1.Cellular transplant applied with rat bone marrow stromal cells preconditioned with stromal-derived factor 1 to treat acute myocardial infarction

Objective To investigate the effects of preconditioning (PC) with stromal-derived factor 1 alpha (SDF-1) on the levels of apoptosis of bone marrow stromal cells (BMSC) treated with hypoxia plus serum deprivation, and observe the therapeutic efficacy of cellular transplant with BMSC preconditioned with SDF-1 in rats with acute myocardial infarction. Methods BMSC were cultured with the whole marrow-adherence way. RT-PCR and immunohistochemistry were used to determine the expression of CXCR4. BMSC were incubated in medium for 24 h with 10 and 100 μg/L SDF-1 respectively, then treated with hypoxia plus serum deprivation for 6 h. The levels of apoptosis were detected by flow cytometry and TUNEL method. Acute myocardial infarction (AMI) model was established in SD rats, and BMSC preconditioned or non-preconditioned with SDF-1 were transplanted into border zone around infarct area, then heart function was measured after two weeks by ultrasonography. Results BMSC exhibited the CXCR4 expression. The number of apoptotic cells was significantly reduced in SDF-1 PC group than in control group (P<0.05), and 100μg/L SDF-1 PC group had the lowest level of apoptosis. AMI model was established successfully. Two weeks after BMSC transplant, significant improvement in cardiac function was observed in 100 μg/L SDF-1 PC group as compared with the non-PC group (P<0.05). Conclusions PC with the chemokine SDF-1 suppresses the apoptosis of BMSC treated with hypoxia plus serum deprivation. SDF-1 PC is a novel approach for enhancing therapeutic efficacy of cellular transplant in rats with AML

4.Effects of stromal-derived factor 1 preconditioning on apoptosis of rat bone mesenchymal stem cells.

The effects of stromal-derived factor 1 preconditioning (PC) on apoptosis of bone mesenchymal stem cells (BMSCs) treated with hypoxia plus serum deprivation were investigated. Bone mesenchymal stem cells were cultured with the whole marrow-adherence technique. RT-PCR and immunohistochemistry were used to detect the expression of CXCR4. BMSCs were incubated in medium for 24 h with 10 ng/mL and 100 ng/mL SDF-1 respectively, and then they were treated with hypoxia plus serum deprivation for 6 h. Apoptosis rate was determined by flow cytometry and TUNEL method. The results showed that BMSCs had CXCR4 expression. The number of apoptotic cells was significantly reduced in SDF-1 PC group as compared with the control group, and 100 ng/mL SDF-1 PC group had the lowest level of apoptosis. It was concluded that SDF-1 preconditioning suppresses the apoptosis of BMSCs treated with hypoxia plus serum deprivation.

Hydrogen sulfide is found to be the third gasotransmitter.It exerts extensive biological effects in nerve system,cardiovascular system,gastrointestinal system,and so on.In cardiovascular system,it exerts the function of vasorelaxation,inhibition of vascular smooth mu-scle cells and cardioprotection.It is also concerted with many cardiovascular diseases such as pulmonary hypertension,hypertension,coronary heart disease,atherosclerosis.This review discusses the latest research progress in the significance of hydrogen sulfide in cardiovascular diseases.
7.Clinical Significance of Measuring Exhaled Gaseous Signal Molecules

Exhaled gaseous signal molecules such as nitric oxide(NO),carbon monoxide(CO) and hydrogen sulfide(H2S) can be measured with a non-invasive technique.The fraction of NO level in exhaled gaseous of patients with airway inflammatory illness(such as bronchial asthma) significantly increases.No in exhaled gaseous is one of the markers in the diagnosis of asthma,and it is also a marker of anti-inflammatory response for patients with asthma.The fraction of CO level in exhaled gaseous of patients with bronchial asthma is also si-gnificantly elevated.Both NO and CO in exhaled gaseous in other airway inflammatory illness,such as chronic obstructive pulmonary disease,chronic bronchitis and cystic fibrosis are also evidently changed.Measuring exhaled gaseous signal molecules can provide important basis for the diagnosis and treatment of airway inflammatory illness.
8.Effect of Metoprolol Treatment in Vasovagal Syncope in Children

Objective To explore the effect of the ?-blocker —metoprolol on the treatment of vasovagal syncope(VVS) in children.Methods Twenty-nine children with unexplained syncope and positive responses to head-up tilt test(HUT) were included in the study.Sixteen of them took metoprolol(treatment group) and 13 of them took vitamin B or oryzanol (control group) at least 2 weeks and HUT were repeated and syncope episodes were observed.Results In treatment group,9 of 16 patients had no syncope episode while 5 of 16 patients had fewer syncope episodes,1 case had more syncope episodes,and 1 case remained the same.HUT were repeated and 6 of 9 cases had negative outcome.In control group,1 of 13 patients had no syncope episode while 5 of 13 patients had fewer syncope episode,3 cases had more syncope episodes and 4 cases remained the same.HUT were repeated and 3 of 7 cases had ne-(gative) outcome.Conclusion These results indicate that adminstering metoprolol orally may be effective for VVS in children.

10.Indoleamine 2,3-dioxygenase and forkhead boxP3 on immune tolerance of gallbladder carcinoma cells

Objective To investigate the expressions of indoleamine 2,3-dioxygenase (IDO) and transcription factor forkhead box P3(FOXP3)in gallbladder adenocarcinoma; and to evaluate the relationship between the expressions of IDO and FOXP3 protein,and clinicopathology and lymph node metastasis of gallbladder carcinoma.Methods The expressions of IDO and FOXP3 in 51 cases of primary gallbladder cancer,90 cases of chronic cholecystitis,and 20 cases of normal gallbladder tissues were studied by immunohistochemical staining. Results The positive expression rates of IDO and FOXP3 in gallbladder carcinoma were 72.5% (37/51) and 60.8% (31/51),in normal gallbladder mucosa were 5% (1/20) and 20.0% (4/20),in cholecystitis and gallstone were 11.1% (10/90) and 32.2% (29/90),respectively.There were significant differences among the three groups in IDO and FOXP3 expressions (P<0.01).The positive expression rates of IDO and FOXP3 were 7.1% and 14.3% for simple hyperplasia,17.7% and 35.3% for atypical hyperplasia,40% and 35% for gallbladder carcinoma (stages Ⅰ-Ⅲ),and 77.4% and 64.5% for gallbladder carcinoma (stages Ⅳ-Ⅴ ).There were significant differences among the four groups in IDO and FOXP3 expressions (P<0.01).There were significant differences among the Nevin stage groups,lymph node metastasis group and gallbladder stone in IDO and FOXP3 expressions.However,there were no significant differences among the sex groups and the age groups in IDO and FOXP3 expressions (P>0.05).In gallbladder carcinoma,the expression of IDO had a positive correlation with FOXP3 expression in lymphocyte (γ=0.406,P=0.003).Conclusion In gallbladder cancer,a high-expression of IDO and an expression of FOXP3 in lymphocyte are closely related with immune tolerance of gallbladder carcinoma.The results provide a reference for clinical use of immunotherapy in gallbladder cancer.
