2.Expression and clinical significance of survivin and p63 in diffuse large B-cell lymphoma

Objective To investigate the expression and significance of survivin and p63 in diffuse large B-cell lymphoma (DLBCL). Methods Immunohistochemical staining was used to detect the expression of survivin and p63 in the lymphnode tissues from DLBCL patients and lymph node reactive proliferation.Results The positive expression rate of p63 and survivin proteins in DLBCL patients was 63.5 %(33/52) and 76.9 %(40/52), respectively. p63 was not expressed in lymph node reative proliferation. The positive expression rate of survivin in lymph node reactive proliferation was 20.0 %. The expression rates of survivin and p63 between two groups was significant different. The replase rate of groups with positive expression of survivin and p63 were higher than that of the negative groups. The mean survival in groups with positive expression of survivin and p63 were shorter than that of the negative groups. Conclusion Survivin and p63 are useful in therapeutic and prognostic assessment. Patients with poor prognostic can be screened out in the early period of treatment using expression of survivin and p63, which may help to improve the outcome of diffuse large B-cell lymphoma.

Objective: To investigate the modulatory effect of gonadal hormone on peripheral pain. Methods: Orchiectomized and ovariectomized rats models were established. Radio heater and Von-Frey hair were used to determine the peripheral mechanical and thermal pain threshold in gonadectomized rats and their corresponding sham-controls. Results: The body weight was increased and the uterus weight was decreased after ovariectomy in female rats(P<0.01). There was no significant change in the body weight of mate rats after orchiectomization. The 100% hind paw withdrawal threshold to Von-Frey hair stimulation was decreased significantly after ovariectomization in female rats, with no significant change in the thermal pain threshold. There was no significant changes in the 100% hind paw withdrawal threshold after Von-Frey hair stimulation or thermal pain threshold after orchidectomy in male rats. Conclusion: These results suggest that changes in androgen level of male rats have no influence on the peripheral basal pain threshold, and female gonadal hormone may inhibit the peripheral pain signaling and has no effect on thermal pain threshold.

7.A Transposon-Based Genetic Screen in Saccharomyces cerevisiae Reveals a Role of YMR166C in Mitochondrial Magnesium Homeostasis

Magnesium homeostasis is very important to normal functions of cell and plays a role in a number of diseases such as diabetes, hypertension, chronic respiratory disorder, osteoporosis and arrhythmia. MRS2 encode a magnesium transporter protein in Saccharomyces cerevisiae and deletion of MRS2 gene results in decrease in mitochondrial magnesium concentration, group Ⅱ RNA splicing defect and growth defect on nonfermentable carbon source. To obtain more information of magnesium homeostasis in mitochondria, mTn-lacZ/LEU2 transposon library was transformed into mrs2 deletion mutant to screen for suppressor genes of MRS2. YMR166C, a member of mitochondrial carrier family, was identified as a suppressor gene of MRS2. Deletion of YMR166C gene can rescue the defects of mrs2 deletion mutant such as the decrease in mitochondrial magnesium concentration, groupⅡ RNA splicing defect and growth defect on nonfermentable carbon source. For the first time it was demonstrated that YMR166C is involved in mitochondrial magnesium homeostasis.
8.Microenvironment of primary liver cancer and regulatory T cells

Regulatory T cell (Treg) infiltration is an important mechanism for inhibition of tumor immune microenvironment. Treg is highly expressed in peripheral blood and tumor tissue of various cancer patients. A high expression of Treg may inhibit the antitumor effect of CD4 + CD8 + T lymphocytes or induce immunological resistance to tumor antigens among cytotoxic T lymphocytes(CTL) without producing acute or memory CTL responses, resulting in tumor escape from immuneattack and immune suppressionin in the tumor microenvironment.
9.Exploration of duration of intravenous analgesia pump after gynecological surgery

Objective To evaluate the influence of the duration of patient-controlled intravenous analgesia (PCIA)on pain and gastrointestinal function after gynecological abdominal surgery.Methods Patients who received gynecological abdominal surgery were divided into 3 groups according to using time of PCIA for 24 hours,48 hours,72 hours.There were 30 cases in each group.Pain intensity,gastrointestinal peristalsis time,anal exhaust time,getting out-of-bed time,voluntary micturition time,nausea and vomiting were compared among three groups after stopping PCIA.Results There were no significant differences in VAS score among three groups at each time point.There were no significant differences in voluntary micturition time among three groups.The anal exhaust time,getting out-of-bed time in 24-hour group were earlier than those of the other two groups,the differences were significant; The nausea and vomiting in 24-hour group were lower than the other two groups,the difference was significant.Conclusions The application of PCIA can effectively relieve acute pain 24 hours after surgery,but the effect was not obvious on pain induced by activities after 24 hours.But the side effects induced by analgesia pump(anesthetic)still work,such as vomiting,long duration of discharge,activity inconvenience.So PCIA should be stopped at right time in order to reduce complications and facilitate for recovery of patients after the surgery.
10.Radiofrequency chemohyperthermia combined with intravenous chemotheray for malig nant ascites

Purpose:To evaluate the efficacy and toleranc e of radiofrequency chemohyperthermia combined with intravenous chemotherapy for m alignant ascites. Methods:Twenty patients with malignant ascites received the tre atment .After abdominal cavity aspiration and infusion of hot 0.9% sodium chlori de injection 1 500-2 000 ml with 5-FU , Estimated the temperature of abdominal cavity was estimated with a sensor, then treated with SR-1000 radiofrequency h yperthermia for 70 minutes and chemotherapy at the same time.The radiofrequency chemohyperthermia with 5-FU 0.5-1.0 g was given d 1,4,8,11 and intravenous che motherapy with oxaliplatin 100 mg was given d 1,8 or cisplatin 40-60 mg d 1,8. Results:The average treatment temperature was 41.5℃ in 20 case s. The highest was 43.5℃ and lowest temperature was 40.5℃. The response rate o f ascites was 90%(18/20) The response rate of peritoneo-pelvic tumor was 58.8%( 10/17).1-year overall survival rate is 75%. The common side-effects were fat n ecrosis (20%) and adominal pain (25%). Conclusions:Radiofrequency chemohyperthermia combined with intr avenous chemotherapy appears to have a relatively high response with low side-e ffects for malignant ascites and good response for peritoneo-pelvic malignant t umors.
