AIM: The effective antisense sequences targeted VEGF mRNA with computer software would be screened and designed, and effect of them on growth K562 cells and protein expression of VEGF were studied with experiments. METHODS: Seven antisense sequences were selected and synthesized, which consisted of 18-20 deoxynucleotide acid and were modified with phosphorothioate, according to principle of low free energy of overall △G 37 Overall. Cell growth was assayed by trypan blue dye exclusion assay and level of VEGF protien in the media was determined by ELISA. RESULTS: Six of seven sequences were capable of inhibing growth of K562 cells and downregulating the VEGF protein expression significantly, compared with Scrambed control group. It was found that there was a close correlation between low level of overall △G 37 and antisense effectiveness ( r =0 887, P
8.Differentiation of human umbilical cord mesenchymal stem cells into hepatocyte-like cells with the combined induction of hepatocyte growth factor and fibroblast growth factor
Chinese Journal of Tissue Engineering Research 2009;13(45):8811-8816
BACKGROUND:Fibroblast growth factor can promote proliferation of mesenchymal stem cells (MSCs).grew along the wall. However, there are few reports on the differentiation of MSCs into hepatocytes following fibroblast growth factor induction. When mass concentration of hepatocyte growth factor was 1 μg/L,it can promote mitosis of hepatocytes, and is the strongest mitogenic agent for normal hepatocytes.OBJECTIVE:To explore the biological characteristics of human umbilical cord MSCs/n vitro and their differentiation ability to hepatocyte-like cells under the induction of chemical factors.DESIGN,TIME AND SETTING:The cytological in vitro study was conducted at the Institute of Blood,Jinan University from August 2008 to April 2009.MATERIALS:Umbilical cord was obtained from healthy full-term fetus, which was provided by the Guangzhou Huaqiao Hospital. The parturient signed informed consent.Hepatocyte growth factor and flbroblast growth factor were bought from Peprotech,USA.METHODS:The MSCs from human cord were isolated and cultured by type Ⅳ colagenase digestion+differential adherence. At the third passage,MSCs received cell surface antigen analysis and cell cycle determination to detect their ability to differentiate into adipocytes and osteoblasts. At the fifth passage,MSCs were adjusted into 5×10~9/L and assigned into 2 groups.BMSCs in the control group were incubated in DMEM/F12 containing 5% fetal bovine serum.BMSCs in the induction group were treated with above-mentioned medium supplemented with 20 μg/L hepatocyte growth factor and 10 μg/L flbroblast growth factor.MAIN OUTCOME MEASURES:The following parameters were measured:biological characteristics of human umbilical cord MSCs and differentiation of human umbilical cord MSCs into hepatocyte-like cells in vitro.RESULTS:At the third passage,MSCs derived from human cord expressed CD29,CD44,CD105,but not antigens of hematopoietic CD34,CD45,and 92.2% of them were in G_0/G_1 phase.Oil red O staining showed cytoplasm presented red granules.Alkaline phosphatase staining demonstrated that cytoplasm was black, with the differentiation ability into adipocytes and osteoblasts.Following 10 days of combined induction of hepstocyte growth factor and fibroblast growth factor,RT-PCR and Western blot results confirmed that cells expressed alpha fetoprotein and albumin.Negative expression was found in the control group.CONCLUSION:Human umbilical cord contained plenty of MSCs,with strong potential of multi-differentiation.Umbilical cord MSCs can differentiate into hepatocyte-like cells following combined induction of hepatocyte growth factor and fibroblast growth factor.
9.Design,screening and evaluation of VEGF siRNA
Basic & Clinical Medicine 2006;0(06):-
Objective Nine VEGF siRNAs were designed and their efficiencys were evaluated for introducing new and effective design rules of siRNA.Methods Nine VEGF siRNA sequences with nineteen nucleic acids were designed. After construction in vitro,VEGF siRNAs were transferred into neuroglioma cells by lipofectamine. The level of VEGF protein in the media was determined by ELISA 48 hours after transferring. The relationship between the characteristics of VEGF siRNA and its' interfering effect were analyzed. Results A quantitative parameter called H-b index were used to characterize the secondary structure of VEGF mRNA.A low value of the H-b index would mean that the structure of mRNA is loose,in which the nucleotides of the targeting region are likely to be accessible by siRNA and the effects are better. The experiments showed a negative correlation between the suppression of VEGF protein expression and H-b index(r=-0.498).The H-b indexs of siRNAs targeting VEGF mRNA 386~406 and 401~421 position were less, however the effects of suppression were better. The siRNAs conferred a strong RNAi activity with A residue at position 6 and A/U residues at position 19 of sense strand of siRNAs.Thethermodynamic properties of siRNA play a critical role in the transferring effects. The lower average of internal stability values for positions 9~14 of the antisense strand is facilitating the interfering effect. The lower binding energy is the more stable incorporation between antisense strand of siRNA and mRNA present, the more favorable is for RISC cleaving mRNA.Conclusions The sequences of VEGF siRNA have been designed successfully and effectively inhibited the expression of VEGF protein.
10.Inhibition of VEGF expression and SMMC 7721 cell growth by VEGFsiRNA
Chinese Journal of Pathophysiology 1986;0(04):-
AIM: To determine the inhibition of VEGF small interfering RNA (siRNA) on expression of VEGF protein and SMMC 7721 cell growth. METHODS: Nine VEGFsiRNA sequences with nineteen nucleotides were designed under the assistance of computer. The inhibitory effect of VEGFsiRNA was analyzed by CCK8. The protein level of VEGF in the media was determined by ELISA. The change of cell cycle was detected by flow cytometry. RESULTS: All the VEGFsiRNA were capable of inhibiting the proliferation of SMMC 7721 cells significantly compared with control and lipofectamine control, while the inhibitory effects of VEGFsiRNA2, VEGFsiRNA4, VEGFsiRNA6, VEGFsiRNA7, VEGFsiRNA8 and VEGFsiRNA9 were better than that of antisense oligodeoxynucleotide. All the VEGFsiRNA reduced the expression of VEGF protein. The effect of VEGFsiRNA7 and VEGFsiRNA2 were the best with the inhibitory rates of 52.65% and 50.43%, respectively. VEGFsiRNAs induced the S arrest in SMMC 7721 cells. The signs of apoptosis in SMMC 7721 cells induced by VEGFsiRNA were not observed. CONCLUSION: VEGFsiRNA sequences were designed and synthesized successfully. VEGFsiRNA effectively inhibited the proliferation of SMMC 7721 cells and reduced the level of VEGF protein.
