, were divided into 4 groups at random: group A received IT 0.9% NaCl 10?g 15min before sham operation ( n = 16); group B received IT 0.9% NaCl 10?l 15min before ligation of sciatic nerve ( n =24); group C received IT L-NAME 10?l (250?g) 15min before sham operation ( n = 16); group D received IT L-NAME 10?l (250?g) 15min before ligation of sciatic nerve (n =24) . The animal in each group were further divided into 4 subgroups ( n = 4-6) and sacrificed on 1st, 3rd, 7th and 14th day after operation. In addition 4 rats which had received no operation of any kind were sacrificed and served as control. The c-fos expression in ipsilateral dorsal horn of spinal cord was detected by immunohistochemistry technique. Results In normal rats in control group (no IT catheterization, no operation) there were only a few fos-positive neurone (3.4? 0.9). The c-fos expression in the ipsilateral dorsal horn of spinal cord greatly increased both in group A and B. In group B there were more fos-positive neurons (FLIN) in the ipsilateral dorsal horn on the 3rd, 7th and 14th day as compare with group A. With pre-emptive intrathecal administration of L-NAME, the increase in ipsilateral c-fos expression induced by ligation of sciatic nerve was inhibited. In group D the number of fos-positive neurons in ipsilateral dorsal horn decreased by 53.8%, 57.1% and 43.2% on the 3rd, 7th and 14th day after operation as compare with group B. On the 1st day afteroperation there was no difference-in FLIN in the ipsilateral dorsal horn between group B and D. Conclusion Nitric oxide plays an important role in mediating the neuropathic pain which can be effectively attenuated or prevented by pre-emptive IT administration of L-NAME.
