1.Construction of eukaryotic expression vectorinsertedby MafA and its expression in HepG-2 cells

Objective:To construct eukaryotic expression vectors pEGFP-N2/MafA and pEGFP-C1/MafA,encoding the mouse musculus v-maf musculoaponeurotic fibrosarcoma oncogene family,protein A(MafA)gene,for the further research of its expression in HepG-2 cells.Methods:pRNA of the mouse MafA gene was distilled by RT-PCR from the mouse,and inserted into Hind Ⅲand SalⅠrestriction sites by PCR,then cloned into pEGFP-N2 and pEGFP-C1 vectors to obtain plasmids pEGFP-N2/MafA and pEGFP-C1/MafA.Human liver cancer cells(HepG-2) were transfected with formed plasmids by means of lipidosome.The MafA-EGFP fused protein was viewed directly with fluorensce microscope,and expression of MafA and insulin Ⅱ was detected by RT-PCR.Results:The mouse MafA gene was amplified through RT-PCR and successfully cloned into transfer vectors.The favorite gene sequence could be expressed and was completely consistent with that reported in genebank,but the insulin Ⅱ gene expression was not detected.Conclusion:The recombinant plasmids pEGFP-N2/MafA and pEGFP-C1/MafA were successfully constructed.But the insulin Ⅱgene was not expressed.
2.The pharmacokinetics of ropivacaine for single sciatic nerve block in dogs

Objective To evaluate the pharmacokineties of ropivacaine for single sciatic nerve block in dogs.Methods Twelve healthy adult mongrel dogs weighing 14-17 kg were randomized to receive 0.5% ropivaeaine 10 mg/kg or 20 mg/kg for single sciatic nerve block(n=6 each).ECG,BP and HR were monitored and recorded during anesthesia.Blood samples were obtained from femoral artery before ropivacaine injection (baseline)and at 10,20,30,40,60,90,120,150,180,240,360 and 720 min after ropivacaine injeetion for determination of plasma ropivaeaine concentration(by reversc-phasc high performance liquid chromatography).Arterial blood samples were also taken when adverse reactions occurred.The pharmacokinetic parameters were eMeulated with DAS 1.0 software package.Results The concentration-time curve of ropivacaine for single sciatic nenre block was fitted to two-compartment open model in both groups.The peak plasma concentration of ropivaeaine was significantly lower in 10 mg/kg group than in 20 mg/kg group.Two dogs developed convulsion in 20 mg/kg group.The plasma ropivaeaine concentration was 12.56 and 13.67 mg/L respectively during convulsion.Conclusion Pharmaeokinetic profile of ropivaeaine for single sciatic nerve block is best described by two-compartment model.Bopivaeaine 20 mg/kg for sciatic nerve block can hardly be tolerated by dogs.
3.Effects of Nimodipine treatment on the expression of neural cell adhesion molecule after the ischemic brain injury in rats

6.A comparative study of medium-term outcomes of type C distal radial fractures treated with volar locking compression plate and external fixation

Objective To compare the medium-term clinical effects of oblique T-shaped volar locking compression plate (LCP) and external fixation (EF) treating type C distal radial fractures.Methods Data of 60 patients with type C distal radial fractures who underwent surgery in our hospital from May 2010 to November 2012 were retrospectively analyzed.36 patients received open reduction with LCP fixation (LCP group),among which there were 21 males and 15 females,and there were 20 left wrists and 16 right wrists with an average age of 45.94±16.29 years old (range,17-80 years old).24 patients received close reduction and external fixation (EF group),among which there were 16 males and 8 females,and there were 15 left wrists and 9 right wrists with an average age of 43.63±14.55 years old (range,16-79 years old).The clinical results of wrist joint were assessed by range of wrist activity,VAS,Gartland-Werley wrist score,postoperative radial tilt angle,radial inclination angle and radial height.Results All the 60 patients were followed up successfully.The LCP group was followed up for 24 to 55 (42.03 ± 8.62) months.The EF group was followed up for 24 to 54 (37.24±9.77) months.In the LCP group,the average VAS score was 1.28±0.78,Gartland-Werley score 2.28±0.78,wrist flexion angle 50.89°±5.53° and wrist dorsiflexion angle 50.69°±5.38°.In the EF group,the average VAS score was 1.38±0.71,Gartland-Werley score 2.92± 1.69,wrist flexion angle 50.96°±5.01° and wrist dorsiflexion angle 51.08°± 5.73°.No statistically differences were found in wrist joint function between two groups.In LCP group,radiological evaluation 2 years after operation showed that the average radial tilt angle was 10.69°±2.29,radial inclination angle 22.53°± 1.95° and radial height 10.63±1.14 mm.In the EF group,radial tilt angle was 6.63°±2.76°,radial inclination angle 16.96°±4.41° and radial height 9.06±0.98 mm.Significant differences were found in radial tilt angle,radial inclination angle and radial height.No statistically differences were found in postoperative complication rates.Conclusion Locking compression plate (LCP) fixation and external fixation (EF) can obtain similar satisfactory medium-term clinical outcome in treating type C distal radial fractures.Comparison with EF,LCP can achieve anatomic reduction under direct vision,and its radiological assessments including radial tilt angle,radial inclination angle and radial height were significantly better than external fixation.
7.Changes of Protein Metabolism in Rat Liver after Scaldingand Effects of Insulin

Full thickness scalding of 37% TBSA was inflicted io rats. Three days later, the liver of the animals was isolated and the changes of the protein metabolism in the liver and the effects of insulin on the changes were evaluated.It was found that the protein and DMA levels of the liver showed no obvious changes after scalding, but the RNA content increased by 24% and the rate of leucine incorporation by 31%. After the addition of insulin, the leucine content increased by 23.% and 25.8% in the controls and the injured animals respectively. The results indicate that the rate of protein synthesis in the liver of the scalded animals increases, which is likely to be related to the increase of RNA content, and the liver protein synthesis remains to be sensitive to insulin.
8.Changes of protein Metabolism in Rat Soleus Muscle after Burus and Effects of Leucine and Insulin

This paper is to report the changes of the protein metabolism in the rat soleus muscle in vitro after 37% TBSA full-thickness burns covering one hindlimb and the effects of leucine and insulin on the burned muscle.It was found that there werea significant decreases of the protein content and marked exaggeration of the rates of tyrosine incorporation,tyrosine release and tyrosine net release in the burned muscle.No changes of the above-mentioned parameters could be revealed in the muscle of the unburned limb and the controls.Elevation of leucine concentration in the medium could not improve the protein metabolism of the burned muscle.0.1u/ml of insulin could significantly stimulate the tyrosine incorporation in the muscle of the unburned limb and the controls,but no similar effect was observed on the burned muscle.The results indicate that increased protein degradation of the local burned muscle is responsible for the postburn negative nitrogen balance;administration of leucine alone cannot improve the protein metabolism in the local burned muscle;and the sensivity of the burned muscle to insulin is suppressed.
9.Dynamic MR imaging of intrahepatic peripheral cholangiocarcinoma

Objective To evaluate the dynamic MR imaging findings of peripheral cholangiocarcinoma (PCC) of the liver. Methods Seventeen patients with PCC underwent dynamic MRI during the early, middle, and delayed phase after contrast medium administration. Enhancement pattern of the tumor, the wedge-like enhancement area peripheral to the tumor, dilatation of the intrahepatic biliary ducts, vascular involvement, and extrahepatic tumor invasion were evaluated. Results Early peripheral enhancement with progressive and concentric filling by contrast medium was the most frequent enhancement pattern (9 patients). A wedge-like enhancement area was observed in 10 patients. Ductal dilatation was found in 11 patients. Vascular involvement and extrahepatic invasion were seen in 8 and 2 patients, respectively. Conclusion Enhancement pattern is relative to the character of pathology (quantity and distribution of cell and fibrosis). Early peripheral enhancement with progressive and concentric filling by contrast medium may be specific for PCC.
