China Journal of Chinese Materia Medica 2014;39(3):397-401
MicroRNA(miRNA) is involved in virtually all biologic processes, including cellular proliferation, apoptosis, and differentiation. Thus, miRNA deregulation often results in impaired cellular function and disease development, so miRNAs have potential therapeutic relevance. The elucidation of these processes regulated by miRNAs and the identification of novel miRNA targets in the pathogenesis of hypertension is a highly valuable and exciting strategy that may eventually led to the development of novel treatment approaches for hypertension. Several mechanisms have been implicated in the pathogenesis of hypertension: overactivation of therenin-angiotensin-aldosterone system (RAAS), dysfunction of the vascular endothelium, damnification of vascular smooth muscle. To maintain and restore target organ expression of miRNA stable may be a new strategy for treatment of hypertension. The article reviews pathogenesis of miRNA and hypertension, researches of miRNAs as biomarker and therapeutic target, discusses advances in miRNA-based approaches that may be important in treating hypertension.

Objective To investigate the effect of potassium iodide on the incidence of sporotrichosis, and provide the theoretical basis for the prevention of sporotrichosis. Methods Experimental mice were divided into 4 groups, group 1~3 were fed with water containing different doses of potassium iodide, and no potassium iodide was given to the control group. After two weeks, the fungi were inoculated intraperitoneally to all the mice. In the third to the eighth weeks, the mice were sacrificed every week in batches. The infection, pathological changes and mycological findings of the mice were observed. Results The incidence rate of experimental sporotrichosis was significantly decreased in the mice (P

Chinese Journal of Laboratory Medicine 2015;38(4):220-222
Neonatal inherited metabolic diseases are a group of metabolic disorders caused by singe gene defect to cause a series of clinical symptoms.Neonatal dried blood spots have the advantages of simple preparation, safety, good stability, and show strong practicability in different screening methods for inherited metabolic diseases. With the development of screening methods, more and more diseases could be diagnosed by screening. The emergence of tandem mass spectrometry and molecular biological techniques promote the newborn screening and automation for inherited metabolic disease effectively.Inherited metabolic diseases induce great harm to the newborn, which could cause not only system organs damage, but also lead to death. Therefore, early screening is important for patients' prognosis.

Objective To investigate the identification of MiR-221 regulates apoptosis by targeting PUMA in lung cancer. Methods The express levels of miRNAs that collected from lung cancer tissue and lung benign lesion were tested by the particular kid they made, analyzed the correlation between individual miRNA.Results 9 6 possible regulated PUMA related miRNAs had been screened through the bioinformatics database, then customized it into the special kit to test the expression with real time fluorescent PCR.The results showed that 10 types miRNAs were up expression in lung cancer tissue,miR-NA221 were selected for deep analyze. Results with rank sum test showed statistical significance (P<0.05)in lung cancer and para cancer tissue. There were statistically significant differences in the expression between metastasis and nonmetastasis (P<0.05),but had nothing to do with the pathological types. Conclusion Real time fluorescent PCR test showed several types of PUMA related miRNAs up-expressed in tissue lung, which indicate miRNA have relationship with PUMA in gene express regulation and cancer generation and development.Pression situation and clinic pathological parameters.Expression of miRNA-221 in lung cancer was associated with tumor metastasis.

Objective The purpose of this study was to investigate the effect of different level of sex steroids (?-estradiol and progesterone) on the proliferation of uterus endometrial epithelial cells. Methods Rabbit uterus endometrial epithelial cells were isolated by digestion of trypsin and centrifugation. The specificity and homogeneity of the endometrial epithelial cells cultured in media containing ?-estradiol and progesterone were determined by immunofluorescence staining of cytokeratin monoclonal antibody and the flow cytometric analysis. The different concentration of progesterone (P4) and ?-estradiol (E2) were added in media, the proliferative capability of the endometrial epithelial cells cells in vitro was determined indirectly by MTT assay. Results High purity endometrial epithelial cells (96%) were isolated using centrifugation method. The endometrial epithelial cells were examined by immunofluorescence staining. The endometrial epithelial cells were positive stained by anti-cytokeratin antibody. Immunofluorescence staining demonstrated that cytokeratin was detectable in the cytoplasm of epithelial cells. The cell growth curve showed that the proliferation of endometrial epithelial cells was not significantly changed by the treatment of E2 (100nmol/L) or P4 (100nmol/L) individually, but a combination of 100nmol/L E2 and 10nmol/L P4 could promote its proliferation greatly (P
