Objective: To investigate the feasibility for reconstructing an aortic arch by a new integrated two-branched stent graft releasing in experimental canine thoracic aorta simulation system.
Methods: A new type of integrated two-branched stent graft was developed and placed in the canine thoracic aorta, the stent graft releasing in a simulated system was conducted under X-ray guidance to monitor the operating process and to explore the feasibility for invitro sent graft releasing.
Results: The new two-branched stent graft was successfully deployed in canine thoracic aorta simulation system. The releasing process was smooth, two small stent grafts were well landing, and all stent grafts were fully expanded and properly positioned.
Conclusion: Our new two-branched stent graft could successfully reconstruct the aortic arch in experimental canine. The thoracic aorta releasing system may better and truly simulate the whole process of endovascular aortic repairing, which provides a good foundation for further animal experiments.

Objective To investigate the incidience of anxiety or depression in the families of chronic diseases and to discuss the relationship between anxiety or depression and coping style. Methods Using SAS, SDS,SCSQ as main tools,questionnaire investigation was made in 232 families of chronic diseases. Results The incidence of anxiety was 25.9%, and that of depression was 43.1%, and the incidence of concurrence was 25.9%. The mental health level was asscociated with the sex, active response of families, the age, disease course of the patients, hospitalization costs, economic conditions. Conclusions The families of chronic diseases patients have different degrees of negative mood, which affects the patients. Therefore, enchancing the mental intervation of families of chronic diseases will do good to the recovery of diseases and improvement of life quality.

Progression of tumor is a complex process with multiple steps and multiple factors.MicroRNA-296 (miR-296) plays an important role in tumor progression,involved in the proliferation,differentiation,angiogenesis,invasion,metastasis,apoptosis and drug resistance of the tumor cells.

Objective:To investigate the therapeutic efficacy of immunotherapy with dendritic cells-based vaccine against intracranial gliomas in rats.Methods:C6 glioma cells were injected into the brain of Wistar rat under stereotactic monitor to establish an animal model of glioma.The precursors of dendritic cells were isolated from bone marrow of rats,stimulated in vitro with recombinant rat granulocyte-macrophage colony-stimulating factor(rrGM-CSF)and interleukin-4(rrIL-4).Cultured cell populations were confirmed to be functional DCs.These DCs were then pulsed ex vivo with C6 tumor-lysates prepared by three cycles of freezing to -80℃ and thawing to 0℃ and subsequently injected subcutaneously into rats harboring intracranial C6 tumor.Rats from different group were treated with five weekly subcutaneous injections of either control media,unpulsed DCs,or DCs pulsed with tumor-lysates.The animals were followed for survival,the percentage of CD8 +T cells in peripheral blood and cytotoxicity assay in vitro were determined by FACS.The level of cytokine IFN-? and IL-10 were detected by ELISA.Results:The results indicated that C6 glioma model rats treated with tumor-lysate-pulsed DCs led to prolonged survival time,increased the percentage of CD8 + T lymphocyte in peripheral blood in comparing with control group.Cytotoxicity assays suggest that vaccination with these tumor-lysate-pulsed DCs can induce specific cytotoxic T lymphocytes against C6 tumor cells compared with control group.Furthermore,significantly enhanced IFN-? and reduced IL-10 (even undetectable)were observed in rats treated with pulsed-DCs.Conclusion:Data supported the therapeutic efficacy of systemic vaccination with DCs pulsed with tumor-lysates against intracranial glioma.

Objective To evaluate the performance of an improved skin prick test in the screening for allergens.Methods A total of 475 patients with chronic urticaria who aged from 3 to 81 years were enrolled in this study,and classified into the control group (n =235) and research group (n =240).Traditional and improved skin prick test were conducted in the control and research group respectively.The allergen detection rate was compared between the two test methods.Results The allergen detection rate was 65.4％ and 67.2％ respectively for the improved and traditional skin prick test,respectively (P ＞ 0.05).House dust mites were the most common sensitizing agent.Conclusion The improved skin prick test can offer reliable evidence for chnical diagnosis with a relatively convenient and safe procedure.

The fused gene (PTH-HSA) of parathyroid hormone (PTH) gene and Human Serum Albumin(HSA) gene was amplified without linker by Overlapping PCR technology. The spliced gene was clone into Pichia pastoris secretory vector pPIC9K. With the help of promoter AOX1 and mat ? signal peptide, the PTH-HSA gene was designed to secretory expression.Linearized by restriction enzyme SalI, The recombinant plasmid pPIC9K/PTH-HSA was transformed into Pichia pastoris KM71 by electroporation. The recombinant strains which were identified by G418 and PCR analysis were induced by methanol to express protein PTH-HSA. The target protein was expressed in fermentation supernatant. Western blot analysis of the fusion protein showed that the expressed fusion protein PTH-HSA had the antigenicity of HSA.adenylate cyclase assay proved that the fused protein exhibited the bioactivity to stimulate cAMP synthesis The specific activity of broth was about 318IU/ml.

Objective To investigate the diagnostic and therapeutic characteristics of prostate cancer with prostate specific antigen(PSA)4-10?g/L. Methods The data of prostate biopsies for the patients with PSA 4-10?g/L from May 1998 to February 2004 and the treatment and prognosis of these patients were retrospectively analyzed. Results In the 141 cases, 34 were diagnosed as prostate cancer, including 3 cT1, 21 cT2, 6 cT3 and 4 cT4. Mean Gleason score was 5.8, and the mean Gleason scores of cT1 and cT2 were significantly lower than those of cT3 and cT4. Twenty-four cases of cT1 and cT2 underwent radical prostatectomy. Four cases of cT3 recieved radical prostateetomy after 4 months of neoadjuvant endocrine therapy. Two cases of cT3 and 4 cT4 received castration. Pathological section from radical surgeries showed that 21 cases were organ confined and 7 were locally invasive. The difference of mean Gleason score between these two groups was significant in statistics. After 18 to 69 months (mean 42 months)follow-up, 21 organ confined cases were free survived, 4 locally invasive cases had biochemical recurrence and 1 case had multifocal bone metastasis. No cancer-specific death happened. Two of 4 cT4 cases were still alive and 2 died of the tumor. Conclusions Routine needle biopsy is necessary for the cases with“grave area”PSA of 4～10 ?g/L. Prostate cancers with PSA 4～10/?g/L are not always early diseases. Gleason score is a very important index for determine the stage of pathology after surgery. Radical prostatectomy is an effective treatment for organ confined disease.

Objective: To investigate the molecular mechanism of inhibitory effect of Scutellaria baicalensis to uropathogenic Escherichia coli (UPEC). Methods: RNA-seq was used to analyze transcriptome of UPEC NB8 effected by water decoction of S. baicalensis. S. baicalensis group and negative control group were set up. In S. baicalensis group, UPEC NB8 was disposed by water decoction of 10 times of MIC of S. baicalensis (62.5 mg/mL) for 30 min. While in negative control group, UPEC NB8 was disposed by same amount of normal saline. Then, total RNA of UPEC NB8 was extracted, while rRNA was removed, cDNA was reverse transcriptional synthesized. Transcriptome sequencing was sequenced on HiSeq2000, transcriptome data were analyzed by BIGpre, Tophat, and Cufflinks. Expression profile was analyzed for differential expression, GO, COG functional enrichment analysis, and KEGG metabolic pathway analysis. Results: A total of 1665 differentially expressed genes existed between S. baicalensis group and negative control group. Among them, 1169 genes were up-regulated genes, while 496 genes were down-regulated genes. Under the effect of water decoction of S. baicalensis, genes of NB8 down-regulated in glycolysis, Krebs cycle, biosynthesis of fatty acids, and ribosomal proteins, while genes of NB8 up-regulated chemotaxis and assembly path of flagella, and transportation protein ABC pathway. Conclusion: The molecular mechanism of the inhibitory effect of S. baicalensis to UPEC is explained. Targets of S. baicalensis to UPEC are glycolysis, Kreb's cycle, biosynthesis of fatty acids, and translation of proteins. Besides, chemotaxis, the assembly path of flagella, and transportation protein ABC also play a key role in alarm reaction of S. baicalensis.
