1.High-performance porous beta-tricalcium phosphate bone tissue engineering scaffolds using 3D printing

BACKGROUND:Although the preparation of bone tissue engineering scaffolds can achieve satisfactory results by solvent casting/particulate leaching, in situ molding method, electrospinning, phase seperation/freeze drying, gas foaming, there are stil some deficiencies in the accuracy, pore uniformity, spatial structure complexity, personalized stents. <br> OBJECTIVE:To prepareβ-tricalcium phosphate bone tissue engineering scaffolds using 3D printing. <br> METHODS:Drug-loadedβ-tricalcium phosphate scaffolds were prepared with 3D printing, and the structure was observed to measure its porosity and mechanical strength. The scaffold was immersed in simulated body fluid for 15 weeks to observe the quality change. The scaffold was co-cultured with rat bone marrow mesenchymal stem cells for 7 days to observe celladhesion and morphological changes. Rat bone marrow mesenchymal stem cells were cultured in extracts of drug-loadedβ-tricalcium phosphate scaffold and low-glucose Dulbecco's modified Eagle’s medium containing 15%fetal bovine serum for 24, 48, and 72 hours, to determine the absorbance values and cytotoxicity grading, respectively. Meanwhile, the cells were subjected to osteogenic culture for 1 week, and <br> the alkaline phosphatase activities in two groups were detected. <br> RESULTS AND CONCLUSION:The prepared scaffold showed irregular micropores, high porosity, uniform pore distribution, high pore connectivity rate, and large compressive strength. The drug-loadedβ-tricalcium phosphate scaffold degraded completely with 15 weeks, and cancellous bone defect repair was completed in the same period. Rat bone marrow mesenchymal stem cells adhered to the surface of drug-loadedβ-tricalcium phosphate scaffold and went deep into the scaffold, showing good growth and proliferation. The activity of alkaline phosphatase was also improved. These findings indicate that the drug-loadedβ-tricalcium phosphate scaffold has good biocompatibility.
2.Clinical characteristics and treatment of primary ovarian malignant lymphoma

Objective:To investigate the clinical features and prognostic factors of primary ovarian lymphoma(POL). Methods:A retrospective review was performed based on the clinical records of 14 POL cases treated at Tianjin Medical University Oncology in-stitute Hospital verity from 2000-6 to 2010-5. Results:The median age of patients was 47 years at presentation(range 28~62years). Ab-dominal pain was the most common initial symptom. The majority of histological subtype was B cell lymphoma, above 50%of which is diffuse large B-cell non-Hodgkin's lymphoma. A R0 resection was carried out in 2 patients who were lost to follow-up soon after sur-gery. 11 patients were treated by CHOP, FC or CHOPE chemotherapy. Chemotherapy combined with rituximab were given to 5 pa-tients. Until now, 5 patients were alive, 6 patients died, and only one patient was lost to follow up after therapy. Conclusion:POL is an extremely rare lymphoma. The ultimate diagnosis depends on histopathologic examination. Primary ovarian non-Hodgkin lymphoma should be treated with multi-modality strategies. Treatment with doxorubicin-based chemotherapy after oophorotomy is recommended. B cell lymphoma preferred to use rituximab combination chemotherapy. Abdominal cavity chemotherapy prophylaxis and irradiation can decrease the probability for recurrence .
3.Value of NLR,PLR and their changes in predicting occurrence of anastomotic leakage after surgery in patients with rectal cancer

Objective:To explore the value of preoperative neutrophil-lymphocyte ratio (NLR),preoperative platelet-lymphocyte ratio (PLR),changes between pre and postoperative neutrophil-lymphocyte ratio(postoperative NLR/preoperative NLR) and changes between pre and postoperative platelet-lymphocyte ratio(postoperative PLR/preoperative PLR) in predicting the occurrence of anastomotic leakage after surgery in patients of rectal cancer.Methods: 187 rectal cancer patients in the Gastrointestinal Surgery Department in the First Affiliated Hospital of Southwestern Medical University during March,2015-March,2016 were included in our study.Preoperative NLR,preoperative PLR,changes between pre and postoperative NLR and changes between pre and postoperative PLR were observed.At the same time we included age,gender,pathological staging,stage of lymph node metastasis,tumor differentiation,distance between tumor and anus in our study.χ2 test or Fisher′s exact value was used in the univariate analysis,while Logistic Regression Analysis was used in the multivariate analysis.Results: The univariate analysis showed that preoperative NLR (Fisher′s exact value=7.242),preoperative PLR (χ2=6.787),changes between pre and postoperative NLR (χ2=15.656),PLR (χ2=9.298),age (χ2=4.813) and distance between tumor and anus(χ2=5.951) were related to anastomotic leakage(P<0.05).Other risk factors such as,gender(χ2=0.001),pathological staging (Fisher′s exact value =2.107),stage of lymph node metastasis(Fisher′s exact value =1.298),tumor differentiation (Fisher′s exact value =2.206),and were not associated with anastomotic leakage (P>0.05).Multivariate analysis showed that the preoperative NLR(OR=1.647,P=0.000),change between pre and postoperative NLR (OR=1.880,P=0.000)and distance between tumor and anus(OR=4.364,P=0.048) were associated with anastomotic leakage.Conclusion: preoperative NLR,change between pre and postoperative NLR and distance between tumor and anus are independent risk factors for anastomotic leakage after rectal anterior resection.
4.Effect of bortezomib combined with doxorubicin on lymphoblastic lymphoma Molt-4 cells

Objective To investigate the effect of bortezomib combined with doxorubicin on lymphoblastic lymphoma cell line Molt-4.Methods Molt-4 cells were cultured in the presence of bortezomib and doxorubicin,cell viability was monitored by CCK-8 and trypan-blue exclusion.Apoptosis was detected by flow cytometry and mitochondrial membrane potential,expression of Fas was also measured with flow cytometry.Results Molt-4 cell proliferation was substantially inhibited in concentration-dependent manners when treated with either bortezomib or doxorubicin.The combination of both drugs synergistically inhibited Molt-4 cell proliferation at 48 hours [(57.24±0.10) %].Combination therapy further enhanced bortezomib and doxorubicin induced apoptosis [48 h (23.08±1.25) %] (P < 0.05).Detection of mitochondrial membrane potential showed that combination therapy could promote apoptosis (15.84 %,5.38 %,5.52 %) but did not significantly change the level of Fas expression (P > 0.05).Conclusion Combination therapy of bortezomib with doxorubicin efficiently inhibits proliferation and induces apoptosis of Molt-4 cells.Activation of mitochondrial and intrinsic apoptotic pathway may play important roles.
5.Efficacy of combined pranoprofen eye drops and artificial tears on the treatment of mild to moderate dry eye syndrome after trabbeculectomy

AlM:To evaluate the efficacy of combined pranoprofen eye drops and artificial tears on the treatment of mild to moderate dry eye syndrome after trabbeculectomy.METHODS:This prospective case control study included 63 cases (63 eyes) of patients with mild to moderate dry eye syndrome after trabbeculectomy in our hospital from November 2013 to June 2013. All subjects were randomly divided into two groups. Observation group was treated with combined pranoprofen eye drops and artificial tears and control group received simple artificial tears marking the eyes at 1, 2, 4wk. The patient's symptoms, signs, BUT, S▏t, and FL were observed before treatment and 1, 2, 4wk after treatment.RESULTS: After 2wk, the symptoms of observation group were improved, there was statistically significant difference (P<0. 05). FL difference of each group was statistically significant ( P<0. 05 ); After 4wk, symptoms and signs were improved. There was statistically significant difference ( P < 0. 05 ). The BUT of the observation group and corneal FL scores of two groups showed significant differences (P<0. 05). CONCLUSlON: Artificial tears joint pranoprofen eye drops has good curative effect in the treatment of mild to moderate dry eye syndrome after trabbeculectomy.
8.Saccharide mapping and its application in quality control of polysaccharides from Chinese medicines.

Polysaccharides with multiple biological activities are usually considered as one of the major bioactive compounds in Chinese medicines (CMs). At present, the development of drug and functional foods related to polysaccharides have attracted a great deal of attention due to their great potential effects and diverse action mechanisms. However, quality control of polysaccharides is the bottleneck and a challenge due to their complexity and chemical diversity. Actually, the bioactivities of polysaccharides are closely related to their molecular structures. In order to ensure their safety and efficacy, the development of novel approaches based on the molecular structures for the improvement of quality control of polysaccharides is significantly important. Therefore, in this article, the relationship between biological activities and chemical structures, as well as the action mechanisms of polysaccharides from CMs were summarized first. Furthermore, saccharide mapping, a novel strategy for quality control of bioactive polysaccharides from CMs, was introduced and the application and perspectives were also discussed.

9.Research advances in poly (ADP-ribose) polymerase inhibitors in combination with radiotherapy

As a DNA damage sensor,poly (ADP-ribose) polymerase (PARP) is involved in a wide variety of cellular activities,such as DNA damage repair.PARP inhibitors regulate a series of cellular activities by inhibiting PARP function,which have become a focus of current research.Recently,several in vivo and in vitro studies showed that PARP inhibitors combined with radiotherapy effectively enhanced the efficacy of radiotherapy.This paper reviews the research advances in the mechanisms of action of PARP inhibitors and their combination with radiotherapy.
10.Questionnaire Survey for Researchers of Medicine Clinical Trial in Guang’anmen Hospital of China Academy of Chinese Medical Sciences

ObjectiveTo understand the status and requirement of the clinical researchers in Guang’anmen Hospital of China Academy of Chinese Medical Sciences (hereinafter referred to as Guang’anmen Hospital).MethodsSelf-filling questionnaire was used. All medical care personnel in the clinical specialty with the GCP qualification were set as respondents. The three aspects of recognition degree of clinical trials quality, initiative of participation and knowledge demand for GCP were investigated. Data were analyzed through SPSS11.5.Results 95.6% of clinical researchers considered that the course of clinical trial was legal;89.9% of clinical researchers believed that the quality of clinical trial was good. Most clinical researchers participated in the clinical trials actively (86.9%) and they considered that the trials were helpful for them to enhance research ability and to issue paper.Conclusion Clinical researchers are familiar with GCP knowledge and can participate in the clinical trials actively. They still expect to learn more.
