1.Comparison between respiratory syncytial virus infected models of common mice and nude mice

Objective:To compare the infection of respiratory syncytial virus in common BALB/c mice and nude BALB/c mice to establish ideal animal model.Methods: At various time after RSV infection in BALB/c mice and nude mice,viral titers of lung tissue were assayed by plaque forming experiment,total leukocytes and differential subgroup cells in bronchoalveolar lavage fluid(BALF)were counted.RSV antigens were detected by immunofluorescent assay in BALF and immunohistochemistry in situ of lung tissue,lung histologys were identified by light microscope and electronical microscope assay.Results:RSV was isolated from lung tissue of BALB/c mice and nude mice.RSV antigen was detected by immunofluorescent assay in BALF and by immunohistochemistry in situ of lung tissue in both mouse,furthermore the mean light density of RSV antigen was higher in nude mice(0.277?0.085)than in BALB/c mice(0.128?0.031).The viral titers in both mouse peaked at the 3rd day post infection,and higher viral titer in nude mice than BALB/c mice.The viral titers detecting duration in lung tissue of BALB/c mice and nude mice were respectively 6 days and over 9 days after infection.The total leukocyte number in BALF significantly increased in infected BALB/c mice and nude mice compared with the control mice,and the peaks were the 6th day in BALB/c mice and the 9th day in nude mice.The leukocyte subgroups in turn were lymphocyte,macrophage,neutrophil and eosinophil.Interstitial inflammations with vast lymphocytes infiltrating were identified in lung histology of both mouse.On the 6th day,the more severe inflammation was detected than on the 3rd day.Meanwhile the more heavy inflammation was always found in nude mice than BALB/c mice.Conclusions:The similar RSV infectious characteristic exists between BALB/c mice and nude mice.But the more severe infection happens in nude mice,which benefits to assess the effect of anti-RSV drugs and is ideal mice model of RSV infection.
2.MRI Findings of Spinal Epidural Angiolipomas with Literature Review

Purpose To investigate the MRI findings and to improve the understanding of spinal epidural angiolipomas. Materials and Methods MRI findings were retrospectively analyzed in two cases of surgery and pathology proven spinal epidural angiolipomas. Relevant literatures were reviewed. Results Two cases of spinal epidural angiolipomas were located in the lumber and lower thoracic spinal canal. The tumors were elongated or spindle in shape within the epidural space, and parallel to the longitudinal axis. In both cases part of the tumors were hyperintense on T1WI and T2WI, attenuated on fat-saturated sequences with mild enhancement. Some areas were hypointenseon T1WI, hyperintenseon T2WI, unattenuated on fat-saturated sequences with apparent enhancement. Angiolipomas were classified according to the MRI features. One was type I and the other was type III. Both cases were non-infiltrative. Conclusion MRI shows the size, shape, MRI signals and the association with the adjacent structure of spinal epidural angiolipomas. The MRI classification is helpful for improving the understanding of the tumor. MRI is the best imaging modality to diagnose spinal epidural angiolipomas.
3.The clinical effects of pemetrexed or docetaxel combined with cisplatin as the first line treatment for advanced lung adenocarcinoma

Background and purpose:At present, the advanced lung adenocarcinoma is mainly treated by chemotherapy-oriented comprehensive therapy. Pemetrexed as a multi-target antifolate chemotherapeutic drug, combined with platinum, was approved for treatment of advanced lung adenocarcinoma because of its low toxicity and high efifcacy. The purpose of this study was to compare the short-term clinical efifcacy and toxicity of pemetrexed and docetaxel combined with cisplatin for advanced lung adenocarcinoma.Methods:Fifty patients with stageⅢ-Ⅳ lung adenocarcinoma were randomly divided into two groups. Group PC received pemetrexed (500 mg/m2, d1) combined with cisplatin (75 mg/m2, d1) and group TP received docetaxel (75mg/ m2, d1) combined with cisplatin (75 mg/m2, d1) treatment. One cycle was 21 days in both two groups. The response rate (RR), disease control rate (DCR), progression-free survival (PFS) and overall survival (OS) of the two groups were compared. The peripheral blood T lymphocyte subsets and natural killer (NK) cell activity of patients in both groups were measured before and after the treatment, while compared with healthy controls (25 cases).Results:Among 50 patients, no one got complete remission (CR), 20 patients got partial remission (PR), 17 patients had stable disease (SD) and 13 patients had progressive disease (PD).It showed no signiifcant difference in RR, DCR, PFS and OS (P>0.05) between two groups. Compared with group TP, the activity of CD3+, CD4+, CD4+/CD8+ T cells and NK cells increased and CD8+ T cells reduced in group PC after treatment, but the value had no obvious change (P>0.05). The common side effects were hematological toxicity and gastrointestinal response. The incidence of side effects in group PC was lower than that in group TP, and the degree was also slighter (P<0.05).Conclusion:The clinical efifcacy of pemetrexed combined with cisplatin is equal to docetaxel combined with cisplatin in treatment of advanced lung adenocarcinoma, but the former could reduce the adverse reaction rate.
4.Role of relaxin in heart disease and therapeutic potential

Following a brief introduction of relaxin family members, receptors and related signalling pathways,we review effects of relaxin on cardiovascular system,particularly,its anti-fibrotic action.The heart and vessels are target organs of relaxin.Recent studies have documented that the relaxin genes are upregulated in the diseased heart of humans and experimental animals.In several animal disease models,relaxin can reverse cardiac fibrosis.Relaxin may also indirectly promote myocardial regeneration and repair by suppressing fibrotic healing.Further research needs to focus on elucidating the exact mechanisms utilized by relaxin to induce these cardiac actions and translation of experimental findings to novel therapy of cardiovascular disease.
5.A simple way to improve fibroblast seeding efficiency and uniformity in constructing 3-D dermal equivalent

Objective:To explore a simple way to improve seeding efficiency and uniformity in constructing 3-D dermal equivalent,in order to effectively construct tissue engineered skin.Methods: Human dermal fibroblasts were added onto collagen sponge scaffolds and cultured in the following 3 ways:(1)oscillating seeding and oscillating culture(OO),(2)oscillating seeding and static culture(OS),and(3)static seeding and static culture (SS).Samples were obtained and subjected to light microscopy,scanning electron microscopy(SEM) and quantitative cell number assay at planed times.Results: Oscillating seeding achieved more uniform spatial cell distribution within scaffolds as compared to static seeding.Moreover,fibroblast seeding efficiency was significantly higher in oscillating conditions(\%) than in static conditions(\%,P
6.Effect of acceptance and implementation therapy on anxiety and depression of cancer patients' spouses

Objective To investigate the effect of acceptance and implementation therapy on anxiety and depression in cancer patients'spouse. Methods The spouses of 50 hospitalized patients from January to June 2014 were assigned into the control group and the spouses of another 50 cases of hospitalized patients from July to December 2014 into the observation group . All the patients received routine care methods. The control group received general health education and the spouses in the observation group were managed with acceptance and implementation therapy, totally for 3 times. After the intervention, the self-rating anxiety scale (SAS) and self-rating depression scale (SDS) were used to evaluate the levels of anxiety and depressions in both groups. Results Before intervention, there were no differences in SAS and SDS scores between the groups (P>0.05). After intervention, the SAS scores in the observation group and the control group scores were (48.36 ± 6.28) and (55.11 ± 6.74), respectively, with statistically significant difference (t=7.530, P<0.05). The SDS scores of the observation group and control group were (50.03 ± 4.12) and (58.50 ± 4.63) respectively, with statistically significant differences (t=6.672, P<0.05). The SAS and SDS score of observation group were lower than those before intervention (t = 7.283,7.524, all P < 0.05). Conclusion The acceptance and implementation therapy can effectively alleviate the anxiety and depression in the spouses of those patients with cancer.
7. The role of intracellular calcium in endothelin-1-induced proliferation of human lung adenocarcinoma cells SPC-A 1

Objective: To explore the role of intracellular calcium ([Ca2+]i) in endothelin-1 (ET-1)-induced proliferation of human lung adenocarcinoma cells SPC-A 1 and the underlying mechanism. Methods: The mRNA transcription of ET-1 and ET receptor (ETR) were detected by reverse transcription-polymerase chain reaction (RT-PCR) and ET-1 and ETR protein expressions in SPC-A 1 cells were measured by Western blotting. Cell proliferation was measured by (3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyl tetrazolium bromide) MTT assay. Intracellular Ca2+ concentration was determined by Fura-2/AM fluorescent assay. Results: ET-1 and ETAR and ETBR were expressed in SPC-A 1 cells at mRNA and protein levels. ET-1 (10-15-10-8 mol/L) stimulated the proliferation of SPC-A 1 cells in vitro and increased [Ca2+]i in a dose-dependent manner (P < 0.05). The effect of ET-1 (10-10 mol/L) on the proli-feration and [Ca2+]i of SPC-A 1 cells was reversed by a highly selective ET-AR antagonist BQ 123 (10-7 mol/L, P < 0.05), but could not be reversed by a highly selective ET-BR antagonist BQ 788 (10-7 mol/L, P > 0.05). Deletion of extracellular Ca2+ with edetic acid (EDTA, 0.4 mmoL/L) or blockade of voltage-gated calcium channels with nifedipine (1 μmol/L) significantly inhibites ET-1-induced elevation of [Ca2+]i and proliferation of SPC-A 1 cells. Conclusion: ET-1 stimulates cell proliferation and increased [Ca2+]i by activation of ET-AR in SPC-A 1 cells. Ca2+ influx via voltage-gated calcium channel is the main mechanism for [Ca2+]i elevation and cell proliferation.
8. Microarray analysis of the relationship between PTEN and prognostic factors in primary breast cancer tissues

Objective: To construct tissue microarray of breast carcinoma and to investigate the association of hepatocyte-specific phosphatase and tensin homolog (PTEN) protein expression with prognostic factors such as estrogen receptor (ER), progesterone receptor (PR), c-erb B-2, and Ki-67 in primary breast carcinoma. Methods: Tissue microarray of breast carcinoma was constructed. Immunohistochemical staining was used to detect the expression of PTEN, ER, PR, c-erb B-2, and Ki-67 inbreast carcinoma tissues. Results: PTEN was highly expressed in 66 out of 163 breast carcinoma tissues (40.5%). The positive rate of ER, PR, c-erb B-2, and Ki-67 was 51.5%, 35.6%, 34.4%, and 44.8%, respectively. The positive expression level of MN decreased with increase in auxiliary lymph nodes metastasis, histological grading, and TNM staging. They had negative correlations (P < 0.05). The positive expression level of PTEN had no evident correlation with tumor size and onset age. PTEN had positive correlation with the expressions of ER and PR protein (P < 0.01), and had negative correlation with the expressions of c-erb B -2 and Ki-67. Conclusion: There exists some relationships between low or loss of PTEN expression and the imbalanced expression of ER and PR and the growth and prognosis of breast carcinoma. PTEN may be considered as a potential indicator to judge the malignant degree of breast carcinoma. Combined detection of PTEN, ER, PR, c-erb B-2 and Ki-67 have significant importance and practical value for adjuvant therapy of breast carcinoma.
