4.The relationship between human adiponectin gene polymorphism of Guangxi Zhuang ethnic group and gestational diabetes mellitus
Journal of Chinese Physician 2014;16(9):1221-1223
Objective To investigate the association of single nuchotide polymorphism (SNP) + 45 T/G and + 276 G/T in the adiponectin gene with type 2 diabetes in Guangxi Zhuang pregnant women. Methods Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method was used to determine the distributions of allele and genotype frequencies of SNP + 45T/G polymorphism and SNP + 276 G/T polymorphism in adiponect in gene in 98 type 2 diabetic patients and 135 normal control subjects. Results The TG,GG genotype frequencies of the SNP +45T/G were higher in gestational diabetes mellitus (GDM) than those in controls (P < 0.05).The G-allele frequency was significantly higher than those in controls (P < 0.05).The distribution of genotype and allele frequencies of the SNP + 276 G/T polymorphism was not different in diabetes and control groups (P > 0.05).Women carrying the TG and GG allele of SNP +45T/G showed higher TG (P <0.05),and lower APN levels than those with TT genotype (P <0.05).Conclusions G allele of SNP +45T/G was related to GDM possibly.T→G variation might involve in the occurrence of GDM by reducing serum diponectin levels.Monitoring this site in pregnant women could predict the risk of GDM.
5.Cytotoxicity evaluation of nano-Ag-SiO2urethral catheter for external urethral mucosa cells
Chinese Journal of Tissue Engineering Research 2014;(16):2550-2556
BACKGROUND:Nano-Ag is increasingly used as antibacterial materials in medical products due to its excelent antibacterial activity. Antibacterial activated nano-Ag-SiO2 OBJECTIVE: To evaluate the biosecurity of nano-Ag-SiO urethral catheter can reduce the incidence of catheter-associated urinarytract infections obviously. As more and more nano-Ag medical products have been developed, their biological security is becoming a focused issue. 2 urethral catheter through comparing the active influence of nano-Ag-SiO2 METHODS: Mechanical separation and enzymatic digestion were used to isolate and culture urothelial mucosa of benign prostatic hyperplasia patients to make cellsuspension after proliferationin vitro. The medium was replaced by nano-Ag-SiO urethral catheter and ordinary urethral catheter extracts onin vitro cels of human prostatic urethra epithelial cels in different periods. 2 and general catheter tube extracts, respectively. MTT assay was performed to quantify thein vitro cytotoxicity of two extracts on human urethra epithelial cels which were cultured for 2, 5 and 7 days. Absorbance values of nano-Ag-SiO2 RESULTS AND CONCLUSION:The cytotoxicity of extract of nano-Ag-SiO and general urinary catheter groups were measured by enzyme-linked immunosorbent assay to calculate the relative growth rate, and toxicity evaluation was compared and evaluated. Under an inverted microscope, cellmorphology, growth and proliferation were observed. 2 catheter was 1 grade, and that of general medical catheter extract was 0 to 1 grade. There was no statistical difference in group effect of relative proliferation rate (F=0.544,P=0.475); there was statistical difference in time effect of relative proliferation rate (F=3.031,P=0.086); no statistical difference was found in the interaction between group and time (F=0.130, P=0.879). These findings indicate that nano-Ag-SiO2 catheter has no or low cytotoxicity effect on cellgrowth and proliferation, which is consistent with the biological evaluation standards of medical devices.

Chinese Journal of Clinical Infectious Diseases 2014;7(2):157-160
Objective To investigate the virologic responses to pegylated-interferon alfa-2a (PegIFNα-2a) plus ribavirin therapy for cryoglobulinemia positive chronic hepatitis C in patients with hepatitis C virus (HCV) genotype 1 b.Methods Clinical data on 60 chronic hepatitis C patients with HCV genotype 1b were collected from Huzhou Central Hospital during January 2008 and December 2012.The patients were divided into cryoglobulinemia positive group (n =22) and cryoglobulinemia negative group (n =38),and both received 48-week PegIFNα-2a (180 μg) plus ribavirin treatments. For those who could not tolerate the treatment,PeglFNα-2a was reduced to 135 μg from 12th week.Rapid virologic response (RVR),end of treatment virologic response (ETVR) and sustained virologic response (SVR) in two groups were observed and compared using Chi-square test. Results RVR,ETVR and SVR in cryoglobulinemia positive group were 59.1% (13/22),81.8% (18/22) and 31.8% (7/22),while those in cryoglobulinemia negative group were 55.3% (21/38),76.3% (29/38) and 60.5% (23/38).There was significant difference in SVRs between two groups (x2 =4.674,P < 0.05).For those completed full course of antiviral therapy with 180 μg PegIFNα-2a,the RVR and SVR in cryoglobulinemia negative group tended to be higher than those in cryoglobulinemia positive group, but the differences were not statistically significant (x2 =1.524 and 1.009,P > 0.05).Serious adverse reactions were observed in 9 (40.9%) patients of cryoglobulinemia positive group, while in 11 (28.9%) patients of cryoglobulinemia negative group (x2 =0.887,P > 0.05).Conclusion Cryoglobulinemia may not have considerable impact on virologic responses to PegIFNα-2a plus ribavirin therapy in patients with chronic genotype 1b hepatitis C.
7.Prognostic evaluation of microRNA-192 levels in diabetic nephropathy patients
Chinese Journal of Postgraduates of Medicine 2013;36(34):35-39
Objective To explore the relevance of the microRNA-192 (miR-192) levels and prognosis in patients with diabetic nephropathy (DN).Methods A total of 59 cases with duration of type 2 diabetes mellitus (T2DM) ≥10 years from January 2010 to July 2011 were retrospectively admitted. The miR-192 levels were determined after admission, and other relevant clinical data were collected. And the patients were followed-up for 24 months.According to the patient's prognosis, the patients were divided into DN group(20 cases) and non-DN group(39 cases),and the relevant information was analyzed in two groups. Results The miR-192 levels in DN group were lower than those in non-DN group[(4.47 ± 2.27) μ g/L vs.(7.28 ± 2.39) μ g/L],and there was significant difference (P < 0.05).Multivariate Logistic regression analysis showed that hypertension history, smoking history, high glycosylated hemoglobin, low fasting C-peptide,low 24 h urine protein, low miR-192 level, high body mass index was the independent risk factors of prognosis of T2DM (P < 0.05).The area under the receiver operating characteristic (ROC) curve was 0.697 in miR-192 levels individual predicting progression to DN stage,0.823 in six others without miR-192.After joining seven indicators forecast area under the curve increased significantly(0.856,95% CI:0.832-0.904).The miR-192 levels 5.14 μ g/L was the best predictive value to the critical value of community, and the patients were divided into miR-192 < 5.14 μ g/L group (28 cases) and miR-192 ≥ 5.14 μ g/L group (31 cases). The number of progression to DN within 24 months in miR-192 < 5.14 μg/L group was more than that in miR-192 ≥5.14 μ g/L group (16/28 vs.4/31,P < 0.05).Conclusions A reduced level of miR-192 is an independent risk factor that can lead to poor prognosis in patients with DN,and can predict adverse outcomes in patients with T2DM effectively.
8.THE IMPLICATION OF PLC-?2 IN GASTRIC CANCER MGC80-3 CELLS TREATED BY TPA
Acta Anatomica Sinica 2002;0(05):-
Objective To study the implication of phosphoinositide-specific phospholipase C?2(PLC-?2) in gastric cancer MGC80-3 cells treated by TPA.Methods The effect of 12-O-tetradecanoylphorbol-1,3-acetate(TPA) on gastric cancer MGC80-3 cells was detected and analyzed by DAPI staining under fluorescence microscope. The effect of TPA on the expression level of PLC-?2 protein was detected by Western blotting, when nuclear and cytoplasmic protein fractions were prepared through lysis of cell and centrifugation.Localization and translocation of PLC-?2 were observed under laser scanning confocal microscope with immune-fluorescence technique. After MGC80-3 cells were pretreated by U73122(PLC-?2 inhibitor),the effect of TPA on PLC-?2 was detected by Western blotting and laser scanning confocal microscope with immunefluorescence technique, and the effect of U73122 on MGC80-3 cells followed by TPA was observed and analyzed by DAPI staining under fluorescence microscope. Results TPA induced the apoptosis of gastric cancer MGC80-3 cells.Meanwhile,TPA increased the expression level of PLC-?2 protein and induced its translocation from cytoplasm to nucleus.U73122 inhibited the effect of TPA on the expression level of PLC-?2 protein, but it did not affect on the cell apoptosis induced by TPA.However, the translocation of PLC-?2 protein induced by TPA was not inhibited by its inhibitor(U73122).Conclusion Although TPA could increase the expression level of PLC-?2 protein,it is not directly related to the cell apoptosis induced by TPA.However, its translocation might be related with the apoptosis of gastric cancer.
9.Protection of indole-3-carbinol against ethanol-induced hepatotoxicity in precision liver-cut slices of rats
Chinese Pharmacological Bulletin 1986;0(06):-
AIM To study the hepato-protection of indole-3-carbinol (I3C), one of glucosinolate compounds in cruciferous vegetables, on ethanol-induced hepatotoxicity. METHODS Ethanol 50 mmol?L -1 was used to induce hepatotoxicity in precision liver-cut slices of rats. The release of glutamic pyruvic transaminase(GTP), glutamic oxaloacetic transaminase (GOT), lactate dehydrogenase (LDH) and glutathione S-transferase (GST) into culture medium, and aniline hydroxylase (ANH) and alcohol dehydrogenase (ADH) in cytosol of hepatocytes in the precision liver-cut were measured with different doses of I3C. The histopathological observation of liver slices was also done to evaluate the effect of I3C. RESULTS The release of GTP, GOT, GST and LDH in the medium increased significantly when the slices were treated with 50 mmol?L -1 ethanol for 4 h. I3C at concentrations of 100~400 ?mol?L -1 effectively reduced the release of GTP, GOT, GST and LDH in the medium of ethanol-treated slices. Meanwhile, the activities of cytosol ANH and ADH decreased to the normal levels. The histopathological changes of liver slices induced by ethanol were lessened by I3C treatment. CONCLUSION I3C can markedly protect rat liver slices from ethanol-induced hepatotoxicity, which partly results from the change of ethanol metabolism in liver.
10.Comparison of the effect of different methods of anesthesia on cerebral autoregulation in patients undergoing neurosurgery
Chinese Journal of Anesthesiology 2011;31(2):144-146
Objective To compare the effect of different methods of anesthesia on cerebral autoregulation in patients undergoing neurosurgery. Methods Sixty-nine ASA Ⅱ orⅢ patients with brain tumor, aged 23-62 yr, scheduled for neurosurgery under general anesthesia, were randomly divided into 3 groups ( n = 23 each) : propofol-remifentanil group (group PR), sevoflurane-remifentanil group (group SR) and propofol-sevoflurane-remifentanil group (group PSR) . Anesthesia was induced with target-controlled infusion (TCI) of propofol (target plasma concentration3 μg/ml, PR and PSR groups) or inhalation of 8% sevoflurane (group SR) and iv injection of remifentanil 1 mg/kg and atracurium 0.5 mg/kg. The patients were mechanically ventilated after tracheal intubation. PETCO2 was maintained at 32-35 mm Hg. Anesthesia was maintained with TCI of propofol (target plasma concentration 2.0-3.5 μg/ml) in group PR, with inhalation of 1.5%-2.5% sevoflurane in group SR, with TCI of propofol (target plasma concentration 1.5-3.0 μg/ml) and inhalation of 1% sevoflurane in group PSR, and with TCI of remifentanil (target plasma concentration 2.0-4.5 ng/ml) and iv infusion of atracurium at 6 μg · kg-1 · min-1 in all groups. Auditory evoked potential index was maintained between 40-45. The middle cerebral artery time-average peak flow velocity was recorded before induction (baseline) , immediately after intubation, immediately before craniotomy and at the beginning of skin suture. The unilateral carotid artery was compressed for 7 s at the corresponding time points mentioned above. The transient hyperemic response ratio (THRR) was calculated to reflect cerebral autoregulation. Results Compared with the baseline value at T0, THRR was significantly increased at T2in group PR and decreased at T2,3 in group SR (P <0.05) ,while no significant change was found in THRR at T1-3in group PSR (P >0.05). The THRR was significantly lower in SR and PSR groups than in group PR, and higher in group PSR than in group SR ( P < 0.05). Conclusion Propofol-remifentanil anesthesia can improve cerebral autoregulation, sevoflurane-remifentanil anesthesia can reduce cerebral autoregulation, and propofol-sevofluraneremifentanil anesthesia exerts no effect on cerebral autoregulation in patients undergoing neurosurgery.
