2.Significance of Changes of Blood Lipids,Endothelin and Carbohydrate Tolerance in Simple Obesity Children

Objective To investigate the changes of blood lipid,endothelin(ET)and carbohydrate tolerance in simple obesity children.Methods Serum total cholesterol(TC),triglyeride(TG),low density lipoprotein(LDL),endothelin(ET) and oral glucose tolerance test(OGTT) of 436 cases(light obesity group 242,moderate obesity group 138,heavy obesity group 56) were detected respectively.Results The levels of TC,TG,LDL,ET and OGTT in 3 simple obesity groups were all higher than those in control group obviously and there were significant differences.The significant positive correlation between the obesity children serum lipids,ET and obese degrees were observed.Four insulin resistant children were found in heavy obesity group and none in other groups.Conclusions The levels of serum lipids and ET increases in obesity children and the OGTT decreases in some heavy obesity children.For preventing the obesity related health problems,the simple obesity children should be paid more attention to earlier.

6.Degradation of earthworm extracts prepared by wet superfine grinding in simulated gastrointestinal environment.

This is to report the study of degradation of earthworm extracts prepared by wet superfine grinding in simulated gastrointestinal environment. Enzymatic reactions were terminated by adjusting the solution pH or using membrane bioreactor principle. Earthworm protein concentration change was detected by Bradford method, the degraded state of protein was described with SDS-PAGE technology, and the degraded state of small molecule substances was detected by HPLC. The results showed that earthworm protein degraded completely in artificial gastric juice. High molecular weight protein degraded greatly in artificial intestinal fluid, while low molecular weight protein was not significantly degraded. Small molecular substances degradation did not degrade in artificial gastric juice, while they degraded obviously in artificial intestinal fluid, there is even new small molecule substance appeared. Finally it is concluded that the substance that having therapeutic effects in vivo may be some degraded peptide, amino acid and stable small molecules existed in artificial intestinal fluid.

Object To explore the feasibility of breeding genetic modified (GM) medicine by expressing human cytokine in transgenic Chinese materia medica Methods Human interferon ? gene and RANTES gene available from the amplification in vitro were enzymatically excised, recoveried, and inserted into intermediate vectors The recombinants were identified by double enzyme digestion of EcoRⅠand HindⅢ The plasmids were extracted from Escherichia coli and introduced into A tumefaciens, and the transformants harboring binary vectors were screened by addition of antibiotics of kanamycin (Km) and rifampicin (Rif), and the explants of M charantia and P vulgaris were transformed by co cultivation of leaf disks with A tumefaciens strain Results RT PCR was applied to detect the transient expression of human interferon ? gene and RANTES gene in transformed medicinal herbal calli Conclusion The expression of recombinant human interferon ? gene and RANTES gene in transgenic M charantia and P vulgaris cells was firstly reported, which opens an alternative road to antivirus, especially anti AIDS virus, by using transgenic Chinese materia medica

AIM To set up a method which can determining the blood concentration of omeprazole and its metabolite 5′ omeprazole, omerpazole sulphone in order to study its cinical pharmacokinetics. METHODS The blood concentration of omerpazole was determinated by HPLC. RESULTS Calibrated standard curve of omeprazole in blood is Y=-0 004 499+0 001 909X (r =0 9990), the recoveries of three concentrations 50, 500, 2 000 mg?L -1 are 90 36, 109 62, 108 91%, respectively; and the precisions are 9 86,7 86, 15 52% , respectively. Calibrated standard curve of its metabolite 5′ OH omeprazole in plasma is Y=-0 003 659+0 001 328X(r =0 9970), the recoveries of three concentrations 20, 200, 1 000 mg?L -1 are 79 42%, 96 49%, 95 04%, respectively; and the precisions are 8 95%, 4 52%, 9 73%, respectively. Calibrated standard curve of its another metabolite omeprazole sulphone in plasma is Y=0 009 248+0 001 765X (r =0 999 2), the recoveries of three concentrations 20, 200, 1 000 mg?L -1 are 94 44%, 105 59%, 104 26%, respectively; and the precisions are 8 72, 8 58, 9 60%, respectively. After 20 mg omeprazole were administered by a voluteer via oral, C max of 5′ OH omeprazole, omeprazole, and omerpazole sulphone were 14 622 7, 408 433 2, 454 363 7 mg?L -1 . CONCLUSION The method is good enough to study pharmacokinetics of omeprazole.
9.Comparison of genotype polymorphism of cytochrome CYP2C19 between Chinese Han subjects and Chinese Meng subjects

AIM: To compare genotype polymorphism of cytochrome CYP2C19 between Chinese Meng subjects and Chinese Han subjects. METHODS: The genotype polymorphism was analyzed by PCR RFLP (Restriction Fragment Length Polymorphism) in 74 subjects of healthy Chinese Han and 6 subjects of Chinese Meng. RESULTS: Of the 74 genotyped healthy subjects, 31 ( 41.9 %) were homozygous for wildtype (wt/wt), 9 ( 12.2 %) were homozygous for CYP2C19m1 and CYP2C19m2 (m1/m1 or m1/m2 or m2/m2), and 34 ( 45.9 %) were heterozygous for CYP2C19m1 or CYP2C19m2 (m1/wt or m2/wt). Among the 6 genotyped Chinese Meng, 1 ( 16.7 %) was homozygous for wildtype (wt/wt), 2 ( 33.3 %) were homozygous for mutant allele CYP2C19m1 and CYP2C19m2 (m1/m1 or m1/m2 or m2/m2), and 3( 50.0 %) were heterozygous for CYP2C19m1 or CYP2C19m2 (m1/wt or m2/wt); no homozygous genotype for CYP2C19m2(m2/m2)was found in this study. CONCLUSION: There is no statistical difference in occurrence of wt/wt and m1/m1 between in 74 subjects of healthy Chinese Han and 6 subjects of Chinese Meng.
10.Effects of Sinomenine on the rat dendritic cells after hepatic ischemia-reperfusion

Objective To study the effects of Sinomenine on the dendritic cells after hepatic ischemia-reperfusion. Methods Forty-eight BN rats were equally divided into control group, low dose (40 μg/g) and high dose (80 μg/g) of Sinomenine groups after the liver transplantation models were established by two cuff tech-nique. Three days after the orthotopic liver transplantation, the livers were resected, then the dendritic cells were separated and purified. The phenotypes [OX62, major histocompatibility complex Ⅱ (MHC-Ⅱ) and CD86] of dendritic cells were examined by FACS, the expression of IL-12, IL-1, and TNF-a mRNA by RT-PCR, and the expression of Toll-like receptor 4 (TLR4) by Western blot. Results The dendritic cells treated with Sinomenine showed immature phenotypes. The expressions of MHC-Ⅱ and CD86 were significantly deceased. The expressions of IL-12, IL-1, TNF-a mRNA and TLR4 were low. Conclusions Sinomenine can significantly inhibit the maturity and immunologic function of dendritic cells after hepatic ischemia-reperfusion.
