Growth year is one of the important factors for the quality of Polygala tenufolia. In this study, primary metabolites and secondary metabolites were compared in 1, 2 and 3 years old P. tenufolia cultivated in Shaanxi Heyang. The samples were subjected to ultra-high performance liquid chromatography (UPLC)-quadrupole time-of-flight mass spectrometry (Q-TOF MS) and nuclear magnetic resonance (NMR) analysis, and the obtained data were analyzed using principal component analysis (PCA) and other statistical analysis methods. In addition, content and correlation of different metabolites were also calculated. The results showed no significance between main component contents in 2 year-old and 3 year-old P. Tenufolia, but 1 year-old was statistically different. The contents of primary metabolites, such as fructose, sucrose, and choline increased as time goes on, while glycine and raffinose decreased. The contents of secondary metabolites, such as onjisaponin Fg, polygalasaponin XXVIII, polygalasaponin XXXII increased, while polygalaxanthone III and parts of oligosaccharide multi-ester including tenuifoliose A, tenuifoliose C, tenuifoliose C2 and tenuifoliose H decreased with the extension of the growth years. Growth years has important impact on the quality of P. tenuifolia and the existing growing years of commodity P. tenuifolia have its scientific evidence. This study supplied a new method for the quality evaluation of Chinese medicinal materials.
3.Study on Quality Standard of Chushi Pill

OBJECTIVE:To establish the quality standard of Chushi pill. METHODS:Microscopic identification and TLC were adopted for the qualitative identification of Cortex moutan,C. dictamni,Angelica sinensis,Rubia cordifolia and Gardeniae fructus in Chushi pill;HPLC was performed to determine the contents of paeonol and baicalin. It was performed on column of Kro-masil 100-5 C18 with mobile phase of methanol-water-phosphoric acid(47∶53∶0.2,V/V/V)at the flow rate of 1.0 ml/min,the detec-tion wavelength was 280 nm,the temperature was 25 ℃ and the volume was 10 μl. RESULTS:The microscopic identification showed microscopic characteristics of C. moutan and C. dictamni,and characteristics of A. sinensis,R. cordifolia and G. fructus were identified by TLC;the linear range of paeonol was 0.106 24-2.124 8 μg(r=0.999 9)and baicalin was 0.059 04-1.180 8 μg (r=0.999 9);RSDs of precision,stability and reproducibility tests were no more than 2.06%;average recoveries were respective-ly 101.56%(RSD=1.68%,n=9)and 100.16%(RSD=1.13%,n=9). CONCLUSIONS:The method is simple,accurate and re-producible,and can be used for the quantity control of Chushi pill.
4.Comparison of the effects of Ginkgo biloba extract and minocycline hydrochlovide on periodontitis.

OBJECTIVETo compare the clinical effect of Ginkgo biloba extract gel (Ginkgo biloba extract,EGB) and minocycline hydrochloride (Periocline) on periodontitis and their inhibition on putative periodontal pathogens.
METHODSThirty patients with moderate-to-severe periodontitis were selected. The patients were divided into an experimental group and a positive control group (minocycline hydrochloride). Supragingival and subgingival scaling were performed on all patients. Subgingival plaque samples were collected before treatment, 1 week, 2 months and 4 months after treatment. The four major periodontal pathogens Porphyromonas gingivalis (Pg), Bacteroides forsythus (Bf), Prevotella intermedia (Pi), Treponema denticola (Td) were detected by polymerase chain reaction. Clinical indexes plaque index (PLI), bleeding index (BI) and probing depth (PD), attachment loss (AL) were examined before treatment, 3 months and 6 months after treatment. The results were statistically analyzed.
RESULTSThe detection rates of the 4 periodontal pathogens were Td (83.3%), Tf (95.0%), Pi (80.0%), Pg (81.7% ) in experimental group and Td (83.3%), Tf (95.0%), Pi (80.0%), Pg (81.7%) in control group before treatment. The detection rates in experimental group were not significantly different with those in control group after treatment, except for the detection rate of Pg 1 week after treatment (P < 0.01, the detection of Pg was 56.7% in experimental group and 53.3% in control group 1 week after treatment). The PLI and BI were not significantly different between experimental group and control group after treatment (P > 0.05). The difference was statistically significant at 6 months after treatment [(3.5 ± 0.5) mm for experimental group and (3.2 ± 0.4) mm for control group, P = 0.00]. The mean of AL decreased with time. The difference was statistically significant at 6 months after treatment [ (4.5 ± 0.4) mm for experimental group and (4.3 ± 0.4) mm for control group at 6 months, P = 0.01].
CONCLUSIONSThe inhibition effects of EGB and minocycline hydrochloride were comparable for major periodontal pathogens within short term.

5.Study on HPLC chromatographic fingerprint of anti-tumor active site SSCE of Caulis spatholobi.

OBJECTIVETo establish the chromatographic fingerprints for the anti-tumor flavonoids of Caulis spatholobi (SSCE). It could used to reflect the chemical information in this part comprehensively, and identify the chemical consitituents preliminarily.
METHODThe HPLC-DAD analysis method was performed on the column Kromasil 100-5PHENYL (4.6 mm x 250 mm, 5 microm). The mobile phase was water (0.5% acetic acid)- methanol in gradient elution and the detection wavelength was 254 nm.
RESULTThe chromatographic fingerprint of SSCE was established, which showed 16 characteristic peaks from 10 batches of medicinal materials. Among them, the peaks 1, 3, 4, 5, 8, 9, 10, 12, 13, and 16 were identified 3,4-dihodroxybenzoic acid, 4-Hydroxybenzoic Acid, epicatechin, puerarin, daidzein, liquiritigenin, calycosin, genistein, formononetin, and prunetin, respectively.
CONCLUSIONThe method is convenient, reproducibility and stability. It can used for quality control of the anti-tumor flavonoids of C. spatholobi (SSCE).

6. Effect of comfrey cream combined with NB-UVB on skin barrier function and immune function in the patients with psoriasis vulgaris

Objective:
To investigate the effects of comfrey cream combined with narrow-band UVB (NB-UVB) on skin barrier function and immune function in the patients with psoriasis vulgaris.
Methods:
A total of 100 patients with psoriasis vulgaris admitted from March 2016 to June 2017 were enrolled in the study. They were randomly divided into observation group (Purple Cream + NB-UVB) and control group (NB- UVB), 50 cases in each group. Psoriasis lesion area and severity index (PASI) and degree of pruritus were evaluated before treatment, after 2 weeks and 4 weeks of treatment. The skin barrier function and immune related indicators were compared between the two groups before and after the treatment.
Results:
After treatment, the PASI score and pruritus score of the observation group and the control group showed a decreasing trend, and the observation group decreased more significantly. The interaction between the two groups at different time points, and between groups and at different time points were statistically significant (
7.Components analysis and anti-psoriatic effect of standard decoction of Lithospermum

Objective To analyze the main ingredients and investigate the anti-psoriasis effect in the standard decoction of Lithospermum. Methods The extraction rate was determined by extract determination method, and the content of total polysaccharide and total phenolic acid was evaluated by spectrophotometry. Moreover, the effects of different concentrations of the standard decoction of Lithospermum on skin lesions induced by imiquimod (IMQ) in psoriatic mice were observed. Results The extracting rate was 4.65 %, the total polysaccharide content was 1.182 mg/ml and the total phenolic acid content was 3.506 mg/ml. The different concentrations of the standard decoction of Lithospermum could ameliorate the scales, erythema and psoriasis-like mice skin and reduce the thickness of epidermis. Conclusions The standard decoction of Lithospermum could improve the psoriasis-like lesions induced by imiquimod in mice and possess the anti-psoriasis effect.
8.Study on the quality standard of Qingre-Chushi granules

Objective To study the quality standard of Qingre-Chushi granules, including TLC identification of main components and ultra performance liquid chromatography determination of main components, so as to lay a foundation for further development of new drugs. Methods Seven constituents, including Gentianae Radix et Razoma, Scutellaria Radix, Plantaginis Herba, Rehmanniae Radix, Imperarae Rhizoma, Poria and Glycyrrhiza Radix et Rhizoma, were determined by TLC. The content of gentiopicroside and Baicalin in Gentiana officinalis and Scutellaria baicalensis was determined by UPLC on Acquity UPLC HSS T3C18 column (2.1 mm×100 mm, 1.7 μm) with acetonitrile-0.01% formic acid water as mobile phase and gradient elution. Results TLC was highly specific and could effectively identify Gentian, Scutellaria baicalensis, Plantain, Rehmannia glutinosa, Rhizoma cocos, and licorice. The results of UPLC assay showed that there was a good linear relationship between gentiopicroside and Baicalin in the range of 0.014-0.028 μg and 0.015-0.449 μg, and the recovery of the three concentrations were all up to standard. Conclusions TLC can be used to identify the seven components of Qingre-Chushi granules, and UPLC can be used to determine the contents of three indicators, which can control the quality of Qingre-Chushi granules.
