1. Effects of vector-mediated targeting human telomerase reverse transcriptase gene RNA interference on proliferation and apoptosis of leukemia HL-60 cells

Objective: To investigate the effects of vector-mediated targeting human telomerase reverse transcriptase ( hTERT) gene RNA interference (RNAi) on the proliferation and apoptosis of leukemia HL-60 cells. Methods: The targeting hTERT gene RNAi recombination vector (pSilencer1.0-U6/hTERT) was transfected into HL-60 cells. The HL-60 cells transfected with empty vector (pSilencer1.0-U6), RNAi-mate or HL-60 cells were used as the controls. The expression of hTERT protein was detected by Western blotting. The proliferation and apoptosis of HL-60 cells were measured by MTT assay and flow cytometry, respectively. Results: The expression levels of hTERT protein in HL-60 cells were lower and the proliferation inhibitory rate and apoptosis rate of HL-60 cells were higher than those of each control group after transfection with pSilencer1.0-U6/hTERT for 24, 72, and 120 h ( P<0.05). There was no differences among HL-60 cells transfected with pSilencer1.0-U6/hTERT for 24, 72, and 120 h, and no differences were also observed among the three control groups ( P>0.05). Conclusion: Vector-mediated targeting hTERT gene RNAi can down-regulate the expression of hTERT protein and inhibit the proliferation and induce apoptosis of HL-60 cells in vitro.
2.A retrospective analysis of the assessment results of arsenide quality control in laboratories of national endemic diseases prevention and control institutions

Objective:To analyse the quality control assessment results of arsenic testing laboratories of national endemic disease prevention and control institutions from 2006 to 2020, so as to further strengthen the quality control of arsenic determination in laboratories of national endemic disease prevention and control institutions, and to ensure the accuracy and reliability of national endemic arsenic poisoning monitoring data.Methods:The water arsenic and urinary arsenic test results of arsenic quality control laboratories from 2006 to 2020 were collected and evaluated by Z-ratio scoring method(│Z│≤2 was qualified, 2 < │Z│ < 3 was basically qualified, │Z│≥3 was unqualified), and the qualification rates of water arsenic and urinary arsenic in different years and different provinces were calculated.Results:From 2006 to 2020, the number of laboratories participating in water arsenic quality control in 14 provinces has increased from 25 to 167, and the number of laboratories participating in urinary arsenic quality control increased from 22 to 90. The standard deviation of laboratory assessment samples was relatively stable, all < 0.05 mg/L. The quality control assessment results from 2006 to 2020 were calculated according to │Z│ < 3, the qualified rate of water arsenic ranged from 64.0% to 100.0%, and that in urine was 54.5% to 100.0%. The quality control assessment results from 2009 to 2020 were calculated according to │Z│≤2, the qualified rate of water arsenic was 84.6% - 100.0%, and that in urine was 83.6% - 100.0%. Among the 13 water arsenic assessment results from 2009 to 2020, according to │Z│≤2, provincial laboratories reported all qualified years for 8 times, prefecture, city and county-level laboratories reported all qualified years for 2 times; there were 8 provinces with all qualified laboratories at province level and 3 provinces with all qualified laboratories at prefecture, city and county-level. Among the 13 assessment results of urinary arsenic from 2009 to 2020, according to │Z│≤2, the provincial laboratories reported all qualified years for 6 times, and the prefecture, city and county-level laboratories all qualified for 1 times; there were 6 provinces in which all provincial laboratories were qualified, and there were 6 provinces in which all prefecture, city and county-level laboratories were qualified.Conclusions:The quality control work of laboratories is improving day by day, but there are still some problems in individual links. Laboratories at all levels should continue to strengthen the construction of laboratories and the training of relevant personnel, further improve the theoretical and technical level of inspectors, and continuously improve the detection quality, to make the analysis data more reliable and comparable, so as to ensure the quality of arsenic poisoning prevention and monitoring work.

4.Effects of intranasal dexmedetomidine as premedication on postoperative behavioral outcomes in children

Objective To observe the effects of intranasal dexmedetomidine (DM)as premedi-cation on postoperative behavioral outcomes in children.Methods Sixty ASA physical status Ⅰ or Ⅱchildren of both genders,2-5 years,weighing 10-30 kg,undergoing hernia surgery,were equally as-signed into three groups (n =20 each)using a random number table:control group (group C),mid-azolam group(group M)and dexmedetomidine group(group D).Thirty minutes before anesthesia in-duction,the children were respectively received intranasal normal saline 0.02 ml/kg (group C),in-tranasal midazolam 0.2 mg/kg (group M)and intranasal DM 2 μg/kg (group D).The sedation score of children apart from parents,the receipt score of face mask for sevoflurane anesthesia induction,the postoperative recovery time,adverse effects,and the percentage of patients requiring rescue analgesic were recorded.To observe the postoperative behavioral outcomes on 1th、7th、30th day using the PH-BQ.Results Compared with group C,the sedation score and the receipt score of face mask of groups M and D were significantly increased (P <0.05).Compared with groups C and M,the adverse effects and the percentage of patients requiring rescue analgesic of group D were decreased (P <0.05).The incidence of the postoperative behavioral outcomes of group C was higher than groups M and D on 1th,7th day (P <0.05).And on 30th day after operation,there was no significant difference among three groups.Conclusion Intranasal dexmedetomidine as premedication can significantly decrease the incidence of the postoperative behavioral outcomes in children.
5.Efficacy of conjunctival autograft transplant and limbal - conjunctival autograft transplant for primary pterygium

AIM: To compare the safety and efficacy of conjunctival autograft transplant ( CAT ) and limbal - conjunctival autograft transplant (LCAT) for primary pterygium.
METHODS: In this prospective, randomized and controlled clinical study, 120 patients ( 120 eyes ) diagnosed with primary pterygium were collected in NO. 474 Hospital of Chinese PLA from January 2014 to January 2015. The 120 patients enrolled in the study in turn, and divided into two group by the odd and even number. The odd number underwent CAT and the even number underwent LCAT.
RESULTS:One hundred and seven patients (107 eyes) completed the follow-up of 1a, including 54 patients (54 eyes) from the CAT group and 53 patients (53 eyes) from the LCAT group. Four patients (4 eyes, recurrence rate 7. 4%) in the CAT group and 2 patients ( 2 eyes, recurrence rate 3. 8%) in the LCAT group developed recurrence. However, there were no significant in recurrence rate between CAT group and LCAT group (P =0. 678).
CONCLUSION: CAT and LCAT might be both effective treatment for primary pterygium for reducing the recurrence rate of pterygium. However, LCAT is the optimal surgical method for primary pterygium.
6.Research on ultrasonic permeability of low intensity pulsed ultrasound through PTFE membrane and Bio-Gide collagen membrane.

The aim of the present study was to detect the transmission rate of ultrasonic low intensity pulsed ultrasound (LIPUS) through polytetrafluoroethylene (PTFE) membrane (Thickness: 0.01 mm) and Bio-Gide collagen membrane, and to provide the basis for the barrier membrane selection on the study of LIPUS combined with guided tissue regeneration (GTR). The ultrasonic (LIPUS, frequency 1.5 MHz, pulse width 200 micros, repetition rate 1.0 kHz) transmission coefficient of the two kinds of barrier membrane were detected respectively through setting ten groups from 10 to 100mW/cm2 every other 10 mW/cm2. We found in the study that the ultrasonic transmission coefficient through 0.01 mm PTFE membrane was 78.1% to 92.%, and the ultrasonic transmission coefficient through Bio-Gide collagen membrane was 43.9% to 55.8%. The ultrasonic transmission coefficient through PTFE membrane was obviously higher than that through Bio-Gide collagen membrane. The transmission coefficient of the same barrier membrane of the ultrasonic ion was statistically different under different powers (P < 0.05). The results showed that the ultrasonic transmittance rates through both the 0.01 mm PTFE membrane and Bio-Gide collagen membrane were relatively high. We should select barrier membranes based on different experimental needs, and exercise ultrasonic transmission coefficient experiments to ensure effective power.

7.Analysis of the factors associated with clinicopathological characteristics of papillary thyroid carcinoma

Objective BRAFV600E mutation, RET/PTC rearrangement, and the concomitant of Hashimoto's thyroiditis(HT) could influence clinicopathological features of papillary thyroid carcinoma (PTC).This study is to investigate the distribution of three factors in PTC and to analyze their associations with clinicopathological characteristics.Methods Fine-needle aspiration samples were collected in a total of 122 conventional PTC patients, who were confirmed by surgery.The clinicopathological features were collected to analyze its association with different factors.BRAFV600E mutation and RET/PTC rearrangement were detected by pyrosequencing and Taqman-qPCR, respectively.Results BRAFV600E mutation was significantly correlated with an older age and a less coexistence with HT(P<0.05).In contrast, RET/PTC rearrangement was more prevalent in young patients and was associated with the concomitant of HT(P<0.05).In the age of ≥20 year and<45 year groups, BRAFV600E mutation was significantly associated with extrathyroidal invasiveness.RET/PTC rearrangement was significantly associated with bilateral lymph node metastasis and the number of metastatic lymph node.Conclusions The distribution of three factors were different in PTC patients.In addition to the age at diagnosis, all of three factors should also be considered together to analyze the association of clinicopathological features of PTC.
8.Effect of drug-sustained capsular tension ring on proliferation of lens epithelial cells in vitro

Objective Our study was performed to design a drug-sustained capsular tension ring (CTR) to evaluate its potentiality on prevention of PCO in the swine capsular bag model in vitro.Methods Following the continuous capsule curvilinear capsulorhexis ( CCCC),Phacomulsification with capsular tension ring implantation was pedormed.CTR-supported swine capsular bag models were prepared and divided into two groups,group CTR ( n =13 ) implanted with the original CTR without any modification and group CTR-PLGA-MG132 ( n =13) implanted with the CTR covered with PLGA and MG132.The CTRsupported capsular bags were cultured in vitro for up to 3 weeks.The area of lens epithelial cells (LEC) coverage over the posterior capsule surface was quantified every day under microscope.The capsules were treated for histological examination.The change of fibronectin was assessed by ELISA assay kit.Results After 2 ~ 3 days,outgrowth of LEC across the posterior capsule was observed,and the posterior capsule was totally covered by a confluent monolayer of cell after (9.06 ± 1.61 ) days in group CTR.Capsular wrinkles became increasingly apparent as time progressed.An increase in capsular thickness was also observed.In contrast,there was less LEC deposition in group CTR-PLGA-MG132.Histological examination showed LEC layers were closely arranged on the posterior capsular surface in group CTR.In group CTR-PLGA-MG132,there was comparatively looser cell arrangement.Compared with group CTR,the mean fibronectin level of posterior capsule by week 3 in group CTR-PLGA-MG132 was 25.14 μg/ml and 106.09 μg/ml respectively.Statistical analysis showed a significant difference ( P < 0.01 ).Conclusions LEC migration,proliferation,and synthesis of EMT markers were inhibited in Group CTR-PLGA-MG132,compared with Group CTR.Drug-sustained capsular tension rings can effectively inhibit the migration,proliferation of LEC and the change of EMT ( epithelial-to-mesenchymal transition) in swine capsular bag models.Drug-sustained capsular tension rings might be a potential therapy to prevent the posterior capsular opacification in the future.
9.Comparative proteomics analysis of aging rat aorta during the process of increasing age

Objective To study the proteins related to aging in aortic of old rats for laying the foundation of further study of aging mechanism.Methods The rat model of aging was built,and all model rats were divided into 4 groups:the adult group(9 weeks),the old group(12 months)of WistarKyoto (WKY) rats,and the age-matched spontaneously hypertensive rats (SHR).Blood pressure of 4 groups was observed.Morphological change of aorta was observed by HE staining.Differential proteins were identified by isobaric tags for relative and absolute quantitation,(iTRAQ)-coupled liquid chromatography and tandem mass spectrometry technology.Part of differential proteins was subsequently detected by real time PCR and Western blot.Results The mean SBP of the old group SHR was higher than WKY of 97.1% (t=39.00,P<0.05),and the adult group of SHR was also higher than WKY of 5.4%(t=3.64,P<0.05).Compared with the adult group,aging change in the aortic morphology of old SHR and WKY were shown in HE staining,and the change in SHR rats was more marked.7 proteins related to aging were identified by Mass spectrum analysis,and they were Profilin-1,Prelamin A,HSP70,creatine kinase-M,Fibulin-5,eIF5A and Prohibitin.Part of differential proteins was subsequently confirmed by real time PCR and Western blot.Prelamin-A was up-regulated in the old group of WKY and SHR (0.15±0.01 vs.0.45±0.04,0.34±0.02 vs.0.78±0.06) (t=12.67,12.06,all P<0.01),Prohibitin was down-regulated in the old group of WKY and SHR(1.34±0.05 vs.1.01± 0.06,1.24±0.05 vs.0.88±0.08) (t=7.41,7.09,all P<0.01).Profilin-1 was up-regulated in the old group of WKY and SHR (9.12±0.4 vs.20.76±0.8,16.84±0.5 vs.55.16±0.9) (t=22.55,64.46,both P<0.01),and Profilin-1 expression in the old group of SHR was higher thanWKY (55.16±0.90 vs.20.76±0.8,t=49.49,P<0.01).Conclusions Differential proteins of the old rat aorta are identified through the comparative proteomics method.These differential proteins will provide new targets for the prevention and control of vascular aging.
10.Dorsal ligament reconstruction for old dorsal dislocation of distal radioulnar joint

Objective To evaluate the efficacy and safety of dorsal ligament reconstruction in treatment of old dorsal dislocation of distal radioulnar joint.Methods Seven patients with old dorsal dislocation of distal radioulnar joint were treated with dorsal ligament reconstruction using the palmaris longus tendon from March 2005 to May 2012 in our institute,including 4 males and 3 females with a mean age of 37 years.All patients had a history of wrist injury for more than 3 months and were diagnosed as isolated dislocation of distal radioulnar joint without fractures.During the operation a bone tunnel was made at dorsal ulnar side of radius near the ulnar notch,which was parallelized to long axial of ulna,two holes were drilled from dorsal to palmarulnaris side through the extensor carpi ulnaris sulcus of the ulna.The palmaris longus tendon was harvested and the strip of the tendon was penetrated through the radial hole.After the tips being crossed,put them through the holes of ulna,reduct the distal radioulnar joint by supinating the forearm,the strip of the tendon was sutured after being tightened,the reversed back the free end of the tendon to reconstruct the sheath of extensor carpi ulnaris tendon.Postoperatively,the upper extremity were kept in a long arm plaster in the position of elbow flexion 90° and forearm supination for 3 weeks,then the below elbow cast was replaced for another 3 weeks.Results Patients were followed-up for 1 year and 8 months 4 years and 2 months with the average of 2 years and 9 months.The rotation of wrist was improved and the handgrip strength was increased significantly.A functional evaluation was performed using the modified Mayo wrist scoring system.All patients had better wrists scores postoperatively (mean,93) compared to preoperatively (mean,68).All patients satisfied with the final result.Conclusion Dorsal ligament reconstruction should be a promise surgical modality for the old dorsal dislocation of distal radioulnar joint.
