AIM:There are some reports about the treatment of spinal cord injury with neural stem cells(NSCs) transplantation,but the ideas about the transplantation time and patterns and detection indexes are still different.In this study,the effects of cryopreserved NSC transplantation on the axonal regeneration after the spinal cord injury in rats were observed.METHODS:The experiment was carried out in the experimental animal center of China Medical University from June 2005 to June 2006.①Thirty-six adult Wistar rats,either male or female and 250-300 g,were provided by the experimental animal department of China Medical University.Neural stem cells were isolated from 10 neonate rats and cultured.All treatments for animals were accorded with the animal ethical criteria.②NSCs at logarithmic phase were cryopreserved at-70 ℃ for 2 weeks and labeled with 5-bromodeoxyuridine(Brdu) after rewarming.After the models of spinal cord injury were established,the NSCs were transplanted into the injured site immediately.Thirty-six rats were randomly divided into NSC transplantation group,DMEM solution group,and control group.③NSC survival and migration were detected by immunohistochemistry,and the reconstruction of spinal cord were detected by horseradish peroxidase(HRP) staining.RESULTS:Of the 36 spinal injured model rats,4 died of overdose of anesthesia,and 5 died of infection,but all were supplemented.Brdu-labeled positive NSCs were detected in the injured spinal cord after transplantation on the 7th day,and increased on the 14th day,then gradually decreased since the 28th day till disappeared.The number of HRP positive cells in transplantation group was significantly higher than DMEM solution.CONCLUSION:Cryopreserved NSCs can survive in the injured site after transplantation and promote the reconstruction of axoplasm pathway of injured spinal cord.
5.Inhibitory Effect of Total Flavonoids of Ampelopsis Grossedentata on Human Hepatoma Carcinoma Cells in Vitro

OBJECTIVE:To investigate the inhibitory effect of the total flavoniods of Ampelopsis grossedentata on human hepatoma carcinoma cells(HCC) in vitro.METHODS:The inhibitory effect of the total flavoniods of Ampelopsis grossedentata extract on human hepatoma carcinoma BEL7404 cells in vitro was analyzed by MTT,cell growth curve method and colony formation method,respectively.RESULTS:The total flavonoids of Ampelopsis grossedentata significantly inhibited the growth of BEL 7404 cells in vitro.MTT assay showed that the IC50 was 28.59 ?g?mL-1;the cell growth curve measurement showed that the total flavonoids of Ampelopsis grossedentata significantly inhibited the growth of BEL 7404 cells;the colony formation assay showed that the IC50 was 100% at a drug concentration of above 22.22 ?g?mL-1 and the IC50 was 58.05% at a drug concentration of 14.81 ?g?mL-1.CONCLUSION:The total flavonoids of Ampelopsis grossedentata have a remarkable inhibitory effects on the growth of BEL 7404 in vitro.
6.Effect of ampelopsis of Ampelopsis grossedentata on duck hepatitis B virus.

OBJECTIVEWe have evaluated the direct effect of ampelopsis (APS) on duck hepatitis B virus (DHBV) replication in ducklings in vivo.
METHODOne-day-old ducklings were infected with DHBV. After infection for 7 days, the animals were treated with APS at dosages of 70, 150, 300 mg x kg(-1) of body weight via the oral route. The drug was given twice per day for 10 days continuously, and normal saline was used as control. The blood was drawn from the posterior tibial vein of all ducks before treatment (T0), after the medication for 5 (T5), 10 (T10) days and withdrawal of the drug for 3 days (P3). DHBV DNA in duck serum was detected by dot blot.
RESULTThe duck serum DHBV-DNA levels were reduced in the group of APS (150, 300 mg x kg(-1)) after treated for 5 and 10 days and the levels of DHBV-DNA did not markedly relapse in both groups of APS after withdrawal of the drug for 3 days. We provide the first evidence that APS can efficiently inhibits DHBV replication in ducks in vivo.
CONCLUSIONAPS therefore warrants further investigation as a potential therapeutic agent for HBV infections.

7. Correlation between CYP2C19 gene polymorphism and individualized medication in patients with ischemic stroke

AIM: To investigate the guiding role of individualized medication adjustment based on CYP2C19 metabolic typing in the treatment of ischemic stroke with clopidogrel, and to provide reference for clinical individualized medication. METHODS: The total of 80 patients with ischemic stroke were divided into the individualized drug instruction group with gene detection (n=40) and the control group without gene detection (n=40) according to whether they received CYP2C19 gene detection. According to the metabolism of CYP2C19, the individualized medication instruction group was divided into slow metabolic type, intermediate metabolic type, fast metabolic type and ultra-fast metabolic type. Patients with fast and ultra-fast metabolites were given clopidogrel dose of 75 mg once a day. Patients with intermediate metabolic type were given double clopidogrel dose of 150 mg once a day. Patients with slow metabolism were given tigrillo dose of 90 mg twice a day or aspirin dose of 100 mg once a day. The control group received 75 mg clopidogrel once a day. All patients enrolled in the groups were followed up for 3 months by outpatients or telephone. The incidence of vascular events and mRS scale scores were compared between the two groups. RESULTS: The incidence of vascular events in the individualized drug instruction group was significantly lower than that in the control group, and the incidence of mRS score(0-1) was significantly higher than that in the control group, with statistically significant differences (P<0.05). CONCLUSION: The individualized medication for patients with ischemic stroke by CYP2C19 gene detection can significantly reduce the incidence of adverse vascular events and improve the prognosis and living ability of patients.
