2.Preparation Technology of Chinese Traditional Medicine Seborrhea Liniment

OBJECTIVE:To study the preparation technology of Chinese traditional medicine Seborrhea liniment.METHODS:The preparation technology of Seborrhea liniment was optimized by orthogonal experiment design L9(34)using 60% alcohol as solvent with the content of Berberine hydrochloride as index for evaluation and with the extraction method,extraction time,amount of solvent and number of extraction times as factors.RESULTS:The optimum preparation technical conditions were as follows:by impregnating method,the extraction time was 48 h using 1 500 mL alcohol and the extraction frequency was 2 times.CONCLUSION:The preparation technology of Chinese traditional medicine Seborrhea liniment is stable and feasible,and it serves as a theoretical basis for the industrial production.
3.Expression of neuroglobulin during the process of brain edema induced by endotoxin in rats

Objective To study the expression of neuroglobulin(Ngb) in cortex of frontal lobe,hippocampus,cerebrospinal fluid(CSF) and plasma of rats during the process of brain edema induced by endotoxin(LPS).Methods Seventy SD rats were randomly divided into the control group(n=10) and the LPS groups(10 each at six time points).An intraventricular injection of LPS(0.1mg/kg,0.2ml) was given to rats in LPS groups,while the same amount of normal saline was given in control group.The plasma,CSF,hippocampus and the cortex of frontal lobe of rats were collected 3,6,12,24,48 and 72 hours after the injection.Ngb was detected by ELISA,Western blotting and immunohistochemical(IHC) method.The brain water content was measured by wet/dry method.The swelling of organelles in hippocampus and the cortex of frontal lobe were observed by transmission electron microscopy(TEM).Results The brain water content was significantly higher in LPS groups than in control group(P
4.The evaluation of efflux transporter model based on RNA interference technology in vitro.

In the present study, the specifically knockdown models of P-gp or MRP2 were constructed by using a series of chemically synthesized small interfering RNA (siRNA) in vitro. The expression of P-gp and MRP2 was measured by real-time PCR and Western blot, and the function was evaluated by applying P-gp and MRP2 substrate, rhodamine and methotrexate. The results showed that MRP2 siRNA-3 or P-gp siRNA-2 significantly decreased the mRNA expression of MRP2 or P-gp, the inhibition ratio was 68% or 84%; MRP2 siRNA-3 or P-gp siRNA-2 at a dose of 80 nmol x L(-1) significantly reduced the protein expression of MRP2 or P-gp at 48 h after treatment, the inhibition ratio was 62% or 70%. Meanwhile, other transporters were not influenced by siRNA. When pretreatment with MRP2 siRNA-3 or P-gp siRNA-2, the efflux of methotrexate or rhodamine decreased significantly and the intra-cellular concentration increased. The results suggested that chemically synthesized siRNA could significantly inhibit the expression and function of MRP2 and P-gp, and the model of RNAi in vitro could be used to evaluate the role of efflux transporters in transportation of drugs.

5.Research progress of Chinese herbal medicine raw materials in cosmetics.

Advocating green, nature, environmental protection, safety and the pursuit of efficacy are the trends of cosmetics in the world. In recent years, more and more Chinese herbal extracts with mild, high safety and small irritation are applied to cosmetics as the natural additives. This has become a new hot spot. The recent application advances of Chinese medicine raw materials in cosmetics are overviewed according to their main functions. This review will provide useful references for the future development and application of Chinese medicinal herbs cosmetics.

6.Effect of xin an granule on electrophysiological response of ventricular muscle cell in rabbits with ischemia.

OBJECTIVETo investigate the electrophysiological effect of Xin' an granule (XAG) on ventricular muscle cell in ischemic rabbits.
METHODSA total of 48 rabbits were divided into the normal group and the ischemic group, and then subdivided into three groups, the control group, the high and low-dose XAG groups, 8 in each group. Rabbits in the low-dose XAG group and the high-dose XAG group were gastrogavaged XAG at the daily dose of 0. 85 g/kg and 3.40 g/kg, while the others in the control group were given the equal dosage of normal saline. All the rabbits were treated three times per day for successive 10 days. The rabbit model of ischemia was established by intravenous injected with 2. 5 U/kg posterior pituitary injection. Five minutes later, the monophasic action potential (MAP) and electrocardiogram (ECG) of each rabbit in the different groups were recorded and compared.
RESULTS(1) To normal rabbits, XAG could significantly shorten the action 50% and 90% potential duration (APD)50 and APD90 of ventricular muscle cell (P < 0.05 ), and high-dose of XAG could significantly increased the Vmax of MAP(P <0. 05). (2) While to ischemic rabbits, XAG could significantly prolong APD50 and APD90, and significantly increased the action potential amplitude (APA) and Vmax of MAP (P < 0. 05).
CONCLUSION(1) XAG can significantly shorten APD50 and APD90 of ventricular muscle cell, and high-dose XAG significantly increase the Vmax of MAP of normal rabbits. (2) XAG can delay and alleviate the manifestation characteristics of action potential of ventricular muscle cell during ischemia.

7.The immunoregulatory effects of bone marrow mesenchymal stem cells on allogeneic peripheral B lymphocyte

Objective To study the immunoregulatory effects of bone marrow mesenchymal stem cells (MSC) on allogeneic peripheral B lymphocytes in vitro. Methods MSCs were isolated and cultured from bone marrow by gradient centrifugation. Mononuclear cells were isolated routinely from peripheral blood, then monocytes were eliminated by L-leucine methy ester method. Remained T lymphocytes were eliminated by AET-SRBC rosette method. The action of MSCs and its supernatant on B lymphocytes proliferation in the presence of anti-human IgM goat antibodies (anti-IgM) was investigated by MTT. The IgG, IgM in the supernatant were detected by ELISA. The percent of apoptosis B lymphocytes, co-cultured with MSCs for 24 or 48h, was assayed by FACS. Results MSCs and its supernatant inhibited B lymphocytes proliferation and Ig secretion. The inhibitory effect depended on the amount of MSCs and condition of its supernatant. The date of FACS indicated that the apoptosis ratio of B lymphocytes, co-cultured with MSCs for different times, were non-significant. The inhibitory effect of MSCs on B lymphocytes was temporary and reversible. Conclusion MSCs have immunoregulatory effects on B lymphocytes, and its mechanisms are complex, not only correlating with the concentration of MSCs but also the action between cells and the secretory cytokine of MSCs.

9.Study on absorbing volatile oil with mesoporous carbon.

Clove oil and turmeric oil were absorbed by mesoporous carbon. The absorption ratio of mesoporous carbon to volatile oil was optimized with the eugenol yield and curcumol yield as criteria Curing powder was characterized by scanning electron microscopy (SEM) and differential scanning calorietry (DSC). The effects of mesoporous carbon on dissolution in vitro and thermal stability of active components were studied. They reached high adsorption rate when the absorption ratio of mesoporous carbon to volatile oil was 1:1. When volatile oil was absorbed, dissolution rate of active components had a little improvement and their thermal stability improved after volatile oil was absorbed by the loss rate decreasing more than 50%. Absorbing herbal volatile oil with mesoporous carbon deserves further studying.

10.Advance in studies on anti-cancer activity and mechanism of flavonoids.

Flavonoids are natural products that are ubiquitous in the natural world, with wide physiological activities and low toxic and side effects. In recent years, their anti-tumor effect has caused widespread concern and studies. According to the findings, flavonoids have prominent effects in preventing and treating lung cancer, breast cancer, colon cancer, prostate cancer, liver cancer, leukemia, ovarian cancer, gastric cancer and so on. Their anti-tumor mechanisms mainly include anti-oxidation, anti-free radical, induction of apoptosis of cancer cells, impact on cell cycle, immune regulation, inhibition of tumor angiogenesis, inhibition of COX-2, inhibition of telomerase activity and so on. This article focuses on the advance in domestic and foreign studies on anti-cancer activity and mechanism of flavonoids, in order to provide theoretical basis and research ideas for the further development and clinical application of flavonoids.
