1.Clinical Observation on Different Administrations of Levofloxacin to Eyes Traumas during Perioperative Period for Preventing Infection

[Objective] To discuss the clinical effect of Levofloxacin in eye drop and oral taking on preventing infection for eyes traumas during perioperative period. [Method] Respectively select 126 cases of non-open eye trauma operation and 162 cases of open eye trauma operation, divide the non-open eye trauma patients into group A(Levofloxacin eye drop) 66 cases and group B(Levofloxacin injection in vein drop) 60 cases, compare both bacteria culture positive rates before administration and after operation;divide the open eye trauma patients into group C(Levofloxacin in eye drop and oral taking)80 cases and group D(Levofloxacin in vein drop) 82 cases, compare both positive rates of bacteria culture before and after the administration and operation.[Result] Comparing group A and group B with groups C and D, the positive rate before and after Levofloxacin administration, there ’s difference of statistical meaning, but without marked statistical meaning after operation. Side effects: the positive rate of bacteria culture was 56.25% before administration, and 10.13% after operation; for group D, they were 59.80% and 12.20% respectively, the difference had statistical meaning on Levofloxacin, but without difference of statistical meaning on positive rate. Purely using focal eye drop had no side effects on group A; the side effects of oral taking and focal administration on group C were much lower than groups B and D. [Conclusion] For eye trauma in perioperative period, the oral taking and eye drop of Levofloxacin have the same effects on infection prevention compared as vein drop, while the focal administration can reduce antibiotics usage and systemic administration, with little side effects, as wel as effectively prevent post-operation infection of eye trauma, worth clinical promotion and application.
3.Expression and function of protease-activated receptor-2 on tumor cells

Protease-activated receptor-2 (PAR-2) belongs to the receptor family which is coupled with G-protein. The N-terminus of PAR-2 is cleaved by some proteases to generate a new N-terminus. The new N-terminus can interact with and activate the receptor itself. The update research reported that PAR-2 could be expressed on some cancers as well as tumor cell lines. There are some relationships between the PAR-2 expression and the behaviors of tumor cell, such as proliferation, angiogenesis, migration and invasion.
4.Combined transvaginal and abdominal ultrasound in the diagnosis of early ectopic pregnancy

Objective To observe the accuracy of abdominal ultrasound combined with transvaginal ultra-sound in the early diagnosis of ectopic pregnancy,thus to provide reference for clinical diagnosis of ectopic pregnancy. Methods The clinical data of 68 patients,who pathologically confirmed or conservative treatment with clinical diag-nosis of ectopic pregnancy,were retrospectively analyzed.All patients were treated with abdominal ultrasound,vaginal ultrasound examination.The accuracy and missed ectopic pregnancy rate of transvaginal ultrasound and abdominal B ultrasound and joint inspection were observed.And the presence or absence of abnormal ovarian uterus were recorded. Results By comparison with pathological diagnosis,joint diagnosis of 32 cases of ectopic pregnancy,the diagnosis rate was 94.1 %,missed 2cases,accounted for 5.9%;Transvaginal ultrasound diagnosis were 26 cases,accounted for 76.5%,missed by 8 cases,accounted for 23.5%;Abdominal ultrasound diagnosis were 24 cases,accounted for 70.6%,missed 1 0 cases,accounted for 29.4%.Joint inspection diagnosis rate was significantly higher than abdominal or vaginal ultrasound(χ2 =1 6.75,1 4.38,all P <0.05),and then compared transvaginal ultrasound and abdominal ultrasound diagnosis rate(P >0.05).Transvaginal ultrasound detected primitive heart tube pulse,mass,blood flow and gestational sac -like structures within the mass were significantly higher than the probability of abdominal ultra-sound(χ2 =26.71 ,7.1 8,1 0.25,50.28,25.69,6.39,all P <0.05).Conclusion United abdominal ultrasound and transvaginal ultrasound can significantly improve the early detection rate of ectopic pregnancy,which can provide refer-ence for clinical diagnosis and treatment.
5.Expression and prognosis in patients with cerebral infarction blood precursor protein and β-tubulin

Objective To investigate the expressions of amyloid precursor protein and β-microtubule protein,and to study their clinical significance in patients with cerebral infarction.Methods 108 cases of acute cere-bral infarction as the observation group and 60 normal adults as the control group were collected in the study.The expressions of amyloid precursor protein and β-microtubule protein in different clinical characters were detected by western blot.Results The results of observation group [APP:(168.78 ±13.64)μg/L,β-tubulin:(124.64 ± 27.08)μg/L]were significantly higher than those in control group (t =22.81,P <0.01;t =21.08,P <0.01).Their expressions were correlated with CT scores and prognosis.Conclusion The higher expressions of amyloid precursor protein and β-microtubule protein may promote the occurrence and development in patients with cerebral infarction. The joint detection should be helpful to predict the prognosis and direct the treatment.
6.Changes and clinical significance of P selectin , lysosome protein expression in patients with ischemic stroke

Objective To study the changes and clinical significance of P selectin (CD 62P), lysosome protein (CD 63) expression in patients with ischemic stroke.Methods 168 cases of acute ischemic stroke (both acute and convalescence) and 40 normal controls were tested for CD 62P and CD 63 expression using flow cytometry. The correlation between the expression and neurological functional deficit scale was analyzed.Results (1) The expression of CD 62p and CD 63 in acute phase (9.48%, 8.36%) of ischemic stroke was markedly higher than that in convalescence phase (5.73%, 4.21%)( P0.05). 7.The DNA Sequence Difference of ERG11 Gene Between the Yeast-form and the Hyphal-form of Candida albicans

Objective To explore the difference of gene sequence between the yeast- and the hyphal-form of Candida albicans. Methods The genomic DNA of both the yeast-form and the hyphal-form of seven isolates were extracted. The isolates were obtained from the same patient infected with HIV, but their sensitivities to fluconazole were different. These isolates were derived from the same C.albican strain as identified by chromosomal analysis and DNA subtype analysis. DNA fragments (310 bp) of ERG11 were amplified by PCR and the difference of DNA sequence between the two forms of each isolates was compared by DNA sequencing. Results Three isolates were found to have sequence difference between the yeast- and the hyphal-form of C. albicans at the position of 1 547 bp, 1 587 bp and 1 617 bp. Conclusion The sequence difference of ERG11 gene exists between the yeast- and the hyphal-form of C. albicans.
8.Effects of Intracerebral Transplantation of Genetically Modified Myoblasts Producing BDNF on Different Brain Regions of Neonatal Rats Subjected to Hypoxic-ischemic Encephalopathy

Objective To investigate the neuroprotection of different brain regions after transplantation of genetically modified myoblasts producing brain derived neurotrophic factor (BDNF) into cortices of neonatal rats subjected to hypoxic ischemic encephalopathy (HIE). Methods Seven day old pups were randomized into sham operated group (C), HIE+BDNF group (B) and HIE+mock transplantation group (A). A rat myoblast cell line expressing and secreting BDNF (BDNF(+)/L 6TG) was constructed. A stereotaxical intracerebroparenchymal transplantation of either BDNF(+)/L 6TG (B) or BDNF(-)/L 6TG (absence of BDNF, A) at 0.8 ?l of cell suspension (4?10 4/?l) into the left cortex of the brain was carried out shortly after HIE undergone by ligation of left common carotid artery followed by a 2.5 h inhalation of humidified 8% O 2+92% N 2 at 37℃. Changes of areas of different brain regions of cortex, hippocampus and striatum at different sections were observed 21 d after the procedure. Injury severity scores of brain tissues were also performed. Results Twenty one days after the manipulation, various degrees of atrophy were observed in the three regions of left hemispheres in group A or B versus contralateral corresponding regions or group C whereas this atrophy in group B was significantly reduced when compared to group A. Not only was the atrophy of cortex near the grafting point alleviated but some other regions of left hemispheres. A similar change pattern was seen in injury severity scores of the tissue. Conclusion Present data suggest that intracerebral transplantation of genetically modified myoblasts producing BDNF has a beneficial effect on the protection from damage of both circumscriptus brain tissues and some other areas distanced from the grafted place.
9.Effects of curdione on cognitive function and expression of inflammatoryfactors in the hippocampus after partial hepatectomy in aged mice

Objective To explore the effects of curdione on cognitive function and expression of inflammatory factors in the hippocampus after partial hepatectomy in aged mice.Methods The animals were divided into control group, sham group, operation group, and high-, medium-and low-dose curdione groups.The model of partial hepatectomy in aged mice was established according to the method reported in literature.The levels of SOD, MDA, CAT and GSH-Px in the hippocampus were detected by enzyme-linked immunosorbent assay (ELISA), while the expressions of NF-κB, IL-1β, IL-6 and TNF-α were measured by Western blot.Results The escape latency was significantly increased (P<0.01), and the number of platform-site crossovers was significantly decreased in operation group (P<0.05).Compared with those in operation group, curdione significantly decreased escape latency and increased platform-site crossovers (P<0.05, P<0.05).The levels of SOD, CAT and GSH-Px in the hippocampus were significantly decreased (P<0.05) and the content of MDA was significantly increased after partial hepatectomy (P<0.01), and these were reversed by curdione (P<0.05, P<0.05, respectively).The expressions of NF-κB, IL-1β, IL-6 and TNF-α protein were significantly inhibited by curdione (P<0.05).Conclusion Curdione can significantly improve cognitive dysfunction after partial hepatectomy in aged mice and the mechanism may be related to its inhibition of oxidative stress, inhibition of NF-κB activation and reduction of inflammatory cytokines IL-1β, IL-6 and TNF-α in the hippocampus.
