2.Expression and clinical significance of survivin and p63 in diffuse large B-cell lymphoma

Objective To investigate the expression and significance of survivin and p63 in diffuse large B-cell lymphoma (DLBCL). Methods Immunohistochemical staining was used to detect the expression of survivin and p63 in the lymphnode tissues from DLBCL patients and lymph node reactive proliferation.Results The positive expression rate of p63 and survivin proteins in DLBCL patients was 63.5 %(33/52) and 76.9 %(40/52), respectively. p63 was not expressed in lymph node reative proliferation. The positive expression rate of survivin in lymph node reactive proliferation was 20.0 %. The expression rates of survivin and p63 between two groups was significant different. The replase rate of groups with positive expression of survivin and p63 were higher than that of the negative groups. The mean survival in groups with positive expression of survivin and p63 were shorter than that of the negative groups. Conclusion Survivin and p63 are useful in therapeutic and prognostic assessment. Patients with poor prognostic can be screened out in the early period of treatment using expression of survivin and p63, which may help to improve the outcome of diffuse large B-cell lymphoma.

Objective: To investigate the modulatory effect of gonadal hormone on peripheral pain. Methods: Orchiectomized and ovariectomized rats models were established. Results: The body weight was increased and the uterus weight was decreased after ovariectomy in female rats(P<0.01). There was no significant change in the body weight of mate rats after orchiectomization. The 100% hind paw withdrawal threshold to Von-Frey hair stimulation was decreased significantly after ovariectomization in female rats, with no significant change in the thermal pain threshold. There was no significant changes in the 100% hind paw withdrawal threshold after Von-Frey hair stimulation or thermal pain threshold after orchidectomy in male rats. Conclusion: These results suggest that changes in androgen level of male rats have no influence on the peripheral basal pain threshold, and female gonadal hormone may inhibit the peripheral pain signaling and has no effect on thermal pain threshold.
6.Microenvironment of primary liver cancer and regulatory T cells

Regulatory T cell (Treg) infiltration is an important mechanism for inhibition of tumor immune microenvironment. Treg is highly expressed in peripheral blood and tumor tissue of various cancer patients. A high expression of Treg may inhibit the antitumor effect of CD4 + CD8 + T lymphocytes or induce immunological resistance to tumor antigens among cytotoxic T lymphocytes(CTL) without producing acute or memory CTL responses, resulting in tumor escape from immuneattack and immune suppressionin in the tumor microenvironment.
7.Radiofrequency chemohyperthermia combined with intravenous chemotheray for malig nant ascites

Purpose:To evaluate the efficacy and toleranc e of radiofrequency chemohyperthermia combined with intravenous chemotherapy for m alignant ascites. Methods:Twenty patients with malignant ascites received the tre atment .After abdominal cavity aspiration and infusion of hot 0.9% sodium chlori de injection 1 500-2 000 ml with 5-FU , Estimated the temperature of abdominal cavity was estimated with a sensor, then treated with SR-1000 radiofrequency h yperthermia for 70 minutes and chemotherapy at the same time.The radiofrequency chemohyperthermia with 5-FU 0.5-1.0 g was given d 1,4,8,11 and intravenous che motherapy with oxaliplatin 100 mg was given d 1,8 or cisplatin 40-60 mg d 1,8. Results:The average treatment temperature was 41.5℃ in 20 case s. The highest was 43.5℃ and lowest temperature was 40.5℃. The response rate o f ascites was 90%(18/20) The response rate of peritoneo-pelvic tumor was 58.8%( 10/17).1-year overall survival rate is 75%. The common side-effects were fat n ecrosis (20%) and adominal pain (25%). Conclusions:Radiofrequency chemohyperthermia combined with intr avenous chemotherapy appears to have a relatively high response with low side-e ffects for malignant ascites and good response for peritoneo-pelvic malignant t umors.
8.Study of mechanism of radiation sensitizing effect of topotecan on nasopharyngeal carcinoma

Purpose:To study the radiosensitising mechanism of topotecan in cell and molecule level on Nasopharyngeal carcinoma.Methods:To detect cell cycle,apoptosis index and the expression s of apoptosis genes such as p53, bcl2, bax by FCM.Results:The proliferation index of RT20Gy +TPT 12.5mg/kg group was decreased, while the apoptosis index was increased, with statistical signifi cance by comparison with TPT and RT group. Meanwhile, the expressions of apoptos is genes, such as p53,bcl-2,bax had no statistic significance between each grou p and the control group. Conclusions:The mechanisms of radiosensitizion might be its act ion on cell cycle,proliferation index and stimulation of apoptosis.However,the i nduction apoptosis might be independent of P53,bcl-2 and bax genes.
9.Detection and clinical significance of G-CSF receptor

Objective: To investigate a method for G-CSFR and analyze its expression in acute leukemia. Methods:20 normal controls and acute leukemic patients were detected by G-CSF receptor antibody and flow cytometry, analyzed relationship between the level of G-CSFR expression and clinical manifestations. Results: G-CSFR expression rates of patients with acute leukemia were significantly lower than that of normal control(P0.05),while the G-CSFR expression rate were significant correlated with blast rate of bone marrow(r. =-0.614, P = 0.025) ;there were no correlation with G-CSFR expression and FAR subtype in acute myeloid leukemia. Conclusion :The method for detecting G-CSFR is very sensitive and differential ,acute leukemic patients have lower expression rate of G-CSFR.
