2.Effects of serum from chronic renal failure rats on endoplasmic reticulum stress and activation of activating transcription factor 4 of rat arterial smooth muscle cells cultured in vitro

Objective To investigate the effects of the serum from chronic renal failure (CRF) rats on endoplasmic reticulum stress (ERS) and activation of activating transcription factor 4 (ATF4) of rat arterial smooth muscle cells (ASMCs) cultured in vitro and explore the possible mechanism. Methods The rat model of CRF was established by 5/6 nephrectomy. ASMCs were incubated in the media with 10% or 20% serum of CRF rats cultured in vitro. ATF4 nuclear translocation were analyzed by immunofluorescence. GRP78 (glucose regulated protein 78), p-PERK (pancreatic ER kinase) and ATF4 proteins in response to the CRF serum in ASMCs were determined by Western blot. The ATF4-DNA binding activity was analyzed by electrophoretic mobility shift assay (EMSA). Results The CRF serum could significantly promote the expression of GRP78 of ASMCs in dose-dependent manner. Under the stimulation of 20% CRF serum, nuclear translocation of ATF4 in ASMCs was found. Compared with control serum group, the expression of ATF4 and p-PERK of CRF serum group was increased significantly (P<0.01). The CRF serum could increase the ATF4-DNA binding activity. Conclusion The CRF rat serum can effectively induce ERS of ASMCs and activate the pathway of PERK-ATF4.
3.Application of ARIMA model to forecasting bacillary dysentery in Jianghan District of Wuhan City

Objective To explore the feasibility of the application of autoregressive integrated moving average(ARIMA) model to forcast the monthly incidence rate of bacillary dysentery in Jianghan District of Wuhan ,and to provide scientific references for prevention and control .Methods SAS 9 .2 software was used to fit the ARIMA model for the monthly incidence rate of bacil‐lary dysentery from 2005 to 2013 .Thereafter ,the fitted model was used to forecast the data from January to June in 2014 ,and was evaluated by comparing the actual incidence .Results The ARIMA(0 ,1 ,1)X(0 ,1 ,1)12 model had a better fitted effect ,it was pre‐dicted that the incidence of bacillary dysentery of Jianghan District of Wuhan City from January to June 2014 would increase slight‐ly ,and the forecast data of bacillary dysentery from January 2005 to December 2013 consisted with the actual situation .Conclusion ARIM A model can be used as short‐term forecasting model to predict the monthly incidence rate of bacillary dysentery in Jianghan District of Wuhan .

Objective To study the clinical characteristics of obstructive sleep apnea-hypopnea syndrome in the aged. Methods Forty-two over 60 patients with obstruction sleep apnea-hypopnea syndrome (OSAHS) diagnosed by polysomnography (PSG) and 42 matched but less than 60 years OSAHS patients were selected, who were admitted to our hospital from February 2003 to December 2005. ObjectiveTo observe the curative effect of pulsed electromagnetic fields (PEMF) treatment system on post -menopausal osteoporosis.Methods16 patients with post-menopausal osteoporosis were treated by Union-2000 PEMF Osteoporosis Trea tment System. The bone mineral density (BMD) and grades of lumbago were measured before and after the treatment.ResultsAfter one p eriod of the treatment, BMD had increased in a field of no statistic significanc e (P>0.05) and lumbago died away in all cases.Conclusions PEMF treatment system is likely to be a practicabl e therapy for post-menopausal osteoporosis.
7.Effects of chronic renal failure rabbit serum on proliferation and nuclear factor kappa B activation of rabbit arterial smooth muscle cells

Objective To investigate the effects of chronic renal failure rabbit serum on proliferation and nuclear factor kappa B (NF-κB) activation of rabbit arterial smooth muscle cells (ASMCs) and to explore the possible mechanism. Methods Rabbit model of chronic renal failure was established by the ligation of renal arterial branches. ASMCs were incubated in the media with various concentrations of chronic renal failure serum cultured in vitro. Cell proliferation was assessed by MTT. Cell apoptosis was detected by Hoechst33342 staining. NF-κB p65 nuclear translocation was analyzed by immunofluorescence. Expression of proliferating cell nuclear antigen (PCNA) and NF-κB p65 proteins in response to chronic renal failure serum in ASMCs was determined by Western blotting. Results Lower concentrations of chronic renal failure serum (≤ 10%) could significantly promot the proliferation of ASMCs in a dose- and time-dependent manner. Higher concentrations of chronic renal failure serum (>10%) could significantly inhibit the proliferation and induce apoptosis of ASMCs compared to the normal control (P<0.05). Under the stimulation of lower concentrations of chronic renal failure serum, the expression of PCNA and NF-κB p65 increased significantly compared to the normal control (P<0.01), while decreased markedly under the stimulation of higher concentrations of chronic renal failure serum compared to the normal control (P<0.01). Under the stimulation of 10% chronic renal failure serum, nuclear translocation of NF-κB p65 in ASMCs was found. Conclusions Different concentrations of chronic renal failure rabbit serum can effectively induce ASMCs proliferation or apoptosis. The mechanism of promoting proliferation may be mediated by activating NF-κB, which will be useful for the treatment of accelerated atherosclerosis in chronic renal failure.
10.Diagnosis and scr eening for antimalarial retinopathy:two cases report and literature review

Objective To analyze antimalarial assosiated retinopathy detected by different methods with of modern ocular fundus examination, and explore screening strategy and the diagnosis of hydroxych-loroquine assosiatedretinopathy according to clinical experience and international guidelines. Methods Full fundus examination was performed in two patients with antimalarial retinopathy. The related literature were reviewed. Results Two patients had bull's eyes maculopathy and abnormal visual fields. Conclusion Clin-icians' alertness to hydroxychloroquine related retinopathy may improve early diagnosis of hydroxychloroquine toxicity. New objective tests are more sensitive than visual fields examination. Visible bull's-eye maculopathy is a late change, and the goal of screening is to recognize toxicity at early stage.
