Aiming at the issue of motor imagery electroencephalography (EEG) pattern recognition in the research of brain-computer interface (BCI), a power feature method based on discrete wavelet packet decomposition is proposed for the channels C3 and C4. Firstly, a six-border Butterworth filter is used to denoise the two-channel EEG signals. Secondly, two-channel EEG signals are decomposed to five levels using Daubechies wavelet and the fourth level and the fifth level are chosen to reconstruct the signals and compute its power feature. Finally, linear discriminant analysis (LDA) is utilized to classify the feature and the Kappa value is utilized to measure the accuracy of the classifier. This method is applied to the standard dataset BCICIV_2b-gdf of BCI Competition 2008, and experimental results show that this method reflect the feature of event-related synchronization and event-related desynchronization obviously and it is an effective way to classify the EEG patterns in the research of BCI.

Objective To construct a plasmid DNA encoding G glycoprotein of respiratory syncytial virus(RSV) and investigate the protective immune response against RSV infection. Methods Recombinant plasmid DNA of pcDNA3.1~G was constructed by standard RT-PCR based cloning procedure. The immunogenicity of recombinant G protein transiently expressed in HEK293 cells was detected by Western blot. BABL/c mice were intramuscularly immunized with pcDNA3.1~G. Samples of lung, sera, bronchoalveolar lavage fluid(BALF) were collected before and after RSV challenge; virus titer in lung was detected by viral titration; sections of paraffin embedding lung tissues were stained by haematoxylin and eosin(HE) for histological analyses; sera anti-RSV IgG levels were examined by ELISA; Th1/Th2 cytokine were detected by ELISA kit, the T lymphocyte subsets of BALF was determined by immunefluorescence staining followed by flow cytometry. Results Plasmid DNA of pcDNA3.1~G was successfully constructed. The expressed target protein possesses immunogenicity. After challenge, pcDNA3.1~G immunized mice presented relieved pathological changes in lung as well as reduced lung viral titers. The RSV specific IgG was detected in sera of immunized mice. There was significantly increased number of CD25~+CD4~+ T cells in mice BALF. Conclusion We constructed a pcDNA3.1~G plasmid DNA vaccination which can induce evident protective cellular immunity against RSV infection in mice with the increased number of CD25~+CD4~+ T cell subpopulation.

Methods The detailed nurse plan and carefully observation and assess of lesion were performed daily. Iodophor water at the concentration of 0.5‰ was used for anal bath and 2% iodine ointment was used to smear perianal to prevent of infection. And gauzes soaked iodophor were used to cover the lesions, and Chinese traditional herb for anal bath and ultraviolet exposure to control infection. Additionally, the necessary psychological support was given to overcome the fear. Results After the careful nursing, the patient passed through the agranulocytosis period and achieved the success of transplantation. Conclusions This case suggests that the methods in prophylaxis of patients with circumferential mixed hemorrhoids is effectious.

AIM: To investigate a new method for identification of traditional Chinese medicine injection by FTIR. METHODS: For the steadiness of spectra, the factors of effecting spectrum's information quality were all investigated scientifically over the experiment procedures and instrumental setting, such as the preparation of samples, resolution ratio, scanning times, repeating scanning times, etc. The traditional Chinese medicine injections were used as the analytical samples such as Radix Isatidis, Rhizoma Chuanxiong, Flos Carthami, Radix Astragali and Herba Houttuyniae. RESULTS: Although all these original spectrums were similar at a certain degree, the FTIR combined with computer aided analysis, such as the cluster analysis and derivative spectrometry comparability calculation could be used to identify these injections. CONCLUSION: The method of identification by FTIR is non destructive testing, cheap, clean, fast, simple and convenient. The result indicates this method is suitable for establishing identification database of traditional Chinese medicine injections.

[ ABSTRACT] AIM:To study the protective effect of Astragalus polysaccharides ( APS) on free fatty acid-induced injury in human umbilical vein endothelial cells (HUVECs).METHODS: Cultured HUVECs were divided into control group, APS group [ APS (200 mg/L) treated for 24 h], free fatty acid group [free fatty acid (0.25 mmol/L) treated for 24 h], free fatty acid plus APS group [free fatty acid (0.25 mmol/L) and APS (200 mg/L) treated for 24 h], and com-pound C group [ free fatty acid (0.25 mmol/L) and APS (200 mg/L) and AMPK inhibitor compound C (10 μmol/L) treated for 24 h] .The cell viability was detected by MTT assay.Nitric oxide ( NO) content in the medium was determined by nitrate reductase assay. The protein levels of total adenosine monophosphate-activated protein kinase (AMPK), phos-phorylated adenosine monophosphate-activated protein kinase (p-AMPK), endothelial nitric oxide synthase (eNOS) and phosphorylated endothelial nitric oxide synthase ( p-eNOS) were measured by Western blot.RESULTS: No significant difference of all indexes between APS group and control group was observed. The cell viability in free fatty acid group de-creased significantly compared with control group. The cell viability in free fatty acid plus APS group was significantly im-proved as compared with free fatty acid group. The cell viability in compound C group was almost the same as that in free fatty acid group. The No content and protein levels of p-AMPK and p-eNOS in free fatty acid group decreased obviously as compared with control group, while the NO content and protein levels of p-AMPK and p-eNOS in free fatty acid plus APS group increased obviously compared with free fatty acid group. No significant difference of the p-AMPK and p-eNOS protein levels between free fatty acid plus APS group and free fatty acid group was observed. No significant difference of the AMPK and eNOS protein levels in all groups was found.CONCLUSION:APS attenuates the free fatty acid-induced injury, and its mechanism is related to the AMPK-eNOS signal pathway.

Objective To investigate the effect of astragalus polysaccharides (APS) on the growth and proliferation of hepatocellular carcinoma cells and its effect on adenosine monophosphate activated pro-tein kinase (AMPK) activity. Methods Hepatocellular carcinoma HepG2 cells cultured for 12,24,and 48 hours were treated with 200,300,and 400 mg/L concentration of astragalus polysaccharides. The cell inhibition rate was detected with methyl thiazolyl tetrazolium (MTT),and apoptosis was observed under the fluorescence microscope.Western blot method was used to measure the expression of total AMPK,phosphorylated AMPK (p-AMPK),and phosphorylate mammalian target of rapamycin (p-mTOR) protein expressions. Results Astragalus polysaccharides of each concentration significantly inhibited the proliferation of human hepatoma HepG2 cells (P < 0.01),and the effect of 300 mg/L concentration astragalus polysaccharides was more significant than that of the 200 mg/L concentration (P <0.01);while inhibitory effect of 400 and 300 mg/L Astragalus polysaccharides on the proliferation of human hepatoma cell line HepG2 was not significant difference. We found that Astragalus polysaccharides of each concentration could promote the apoptosis of HepG2 cells, and the effect of 300 mg/L Astragalus polysaccharides was more significant. However, astragalus polysaccharide of 400 mg/L concentration could promote the apoptosis no more than the 300 mg/L concentration, which was observed by fluorescent microscope.Western blot results showed that astragalus polysaccharides could increase the expression of p-AMPK (P < 0.05),and inhibit its downstream protein expressions of p-mTOR (P < 0.05).The proliferation effect of astragalus polysaccharides was weakened after accession of AMPK antagonist compound C on hepatocellular carcinoma cells. Conclusions APS can inhibit the growth and proliferation of hepatocarcinoma cells, and its mechanism is related to the AMPK-mTOR pathway.
