5.Relationship between microRNA-200c and ZEB1 of epithelial mesenchymal transition factor in cervical cancer

Objective To investigate relationship between microRNA-200c and ZEB1 in cervical epithelial mesenchymal transition factor. Methods 50 cases diagnosed with cervical cancer whose tumor tissue were taken to be the experiment group.50 cases diagnosed with uterine myoma whose normal tissue were taken to be the control group.ZEB1 and microRNA-200c of uterine tissue were were detected.Results The average level of ZEB1 in the experimental group was significantly higher than that of the control group (P<0.05), and in cervical cancer, lymph node metastasis and the depth of myometrial invasion had statistically significant (P<0.05).In experimental group, microRNA-200c average was lower than control group (P<0.05), and in cervical cancer, different pathological grade group, lymph node metastasis and deep muscular layer infiltration had statistically significant(P<0.05).In experimental group, ZEB1 and microRNA-200c showed negative correlation(r =-0.270 P =0.001).Conclusion The expression level of ZEB1 in cervical cancer is higher, and the expression level of microRNA-200c in cervical cancer is lower, suggesting that the two factors may play an important role in the invasion and metastasis of cervical cancer.The correlation between the two factors suggests that microRNA-200c may be involved in the process of cervical cancer by ZEB1.
6.Efficacy and Safety of Sodium Valproate versus Diazepam in the Treatment of Status Epilepticus in Adults:a Meta-analysis

OBJECTIVE:To systematically evaluate the efficacy and safety of sodium valproate versus diazepam in the treat-ment of status epilepticus in adults,and to provide evidence-based reference for clinic treatment. METHODS:Retrieved from Med-line,EMBase,PubMed,Cochrane Library,CJFD,VIP datebase,CBM and Wanfang database,the randomized controlled trials (RCT) about sodium valproate (test group) versus diazepam (control group) in the treatment of status epilepticus in adults were collected. Meta-analysis was performed by using Rev Man 5.0 statistical software after extracting data and evaluating quality. RE-SULTS:A total of 5 RCTs were included,involving 276 patients. Results of Meta-analysis showed that recurrence rate of epilepsy in 48 h in test group was significantly lower than that of control group [OR=0.39,95%CI(0.16,0.96),P=0.04]. Compared with the control group,the effective rate controlling epilepsy within 2 h[OR=1.76,95%CI(0.88,3.52),P=0.11] and incidence of liver injury[RR=1.19,95%CI(0.38,3.70),P=0.77] were no significant differences. CONCLUSIONS:The efficacy and safety of sodi-um valproate was equivalent to diazepam in the treatment of status epilepticus in adults,however,sodium valproate was superior to diazepam in terms of controlling recurrent effect. Due to methodology limit of included studies,large-scale and high quality RCT are required for further validation of the conclusions.
7.Analysis of Prognosis of Children with Non-Hodgkin′s Lymphoma

Objective To analyze the factors affecting prognosis of children with non-Hodgkin′s lymphomas(NHL).Methods Thirty patients of childhood NHL histologically proven in Shanghai children′s medical center from Jan.1996 to Dec.2000 were analyzed.The patients were divided into groups according to some clinical factors that may have predictive significance including age,gender,immunologic phenotype,general system symptom,the size of tumor bulk,bone marrow involvement,mediastinal involvement,hepatomegaly or liver involvement,splenomegaly or spleen involvement and early response to chemotherapy,then the difference of survival rate were compared between groups.Survival analysis was performed by the Kaplan-Meier method,and difference between groups were campared using the Log-rank test.Results The 3-year survival rate was better in patients without giant tumor bulk than that with giant tumor bulk(P0.05).Conclusions Bone marrow involvement and the size of tumor bulk have very significant influence on prognosis of childhood NHL,and splenomegaly or spleen involvement have significant influence on disease-free survival.It can′t be proven in childhood NHL in this study.
9.EFFECTS OF WEHI-3 SUPERNATANT ON GROWTH AND PROLIFERATION OF MURINE BONE MARROW STROMAL CELLS IN VITRO

In order to observe the effects of the supernatant of WEHI-3 cell culture which contains IL-3 (interleukin 3), we have Performed some experiments by adding WEHI-3 supernatant to murine bone marrow cells culture. Cell culture of six days shows that the supernatant of WEHI-3 can promote adherence and proliferation of the stromal cells. There are 1.334?10~6 stromal cells/L in the culture which contained 20% WEHI-3 supernatant. Four kinds of stromal cells can be identified under both light microscope and SEM. Fibroblast-like cells which tend to be spindle-shaped with many filament-like and microvilli-like dendrites on the cell surface; Riticular-like cells which are irregular with some folds on the dendrites surface occasionally; Macrophage-like cells which are round with many phagocytic granules in the cytoplasm, and are characterized by positive ANAE staining, and many lamina-like folds and long filament-like dendrites on the cell sufrace; Fat cells are few in number, the cell body is ovoid with lipid droplet, and show strong cytoplasmic staining for Sudan black B, the cell surface is smooth. At the same time we have also observed the close contact between the hematopoietic cells and the stromal cells. In the culture without WEHI-3 supernatant, we have found few small adhered cells with less cytoplasm and weak enzyme activaties under light microscope, and with atrophied cell body and flat surface under SEM. The cell count of the stromal cells in normal culture is 58.83/mm~2. The results showed that WEHI-3 supernatant can promote the growth and proliferation of the stromal cells of murine bone marrow with some histochemical changes.
