Objective To observe clinical efrect of treating angina pectoris with Shuxuening injection combined with western medicine.Methods 62 cases patients with angina pectoris were randomly recurited into a control group and a treatment group,The control group was treated with western medicine,and the treatment group was treated with Shuxuening injection based on the control group.The clinical effect was observed between the two groups.Results The clinical effect in the treatment group(83.9%)was significantly higher as compared to the control group(64.5%)(χ~2=2.352,P＜0.05).Conclusion The treatment of Shuxuening injection combined with western medicine on angina pectods is better than westem medicine only.

The plasticity of bone marrow stem cells is an important discovery in the field of stem cell research, which provides the possibility of bone marrow stem cells to be used for non-hematopoietic system diseases. Objective To evaluate the effect and safety of ultrasound-guided pereutaneous thrombin injection for the treatment of iatrogenic femoral arterial pseudoaneurysm.Methods Thirty patients [6 male,24 female,age range 45-82 years,mean (63 ± 10) years] were found to have pseudoaneurysms confirmed by ultrasound between 1 and 7 days following femoral arterial puncture from July 2011 through July 2014.Results All patient had pseudoaneurysm thrombosis after the first procedure.Thrombin 200-1000 U (50 U/mL)was injected into the pseudoaneurysm under ultrasound-guidance,performed.One patient showed acute allergy 5 min after thrombin injection,which was relieved by anti-allergic therapy.No thromboembolic complications or infections occurred.Conclusions Ultrasound-guided pereutaneous thrombin injection is a safe and effective noninvasive method for the treatment of iatrogenic femoral pseudoaneurysm and should be considered as first-line therapy.

Objective To explore the validity of oxygen atomizing inhalation pulmicort respulas combined with montelukast in treating children with asthma and its role in pulmonary function and T lymphocyte subset.Methods Totally 78 cases treated in Huizhou People's Hospital from June,2014 to June,2016 were randomly divided into observation group and control group with each group of 39 cases.The control group on the basis of routine treatment with oxygen atomizing inhalation of pulmicort 1 mg,twice daily,and the observation group based on control group added Montelukast Sodium Chewable Tablets 5 mg,once daily,one week for one course.The clinical effect,pulmonary function,and level of T lymphocyte subset were compared between two groups.Results The total effective rate in the observation group was 94.70％,which was significantly higher than control group (82.05％,P ＜ 0.05).After therapy,the symptom scores of two groups were obviously decreased compared with those before therapy (P ＜ 0.05),and those of the observation group were significantly lower than control group (P ＜ 0.05);The pulmonary function of the two groups were obviously improved (P ＜ 0.05) compared with that before therapy (P ＜ 0.05),and the levels of FEV1,FVC,and FEV1/FVC in observation group were significantly higher than control group (P ＜ 0.05);The level ofT lymphocyte subset in control group showed no statistical difference compared to that before therapy,and the levels of CD4+ and CD4+/CD8+ in observation group were obviously decreased,whereas CD8+ was increased.Conclusion Oxygen atomizing inhalation pulmicort respulas combined with montelukast could effectively increase the clinical effect on children with asthma,improve pulmonary function and positively regulate immune function,which deserves clinical expansion.

BACKGROUND:Fibroblast growth factor can promote proliferation of mesenchymal stem cells (MSCs).grew along the wall.However,there are few reports on the differentiation of MSCs into hepatocytes following fibroblast growth factor induction.When mass concentration of hepatocyte growth factor was 1 μg/L,it can promote mitosis of hepatocytes,and is the strongest mitogenic agent for normal hepatocytes.OBJECTIVE:To explore the biological characteristics of human umbilical cord MSCs/n vitro and their differentiation ability to hepatocyte-like cells under the induction of chemical factors.DESIGN,TIME AND SETTING:The cytological in vitro study was conducted at the Institute of Blood,Jinan University from August 2008 to April 2009.MATERIALS:Umbilical cord was obtained from healthy full-term fetus,which was provided by the Guangzhou Huaqiao Hospital.The parturient signed informed consent.Hepatocyte growth factor and flbroblast growth factor were bought from Peprotech,USA.METHODS:The MSCs from human cord were isolated and cultured by type Ⅳ colagenase digestion+differential adherence.At the third passage,MSCs received cell surface antigen analysis and cell cycle determination to detect their ability to differentiate into adipocytes and osteoblasts.At the fifth passage,MSCs were adjusted into 5×10~9/L and assigned into 2 groups.BMSCs in the control group were incubated in DMEM/F12 containing 5% fetal bovine serum.BMSCs in the induction group were treated with above-mentioned medium supplemented with 20 μg/L hepatocyte growth factor and 10 μg/L flbroblast growth factor.MAIN OUTCOME MEASURES:The following parameters were measured:biological characteristics of human umbilical cord MSCs and differentiation of human umbilical cord MSCs into hepatocyte-like cells in vitro.RESULTS:At the third passage,MSCs derived from human cord expressed CD29,CD44,CD105,but not antigens of hematopoietic CD34,CD45,and 92.2% of them were in G_0/G_1 phase.Oil red O staining showed cytoplasm presented red granules.Alkaline phosphatase staining demonstrated that cytoplasm was black,with the differentiation ability into adipocytes and osteoblasts.Following 10 days of combined induction of hepstocyte growth factor and fibroblast growth factor,RT-PCR and Western blot results confirmed that cells expressed alpha fetoprotein and albumin.Negative expression was found in the control group.CONCLUSION:Human umbilical cord contained plenty of MSCs,with strong potential of multi-differentiation.Umbilical cord MSCs can differentiate into hepatocyte-like cells following combined induction of hepatocyte growth factor and fibroblast growth factor.

AIM:To explore the effects of the combined use of human telomerase reverse transcriptase (hTERT) gene antisense oligodeoxynucleotide (ASODN) and cisplatin on apoptosis of HL-60 cells. METHODS:PCR-ELISA was used to determine telomerase activity in HL-60 cells untreated or treated with ASODN, the expression levels of hTERT protein were assayed by immunofluorescence using fluoresce isothiocyanate (FITC) label. Apoptosis was detected by DNA electrophoresis and flow cytomertric cell cycle analysis. RESULTS:The expression levels of hTERT protein and telomerase activity in HL-60 cells treated with ASODN were shown to decrease with time, the DNA fragments were obviously found and the percentage of apoptosis cells was significantly enhanced in HL-60 cells with the combined use of hTERT ASODN and cisplatin compared with the combined use of hTERT sense oligodeoxynucleotide and cisplatin or cisplatin alone, respectively. CONCLUSION:Inhibition of telomerase activity by hTERT ASODN increases the cisplatin-induced apoptosis of HL-60 cells.

AimTo explore the effects of bcl-2 antisense oligonucleotide targeting the coding region of the bcl-2 mRNA on apoptosis induced by Vp16 in HL60 and K562 cells. Methods Drug sensitivity was compared by MTT cytotoxicity assay, and expression of bcl-2 protein and apototic cells were assayed by flow cytometry. Results The bcl-2 antisense oligonucleotide of 10 μmol · L- 1 combined with etoposide inhibited expression of bcl-2 protein,increased apoptosis in HL60 and K562 cells and decreased IC50 of etoposide; The antisense oligonucleotide targeting the coding region of the bcl-2 mRNA had stronger effection than the antisense oligonucleotide targeting the translation initiation. ConclusionThe bcl-2 antisense oligonucleotide targeting the coding region of the bcl-2 mRNA can enhances etoposide induced apoptosis of HL60 and K562 cell.

Objective To investigate the effects of continuous infusion of moderate dose tranexamic acid and ulinastatin on fibrinolysis during orthotopic liver transplantation (OLT).Methods Thirty ASA Ⅲ or Ⅳ patients aged 34-63 yr with a body mass index of 17-37 kg/m2 and end-stage liver disease score 6-34,undergoing OLT,were randomly assigned to one of 3 groups ( n =10 each):group control (group C) ; group tranexamic acid (group T) and group ulinastatin (group U).The patients received continuous infusion of normal saline at 10 ml/h in group C or ulinastatin at 100 000 U/h in group U immediately after tracheal intubation until 120 min after portal vein was declamped,while in group T the patients received a loading dose of tranexamic acid 1 g followed by continuous infusion at 10 mg· kg-1 ·h- 1.Prothrombin time (PT),activated partial thromboplastin time (APTT),international normalized ratio ( INR),fibrinogen (Fg),D-dimers (D-D) and fibrin degradation product (FDP) were measured before induction of anesthesia (baseline),at 120 min after skin incision,30 min after clamping of portal vein (anhepatic phase),30 and 120 min after declamping (neohepatic phase) and at the end of operation.The amount of blood loss and transfusion were recorded.The patients were followed up after operation for hepatic artery and portal vein thrombosis in groups U and T.Results There were no significant differences in PT,APTT,Fg,INR and amount of blood loss and transfusion among the 3 groups.The plasma D-D concentration and percentage of patients with plasma FDP ＞ 20μg/ml were significantly lower in group T than in group C.There were no significant differences in plasma D-D concentration and percentage of patients with plasma FDP ＞ 20 μg/ml between groups U and C.No hepatic artery and portal vein thrombosis was detected after operation in groups T and U.Conclusion Continuous infusion of moderate dose tranexamic acid can inhibit fibrinolysis during OLT,but can not reduce the amount of blood loss.Continuous infusion of ulinastatin ( 100 000 U/h) has no significant effect on fibrinolysis during OLT.

Objective:To determine whether the bcl-2 antisense oligonucleotide (ASODN) can increase the sensitivity of NCI-H460 cell lines to docetaxel or radiation. Methods: Docetaxel in combination with bcl-2 ASODN or non-sense oligonucleotide (NSODN) was used to treat NCI-H460 cells, and then IC_~50 of docetaxel against NCI-H460 cells was determined with MTT method. In radiation group, NCI-H460 cells were divided into pure radiation, radiation plus bcl-2 ASODN, and radiation plus NSODN groups. The inhibition rates of cell growth were assayed by MTT, and the expression levels of Bcl-2 protein were assayed by immunofluorescence.
