1.Chemical constituents from roots of Andrographis paniculata.

To investigate the chemical constituents of the roots of Andrographis paniculata, 28 compounds were isolated and identified from the 80% ethanol extract. There are 20 flavonoids: 5, 5'-dihydroxy-7, 8, 2'-trimetroxyflavone (1), 5-hydroxy-7, 8, 2', 6'-tetramethoxyflavone (2), 5, 3'-dihydroxy-7, 8, 4'-trimethoxyflavone (3), 2'-hydroxy-5, 7, 8-trimethoxyflavone (4), 5-hydroxy-7, 8, 2', 3', 4'-pentamethoxyflavone (6), wightin (7), 5, 2', 6'-trihydroxy-7-methoxyflavone 2'-O-beta-D-glucopyranoside (8), 5, 7, 8, 2'-tetramethoxyflavone (10), 5-hydroxy-7, 8-dimethoxyflavanone (11), 5-hydroxy-7, 8-dimethoxyflavone (12), 5, 2'-dihydroxy-7, 8-dimethoxyflavone (13), 5-hydroxy-7, 8, 2', 5'-tetramethoxyflavone (14), 5-hydroxy-7, 8, 2', 3'-tetramethoxyflavone (15), 5-hydroxy-7, 8, 2'-trimethoxyflavone (16), 5, 4'-dihydroxy-7, 8, 2', 3'-tetramethoxyflavone (17), dihydroneobaicalein (18), andrographidine A (19), andrographidine B (20), andrographidine C (21) and 5, 2'-dihydroxy-7, 8-dimethoxyflavone 2'-O-beta-D-glucopyranoside (22); three diterpenoids: andrograpanin (23), neoandrographolide (24) and andrographolide (25); two phenylpropanoids: trans-cinnamic acid (26) and 4-hydroxy-2-methoxycinnamaldehyde (5); and oleanolic acid (9), beta-sitosterol (27) and beta-daucosterol (28). Compound 1 is a new flavone, compound 4 is a new natural product, compounds 2, 3 and 5 were isolated from the Androggraphis genus for the first time and compounds 6-9 were isolated from this plant for the first time.
2.Regulation Value and Social Effect Analysis of Drug Proportion in the Hospital Management and Evalua-tion

OBJECTIVE:To evaluate the value and social effect of the regulation by the indicator of drug proportion. METH-ODS:The scientificity and rationality of pharmaceutical proportion and the regulation activity by the indicator of drug proportion was analyzed by analyzing the connotation and influential factors of drug proportion and the method of comparative study,qualita-tive analysis and quantitative analysis. RESULTS:There was great difference in connotation and value between international drug proportion and domestic one. Indicator of drug proportion had no direct relation to pharmaceuticals consumption control and rational drug use;the value and effect of the regulation activity in a hospital by the indicator of drug proportion were far from complete and required further research. CONCLUSIONS:Health administrative department and policy decisions department should further re-search and objectively evaluate the value and effect of the regulation activity in a hospital by the indicator of drug proportion. Whether the regulation activity in a hospital by the indicator of drug proportion carry out based on scientific verification;if neces-sary,the indicator of drug proportion should be cancelled or adjusted in hospital management and evaluation indicator system.
3.Application of the domestic rebounder tonometer in children

AlM:To evaluate the clinical application of the domestic rebound tonometer ( RBT ) in measuring intraocular pressure ( lOP) in healthy children.
METHODS:lOP measurement was measured in bilateral eyes of 108 cases with domestic RBT. Then the lOP for the left eyes of the children older than 13 years was obtained with Goldmann applanation tonometry ( GAT) .
RESULTS: Of the 108 children, 100 ( 92. 6%) had lOP measurement both eyes successfully. Of the 100 children, 24 were older than 13 years. The mean lOP values for the left eyes of the 24 children of RBT and GAT were (16. 0± 3. 0) mmHg (1kPa=7. 5mmHg) and (15. 7±2. 8) mmHg, respectively. There was no significant difference between the lOP of the two tonometers (P>0. 05). The lOP of RBT had a good liner relationship with that of GAT ( r=0. 849, P<0. 05). The lOP values were normally distributed for the both eyes. Mean lOP values were (16. 0±2. 8) mmHg and (16. 2 ± 2. 6 ) mmHg for the left eyes and right eyes, respectively. There was no statistically significant difference in mean lOP readings between them, which had a good liner relationship (r=0. 863, P<0. 05).
CONCLUSlON: Domestic RBT is a good instrument in detecting lOP in children and it seems to be very comfortable and easy when performing lOP measurement in children without an anesthetic.

The curative effects of multi-disciplinary and comprehensive treatments for colorectsl cancer have been widely recognized in recent years.Because of the diversification of colorectal cancer treatment and the relatively mature technology,the current treatment of colorectal cancer could embody the multi-disciplinary and comprehensive treatment.The curative effect of neo-adjuvant chemoradiotherapy has been validated by several recent clinical trials.Other treatment methods such as Chinese medicine,radiofrequency ablation and hepatic artery infusion therapy are also involved in the comprehensive treatment,which provide considerable curative effects.
6.Effect of Yiqi Yangyin Recipe and Its Different Assembling on Apoptosis Rate and Expression of Associated Protein in Acute Myeloid Leukemia Mice

Objective To observe the effects of Yiqi Yangyin Recipe (supplementing qi and nourishing yin) and its different assembling on apoptosis rate and associated protein expression of c-myc and survivin in mice, and explore its mechanism. Methods NOD-SCID mice were injected with l×106 KG1a cells from the tail vein after irradiation of 60Co. Model mice were randomly divided into control group, whole recipe group, strengthening body resistance group, eliminating pathogens group. The treatment groups were given correspongding administration, and control group was given equal volume of NS. Flow cytometry was applied to measure apoptosis rate. The immunochemistry method was used to detect the expression of c-myc, survivin. Results Apoptosis rate in all treatment groups were increased (P<0.01). The expression of c-myc and survivin in lymph node tissues in all treatment groups were significantly declined compared with control group (P<0.05). The effect of whole recipe group on the all indices was superior to strengthening body resistance group and eliminating pathogens group (P<0.05). Conclusion Yiqi Yangyin Recipe can decrease the expressions of c-myc and survivin, induce apoptosis in leukemia cells, therefore shows the therapeutic effect.
7.THE INFLUENCE OF DIFFERENT CULTURAL CONDITIONS ONEXPRESSION OF ScFv GENES AGAINST ALPHA-TOXIN OFCLOSTRIDIUM PERFRINGENS TYPE A

The ScFv gene containing Nco I and Bam HI was amplified by PCR, and inserted into the prokaryotic expression vector pHOG21 with Nco I and Bam HI digestion. After screening, high-level expression recombinant XL1-Blue (pHOG2E3) were obtained, and the different expression levels of ScFv were detected by SDS-PAGE and EUSA. When the strain was induced by 0.5 mmol/LIPTG and 0.4mol/L sucrose in the culture medium LB at 37℃ for 6 hours, the high level production of ScFv protein was obtained. The molecular weight of the expressed ScFv is 31, 000 D. The ScFv was mainly in the form of inclusion body, but it could also be in the culture medium and soluble periplasmic content. Above all, the ScFv protein could neutralize the phospholipase C activities of alpha-toxin of Gostridium perfrigens type A.
8.Experimental study on the formation of intravascular microthrombosis in brain after traumatic brain injury in rat

Objective To explore the formation of intravascular microthrombosis in the brain after traumatic brain injury in rat in order to study the potentially causal linkage between thrombosis and post-traumatic cerebral ischemia. Methods Lateral fluid percussion injury (FPI) model was reproduced in the present study to prodace acute brain injury in rats. The rats were sacrificed at 12, 24, 72 hours and 7days after traumatic brain injury. Sham-injured animals received identical craniotomy without FPI. Pathological studies were performed to study the distribution and severity of intravascular microthrombosis in the brain. 9.Primary culture and purification of embryonic dorsal root ganglion neurons

Objective To establish a simple and reliable method for primary culture and purification of embryonic dorsal root ganglion neurons. Methods Dorsal root ganglions (DRG) from rat embryos were isolated under microscope and dissociated in trypsin. The dissociated neurons were treated with 5-fluorouracil for purification. Immunocytochemistry was performed by double staining with anti-calcitonin gene-related peptide and anti-NF200 antibodies to evaluate the purification rate of DRG neurons. Results Isolated and cultured DRG neurons survived and grew nicely in the basis medium for neurons, and the purified rate of DRG neurons could reach to around 93%. Conclusion The above method is very useful to obtain dorsal root ganglion neurons and to purify the neuronal population.
10.Imipramine promotes regeneration of hippocampus neuron after traumatic brain injury in rat
