1.Comparison of orthotopically implanted mouse models of primary breast cancer established by different methods

Objective To analyze the biological characteristics and morphologic features between the mouse models using MDA-MB-231 cells established by injecting tumor cell suspension or implanting tumor tissues. Methods Twenty BALB/c mice were randomly divided into two groups,with 10 in each group.The orthotopic models of breast cancer were established by injection of tumor cell suspension or implantation of tumor tissues in the fat pad of right second breast in the nude mice of the two groups,respectively.The time of tumor formation,success rate,tumor growth rate,shape,central tumor necrosis,blood vessel distribution and tumor ulceration were compared between the two groups of models.The histological features of the tumors were also observed. Results There were no significant difference in the success rate between the two methods.In the group of tumor tissues implantation,the tumors formed in an earlier time with various shape configurations,and they were hypovascular and vulnerable to central necrosis with the resultant superficial ulcerations.However,in the group with injection of tumor cell suspension,the tumors formed later with a regular shape,and the superficial ulceration was rare and not vulnerable to central tumoral necrosis since the tumors were abundant in blood vessels resulting in adequate nourishment.There were no significant difference in the pathological findings between the two groups. Conclusion The nude mouse models of breast cancer can be successfully established by both tumor tissues implantation and tumor cell suspension injection,with identical biological characteristics and fairly different morphological features.The investigators are well advised to choose the more appropriate one according to the purpose of the experiments.
2.Serum and tissue levels of matrix metalloproteinase-1 and its tissue inhibitor in patients with intervertebral disc degeneration

BACKGROUND:Matrix metaloproteinases are now generaly considered to be able to degrade al extracelular matrices. Hypersecretion of matrix metaloproteinases or reduction in tissue inhibitors of matrix metaloproteinases leads to destruction of the dynamic balance of extracelular matrix. OBJECTIVE: To elucidate the role of matrix metaloproteinase-1 and tissue inhibitor of matrix metaloproteinase-1 in the pathogenesis and progression of intervertebral disc degeneration. METHODS:A total of 60 patients with intervertebral disc degeneration were included. Mild, moderate, and severe degeneration signals appeared on MRI imaging of the patients. Meanwhile, 20 patients with vertebral fracture, mainly cervical spine fracture, were selected as the control group. Venous blood samples were colected before the surgery; the intervertebral disc specimens were sequentialy colected. RESULTS AND CONCLUSION: Serum and tissue levels of matrix metaloproteinase-1 in patients with intervertebral disc degeneration were significantly increased compared with the control group (P < 0.05), and furthermore those were significantly increased in patients with severe disc degeneration compared with patients with mild and moderate disc degeneration (P < 0.05). However, serum and tissue levels of tissue inhibitors of matrix metaloproteinases did not differ significantly between the disc degeneration and control groups (P > 0.05). These results indicate that hypersecretion of matrix metaloproteinase-1 occurs in patients with intervertebral disc degeneration; however, the expression of tissue inhibitor of matrix metaloproteinase-1 is not correlated with intervertebral disc degeneration.

4.Diagnose nose and treatment of testicular endodermal sinus tumor

Objective To investigate the diagnosis and treatment outcomes of testieulax endodermal sinus tumor. Methods Twenty-four cases diagnosed with testieular endodermal sinus tumors from November 1996 to April 2007 were retruspeetively reviewed. Eighteen patients presented with stage Ⅰ disease, 4 presented with stage Ⅰ , and 2 presented with stage Ⅲ. Inguinal radical or-chieetomy were performed in all patients. Retroperitoneal lymph node dissection was performed in 8 ea-ses. Results The histological structures were rather complicated in 24 tumors. Twenty-three eases (96%) were found with reticular pattern, 22(92%) with hyaline body, 20(83%) with glandlike structure, 16(67%) with Shiller-Dural body, and 13(54%) with solid formation. The former 3 kinds were regarded as the main diagnostic criteria. Twenty-one eases were followed up for 20 months to 12 years. During the follow-up, 2 patients died of the disease. Condesions Early diagnosis and combi-nation therapy, including radical orehieetomy and chemotherapy, are the keys to improve the curative effect of testieular endodermal sinus tumors. Active surveillance of AFP is critical for monitoring the recurrence and metastasis of this tumor.
5.Effect of ephedrine on expression of eotaxin in human bronchial epithelial cells stimulated by tumor necrosis factor-α

Objective To observe the effect of ephedrine on the expression of eotaxin in human bronchial epithelial cells (16HBE) stimulated by tumor necrosis factor‐α(TNF‐α) and to explore the mechanism of Chinese medicine ephedra in treating asthma .Methods The in vitro cultured 16HBE were randomly divided into the control group ,TNF‐αstimulation group(TNF‐α20 ng/mL) and TNF‐αplus ephedrine group (TNF‐α20 ng/mL plus ephedrine 300 μg/mL) .Three complex holes in each group were set to culture for 18 h ,the eotaxin mRNA expression was measured by real time fluorescent quantified PCR and protein level was detected by immunocytochemical stain and Western blot .The eotaxin concentration in cells culture supernatant was quantified by ELISA .Results Compared with the the control group ,the expression level of eotaxin mRNA and protein ,and the concentration of eotaxin in cell culture supernatant in the TNF‐α stimulation group were increased obviously ,there being statisticaly significant difference between them(P<0 .01);however ,all above these parameters in the TNF‐αplus ephedrine group showed decreased obvi‐ously as compared with the TNF‐αgroup ,the difference between them was statistically significant (P<0 .01) .Conclusion Ephed‐rine can inhibit the expression and secretion of eotaxin in TNF‐α induced 16HBE inflammatory model ,which may be one of the mechanisms of Chinese medicine ephedra in treating asthma .
6.Time-and dose-effect of Gardenia Jasminoides extract on hepatoxicity in rats

AIM To observe the relationship between dose effect and time effect on hepatoxicity of Gardenia jasminoides Ellis extract in rats.MOTHODS Wistar rats were divided into four groups:low,middle and high (3,10,and 30 g/kg) dose of G.Jasminoides groups (administrated by gavage),and the normal control group were orally given deionized water.All rats were observed daily during the administration period.On the 7th,14th,28th day after the administration,blood samples were collected;serum alanine transaminase (ALT),aspartate transaminase (AST),alkaline phosphatase (ALP),glutamic dehydrogenase (GLDH) activity and total bile acid (TBA) and total bilirubin (TBIL) were determined.The livers were weighed and the liver index was calculated.HE staining and observation of histopathological changes in the structure of liver tissue under light microscopy were performed.RESULTS After the 7th day of administration,the rats in high dose group showed lower food consumption and slowly increased body weight.Serum ALT,AST,ALP,TBA,TBIL and GLDH in rats from high dose group were significantly higher than those in the normal control group.The liver index of rats in the middle and high dose groups was significantly increased than that in the normal control group.After the 14th day of administration,serum ALT,TBA and TBIL in rats from the high dose group were significantly higher than those in the normal group.The liver index of rats in the middle and high dose groups was significantly increased than that in the normal control group.After 28th day of administration,serum ALT and TBA in the rats from the high dose group,TBIL and GLDH in rats from the middle dose group,and GLDH in rats from the low dose group were significantly higher than those in the normal control group.The liver indexes of rats in all dose groups were significantly increased than those in the normal control group.After the 7th,14th or 28th day of the treatment,histopathological changes such as the liver cell hypertrophy,interlobular bile duct hyperplasia,and inflammatory cell infiltration appeared in the middle and high dose groups.CONCLUSION The high dose of G.jasminoides can induce and increase liver toxicity with the increase in dose,but at high dose level,liver toxicity does not increase with time.
7. Extraction of ginger oleoresin from Zingiberis Rhizoma Recens and inclusion of ginger oleoresin with β-cyclodextrin

OBJECTIVE: To optimize the extraction and inclusion processes of ginger oleoresin from Zingiberis Rhizoma Recens. METHODS: Supercritical carbon dioxide extraction was selected to extract ginger oleoresin from Zingiberis Rhizoma Recens. The extraction condition was optimized by orthogonal experiment. The inclusion method, the ratio of ginger oleoresin to β-cyclodextrin and inclusion time were studied as factors of inclusion process. RESULTS: The optimized extraction technology was as follows: extraction temperature was 50℃, extraction pressure was 25 MPa, separation pressure was 9 MPa and extraction time was 90 min. The optimal inclusion method was triturating inclusion, with the ratio of ginger oleoresin to β-cyclodextrin of 1:8 and inclusion time of 60 min. CONCLUSION: The optimized technology of extraction and inclusion is stable and feasible, and can be used for the extraction and inclusion of ginger oleoresin.
8.Chronic tibial defection in children treated with grafting of fibula with vessel pedicles and Lebone under external fixators

Objective To report one stage treatment of chronic tibial defection in children. Methods 19 children with chronic tibial defection were treated in our department when their infection was at the stage of latency. First their focuses of infection were cleared away. Next a piece of fibula with vessel pedicles whose length was equal to that of the tibial defect was taken as the support to restore the length of the tibia. Then the external fixators were applied and a mixture of antibiotic and Lebone powder was used to fill the defect. Results In 15 cases the incision healed at one stage, slight infection occurred in 4 cases whose incisions healed after treatment of 3 to 4 weeks. Follow-ups of 0.5 to 1.5 years showed that no infection occurred again, bone defects healed well and limb function was satisfactory. Conclusion To treat chronic tibial defection in children at one stage, combined therapy should be applied. Grafting of fibula with vessel pedicles and Lebone under external fixators is a good one, because it can shorten the treatment time at no cost of satisfactory effects.
9.Placing double J stent using a ureteroscope in early management of ureterovaginal fistula

Objective To evaluate the clinical effect of placing double J stent using a ureteroscope in early managing ureterovaginal fistula.Methods Twenty-eight patients cases with ureterovaginal fistula from 2002 to 2008 were treated early with placing double J stent using a ureteroscope and the clinical data were reviewed.Results Twenty-two of 28 cases were treated and double J stent was placed in them by a uretero-scope and 75% (21/28)of cases were cured.Four of 21 cases were treated twice by a ureteroscope and were cured finally.7 cases with failure ureterovaginal treatment underwent ureterocystostomy and were cured.The follow-up from 6 months to 33 months (average 10.1±6.4 months)showed that all of the 28 cases had been cured and had no urinary fistula.Conclusion Placing double J stent using a ureteroscope is the first choice of operative procedure for the early treatment of ureterovaginal fistula.
10.Multiband mucosectomy in treatment of early cancer and precancerous lesions of esophagogastric junction

Eighteen patients with early cancer and precancerous lesions of esophagogastric junction underwent multiband mucosectomy (MBM) in Yangzhou First People's Hospital from January 2013 to February 2014.The clinicopathological data of patients were analyzed retrospectively and the short-term efficacy and safety of MBM were evaluated.Operations were successful in all 18 cases.The mean operative time was 35.8 min.Intraoperative bleeding occurred in 3 cases and successfully managed by endoscopic hemostasis.Small perforation occurred in 1 patient and was closed by metal clips.Pathological examination showed mucosal cancer in 5 cases,high-grade intraepithelial neoplasia in 8 cases and low-grade intraepithelial neoplasia in 5 cases.No relapse of cancer was found during follow-up.Results indicate that MBM is an effective and safe method in treatment of early cancer and precancerous lesions in the esophagogastric junction.
